A Great Space Weather Event in February 1730
Aims. Historical records provide evidence of extreme magnetic storms with
equatorward auroral extensions before the epoch of systematic magnetic
observations. One significant magnetic storm occurred on February 15, 1730. We
scale this magnetic storm with auroral extension and contextualise it based on
contemporary solar activity. Methods. We examined historical records in East
Asia and computed the magnetic latitude (MLAT) of observational sites to scale
magnetic storms. We also compared them with auroral records in Southern Europe.
We examined contemporary sunspot observations to reconstruct detailed solar
activity between 1729 and 1731. Results. We show 29 auroral records in East
Asian historical documents and 37 sunspot observations. Conclusions. These
records show that the auroral displays were visible at least down to 25.8{\deg}
MLAT throughout East Asia. In comparison with contemporary European records, we
show that the boundary of the auroral display closest to the equator surpassed
45.1{\deg} MLAT and possibly came down to 31.5{\deg} MLAT in its maximum phase,
with considerable brightness. Contemporary sunspot records show an active phase
in the first half of 1730 during the declining phase of the solar cycle. This
magnetic storm was at least as intense as the magnetic storm in 1989, but less

A note on the sunspot and prominence records made by Angelo Secchi during the period 1871–1875
Angelo Secchi (1818–1878) was an Italian Jesuit who made relevant scientific contributions in geophysics, meteorology, and astrophysics. He was a well-known pioneer in solar physics due to his theories and observations. Secchi published Le Soleil (The Sun) a summary of knowledge about our star in that time. Moreover, he published in this book his sunspot and prominence observations made during the period 1871–1875. In this work, we present a machine-readable version of these observations and a preliminary analysis of them
The Extreme Space Weather Event in 1903 October/November: An Outburst from the Quiet Sun
While the Sun is generally more eruptive during its maximum and declining
phases, observational evidence shows certain cases of powerful solar eruptions
during the quiet phase of the solar activity. Occurring in the weak Solar Cycle
14 just after its minimum, the extreme space weather event in 1903 October --
November was one of these cases. Here, we reconstruct the time series of
geomagnetic activity based on contemporary observational records. With the
mid-latitude magnetograms, the 1903 magnetic storm is thought to be caused by a
fast coronal mass ejection (~1500 km/s) and is regarded as an intense event
with an estimated minimum Dst' of ~-513 nT The reconstructed time series has
been compared with the equatorward extension of auroral oval (~44.1{\deg} in
invariant latitude) and the time series of telegraphic disturbances. This case
study shows that potential threats posed by extreme space weather events exist

Holistic assessment of the microbiome dynamics in the substrates used for commercial champignon (Agaricus bisporus) cultivation
Microorganisms strongly influence and are required to generate the selective substrate that provides nutrients and support for fungal growth, and ultimately to induce mushroom fructification under controlled environmental conditions. In this work, the fungal and bacterial microbiota living in the different substrates employed in a commercial crop (compost phase I, II and III, flush 1 and 2, and casing material on day 1, 6 and 8 after compost casing and during flush 1 and 2) have been characterized along the different stages of cultivation by metataxonomic analysis (16S rRNA and ITS2), analysis of phospholipid fatty acid content (PLFAs) and RT-qPCR. Additionally, laccase activity and the content of lignin and complex carbohydrates in compost and casing have been quantified. The bacterial diversity in compost and casing increased throughout the crop cycle boosted by the connection of both substrates. As reflected by the PLFAs, the total living bacterial biomass appears to be negatively correlated with the mycelium of the crop. Agaricus bisporus was the dominant fungal species in colonized substrates, displacing the pre-eminent Ascomycota, accompanied by a sustained increase in laccase activity, which is considered to be a major product of protein synthesis during the mycelial growth of champignon. From phase II onwards, the metabolic machinery of the fungal crop degrades lignin and carbohydrates in compost, while these components are hardly degraded in casing, which reflects the minor role of the casing for nourishing the crop. The techniques employed in this study provide a holistic and detailed characterization of the changing microbial composition in commercial champignon substrates. Background
The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd.
Methods
We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background.
Results
First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001).
Conclusions
In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival
SunMap: A Solar Image Processing Software for Obtaining Synoptic Maps
A new open-source software, called SunMap, has been developed to obtain synoptic maps in an easy and quick way from multiple full-disc solar images. Our objective is to provide a free and straightforward application for heliophysicists and geophysicists interested in generating solar synoptic maps. SunMap allows comparison of structures and patterns of solar activity over various periods. Thus, the short- and long-term evolution of solar regions of interest can be studied. To reach this goal, different solar images taken day by day are stored in a single map that uses a sequence of images and allows the positioning of each observable element on it. A simple comparison between a synoptic map generated by SunMap and another previously constructed map is presented to show the versatility of this new available software
