    Mesenchymal stem cells for the treatment of systemic lupus erythematosus: is the cure for connective tissue diseases within connective tissue?

    Mesenchymal stem cells (MSCs) are now known to display not only adult stem cell multipotency but also robust anti-inflammatory and regenerative properties. After widespread in vitro and in vivo preclinical testing in several autoimmune disease models, allogenic MSCs have been successfully applied in patients with severe treatment-refractory systemic lupus erythematosus. The impressive results of these uncontrolled phase I and II trials - mostly in patients with non-responding renal disease - point to the need to perform controlled multicentric trials. In addition, they suggest that there is much to be learned from the basic and clinical science of MSCs in order to reap the full potential of these multifaceted progenitor cells in the treatment of autoimmune diseases

    Diseño estructural de una edificación de Concreto Armado de 5 niveles para mitigar los daños estructurales de sismos severos en edificaciones en el distrito de Los Olivos

    El propósito de esta tesis es el diseño estructural de un edificio utilizando concreto armado en Av. Universitaria Mz. B lote 5, urb. Los Parques de Villa Sol, distrito de Los Olivos, Provincia de Lima, departamento de Lima, posee un área de terreno de 450m2 y un perímetro de 90m, además, tiene un ancho de 15m y un largo de 30m. cuenta con un suelo en su superficie de gravas y arcillas con desechos de construcción a -0.40 a -5.00m de profundidad presenta un suelo fino o limo consolidado. El edificio cuenta con 5 niveles, el 1er nivel cuenta con una recepción, un almacén con un estacionamiento para bicicletas, algunas áreas libres y 2 departamentos. Mientras del 2do al 5to nivel son plantas típicas destinadas a departamentos. Cabe indicar que todo lo concerniente a las partes del proyecto de concreto armado están sujetos a los estándares establecidos por las normas nacionales de construcción y recomendaciones prácticas extraídas de la experiencia de varios especialistas. Los primeros capítulos definen el proyecto, se conceptualiza la edificación a partir del diseño arquitectónico para la estructuración de la edificación. Asimismo, se realiza el predimensionamiento inicial de las partes estructurales y se cuantifica las cargas de acuerdo a la normativa vigente E0.20. Luego se realiza el análisis estático y dinámico con ayuda del Etabs, con la finalidad de verificar su comportamiento ante el sismo y lasas cargas gravitacionales. Asimismo, se verifica el cumplimiento de los parámetros de derivas, participación de masa y se verifica que el 80% de la cortante basal estática no supere la cortante dinámica, según la norma E0.30. Se finaliza con el diseño de los elementos estructurales de la edificación: losas aligeradas, losa maciza, columnas, placas, vigas y cimentación. El diseño se realiza de acuerdo a la normativa vigente de concreto armado E0.60. Siguiendo los alcances y parámetros acerca del diseño por flexión, cortante y flexo compresión.The purpose of this thesis is the structural design of a building using reinforced concrete in Av. Universitaria Mz. B lote 5, urb. Los Parques de Villa Sol, district of Los Olivos, Province of Lima, department of Lima, has a land area of 450m2 and a perimeter of 90m, also has a width of 15m and a length of 30m. has a soil surface of gravel and clay with construction waste at -0.40 to -5.00m depth presents a fine soil or consolidated silt. The building has 5 levels, the 1st level has a reception, a warehouse with a bicycle parking, some free areas and 2 apartments. The 2nd to 5th levels are typical apartment floors. It should be noted that everything concerning the parts of the reinforced concrete project are subject to the standards established by the national construction norms and practical recommendations drawn from the experience of various specialists. The first chapters define the project, the building is conceptualized from the architectural design for the structuring of the building. Likewise, the initial pre-dimensioning of the structural parts is carried out and the loads are quantified according to the current E0.20 standard. Then, the static and dynamic analysis is performed with the help of Etabs, in order to verify its behavior under seismic and gravity loads.     Functional Properties of Human-Derived Mesenchymal Stem Cell Spheroids: A Meta-Analysis and Systematic Review

