    Utilisation of off-river habitats by lowland river fishes

    This study assessed the importance for fish of lateral connectivity of river side-channels to the River Frome, a lowland chalk. stream in Southern England. Lateral connectivity and habitat complexity is under threat in many floodplain river systems due to human disturbance. A holistic approach was used to investigate fish communities in seven sidechannels (including drainage ditches, natural streams and a millstream) and to assess the functionality provided by these habitats to fish species. Seasonal electric fishing over three years was used to monitor fish assemblages in each channel in relation to biotic and abiotic variables. Fish movements between the main channel and lateral habitats were monitored continuously in five locations with PIT (passive integrated transponder) telemetry. In total over one hundred pike (Esox lucius) and dace (Leuciscus leuciscus) were radio tracked to monitor the movements of individual fish with increased sensitivity. Each side-channel provided a distinct habitat and supported a different fish community. Flow was the main discriminating factor between channels and their assemblages. Multiple linear regression of/biotic and abiotic variables did not predict abundances of individual species effectively. In contrast, habitat stability was a good indicator of species diversity and may prove a useful management tool. Side-channels were used for different functions by different species. A single fish species used a range of habitats within the river system, each for different functions with functional differences in use between seasons. The River Frome dace population exhibited a metapopulation structure with a main river source population and sink sub-populations in side-channels. The structure of the population is based upon the availability of both lateral' and longitudinal connectivity, with dace moving between the main river and side-channels and also making excursions over 10 km along the main river channel. Drainage ditches are a particularly important pike spawning location, with males arriving earlier onto this distinct habitat. Protandry (early arrival strategy of some males) was exhibited by slow-growing male pike which arrived as early as December.     Isolation and partial characterization of lipoprotein A-II (LP-A-II) particles of human plasma

    High density lipoproteins (HDL) consist of a mixture of chemically and functionally distinct families of particles defined by their characteristic apolipoprotein (Apo) composition. The two major lipoprotein families are lipoprotein A-I (LP-A-I) and lipoprotein A-I : A-II (LP-A-I : A-II). This study describes the isolation of a third minor HDL family of particles referred to as lipoprotein A-II (LP-A-II) because it lacks ApoA-I and contains ApoA-II as its main or sole apolipoprotein constituent. Because ApoA-II is an integral protein constituent of three distinct lipoprotein families (LP-A-I : A-II, LP-A-II : B : C : D : E and LP-A-II), LP-A-II particles were isolated from whole plasma by sequential immunoaffinity chromatography on immunosorbers with antisera to ApoA-II, ApoB and ApoA-I, respectively. In normolipidemic subjects, the concentration of LP-A-II particles, based on ApoA-II content, is 4-18 mg/dl accounting for 5-20% of the total ApoA-II not associated with ApoB-containing lipoproteins. The lipid composition of LP-A-II particles is characterized by low percentages of triglycerides and cholesterol esters and a high percentage of phospholipids in comparison with lipid composition of LP-A-I and LP-A-I : A-II. The major part of LP-A-II particles contain ApoA-II as the sole apolipoprotein constituents; however, small subsets of LP-A-II particles may also contain ApoD and other minor apolipoproteins. The lipid/protein ratio of LP-A-II is higher than those of LP-A-I and LP-A-I : A-II. In homozygous ApoA-I and ApoA-I/ApoC-III deficiencies, LP-A-II particles are the only ApoA-containing high density lipoprotein with levels found to be within the same range (7-13 mg/dl) as those of normolipidemic subjects. However, in contrast to normal LP-A-II, their lipid composition is characterized by higher percentage of triglycerides and cholesterol esters and a lower percentage of phospholipids and their apolipoprotein composition by the presence of ApoC-peptides and ApoE in addition to ApoA-II and ApoD. These results show that LP-A-II particles are a minor HDL family and suggest that, in the absence of ApoA-I-containing lipoproteins, they become an efficient acceptor/donor of ApoC-peptides and ApoE required for a normal metabolism of triglyceride-rich lipoproteins.     Understanding how education/support groups help lone mothers