    Mesenchymal stem cells (MSC) are adult multi-potent cells that can be isolated from many types of tissues including adipose tissue, bone marrow, and umbilical cord. They show great potential for cell therapy-based treatments, which is why they are being used in numerous clinical trials for a wide range of diseases. However, the success of placebo-controlled clinical trials has been limited, so new ways of improving the therapeutic effects of MSC are being developed, such as their assembly in a 3D conformation. In this meta-analysis, we review aggregate formation, in vitro functional properties and in vivo therapeutic potential displayed by adipose tissue, bone marrow, and umbilical cord-derived MSC, assembled as spheroids. The databases PubMed and SciELO were used to find eligible articles, using free-words and MeSH terms related to the subject, finding 28 published articles meeting all inclusion and exclusion criteria. Of the articles selected 15 corresponded to studies using MSC derived from bone marrow, 10 from adipose tissue and 3 from umbilical cord blood or tissue. The MSC spheroids properties analyzed that displayed enhancement in comparison with monolayer 2D culture, are stemness, angiogenesis, differentiation potential, cytokine secretion, paracrine and immunomodulatory effects. Overall studies reveal that the application of MSC spheroids in vivo enhanced therapeutic effects. For instance, research exhibited reduced inflammation, faster wound healing, and closure, functional recovery and tissue repair due to immunomodulatory effects, better MSC engraftment in damaged tissue, higher MSC survival and less apoptosis at the injury. Still, further research and clinical studies with controlled and consistent results are needed to see the real therapeutic efficacy of MSC spheroids

    Advances in extracellular vesicles as mediators of cell-to-cell communication in pregnancy

    Cell-to-cell communication mediated by Extracellular Vesicles (EVs) is a novel and emerging area of research, especially during pregnancy, in which placenta derived EVs can facilitate the feto-maternal communication. EVs comprise a heterogeneous group of vesicle sub-populations with diverse physical and biochemical characteristics and originate by specific biogenesis mechanisms. EVs transfer molecular cargo (including proteins, nucleic acids, and lipids) between cells and are critical mediators of cell communication. There is growing interest among researchers to explore into the molecular cargo of EVs and their functions in a physiological and pathological context. For example, inflammatory mediators such as cytokines are shown to be released in EVs and EVs derived from immune cells play key roles in mediating the immune response as well as immunoregulatory pathways. Pregnancy complications such as gestational diabetes mellitus, preeclampsia, intrauterine growth restriction and preterm birth are associated with altered levels of circulating EVs, with differential EV cargo and bioactivity in target cells. This implicates the intriguing roles of EVs in reprogramming the maternal physiology during pregnancy. Moreover, the capacity of EVs to carry bioactive molecules makes them a promising tool for biomarker development and targeted therapies in pregnancy complications. This review summarizes the physiological and pathological roles played by EVs in pregnancy and pregnancy-related disorders and describes the potential of EVs to be translated into clinical applications in the diagnosis and treatment of pregnancy complications.</p

Background: Intracoronary delivery of autologous bone marrow mononuclear cells is an interesting therapeutic promise for patients with heart failure of different etiologies. Aim: To evaluate the long-term safety and efficacy of this therapy in patients with dilated cardiomyopathy of different etiologies under optimal medical treatment. Patients and Methods: Prospective, open-label, controlled clinical trial. Of 23 consecutive patients, 12 were assigned to autologous bone marrow mononuclear cell intracoronary transplantation, receiving a mean dose of 8.19 ± 4.43 x 106 CD34+ cells. Mortality, cardiovascular readmissions and cancer incidence rate, changes in functional capacity, quality of life questionnaires and echocardiographic measures from baseline, were assessed at long-term follow-up (37.7 ± 9.7 months) in patients receiving or not the cells.