    <p>Abstract</p> <p>Background</p> <p>Lone-mother led families are at increased risk of psychosocial disadvantage, social isolation and mental health morbidity. Community-based programs are more accessible for families seeking assistance. We examine the experiences of eight lone mothers participating in a larger randomized controlled trial (RCT) of a community-based education/support group program using mixed methods.</p> <p>Methods</p> <p>A purposeful sample of eight mothers participating in the intervention arm of an RCT of community-based support/education groups was selected for the qualitative study. Individual interviews asked mothers about themselves and their relationships with their children before and after the group. Interviews were taped, transcribed and content analysis was used to code and interpret the data. Quantitative data collected in the RCT were used to describe these mothers.</p> <p>Results</p> <p>Mothers participating in the RCT and qualitative study experienced multiple difficulties, including financial and mood problems. These mothers reported that before participating in the group, they had shared experiences of social isolation, stigma, a sense of failure, poor relationships with their children and difficulties with financial management. After the group, mothers identified improved self-esteem, support from other mothers, improved parenting skills and improved communication with their children as outcomes of group participation.</p> <p>Conclusions</p> <p>The qualitative data revealed mothers' perceptions of specific areas that improved by participating in the group. The utility of complementary information provided by qualitative and quantitative methods in understanding program impact, as well as the need for broader assistance is noted.</p

    Do aluminium-based phosphate binders continue to have a role in contemporary nephrology practice?

    Background: Aluminium-containing phosphate binders have long been used for treatment of hyperphosphatemia in dialysis patients. Their safety became controversial in the early 1980's after reports of aluminium related neurological and bone disease began to appear. Available historical evidence however, suggests that neurological toxicity may have primarily been caused by excessive exposure to aluminium in dialysis fluid, rather than aluminium-containing oral phosphate binders. Limited evidence suggests that aluminium bone disease may also be on the decline in the era of aluminium removal from dialysis fluid, even with continued use of aluminium binders

    Investigating the DART Impact Event with the Lucy LOng Range Reconnaissance Imager

    NASA’s Lucy mission is the first to provide flyby reconnaissance of the Jovian trojan asteroids, which are thought to be primordial small bodies that formed at a variety of heliocentric distances during the early stages of the solar system’s formation and were subsequently captured into Jupiter’s L4 and L5 Lagrange stability zones. Since its successful launch on 2021-Oct-16, the Lucy spacecraft has been orbiting the sun within the inner solar system. On 2022-Oct-16, Lucy executes the first of three Earth Gravity Assists (EGAs) that put the spacecraft on the correct trajectory to achieve its encounters with the Jovian trojans. The DART kinetic impact on the secondary body of the Didymos-Dimorphos binarysystem occurs 20 days prior to EGA1, at a time when the Lucy spacecraft is well-placed to observe it. Lucy carries a sensitive panchromatic camera, the Lucy LOng Range Reconnaissance Imager (L’LORRI), which is capable of detecting the binary system with high signal-to-noise ratio (SNR) and with temporal cadences as fast as once per second. The observing geometry from Lucy is similar to that from the Earth: the range to the Didymos system is 0.126 au from Lucy vs 0.0757 au from Earth, and the solar phase angle is 31.9 deg vs 53.2 deg. The L’LORRI investigation of the DART impact event is divided into eight separate observational phases, starting 12 hr before the impact and ending 24 hr afterwards. L’LORRI cannot resolve the binary, but instead records the total brightness, which is expected to increase after the DART impact due to reflected sunlight from the ejecta. The first two phases are designed to obtain baseline photometry of the Didymos system covering both the Didymos-Dimorphos mutual orbit period (11.92 hr) and the rotational period of Didymos (2.26 hr). Phase 3 covers the impact event itself at one second cadence, starting 3 minutes beforeimpact and ending 4 minutes afterwards. Lucy has a clear view of the predicted DART impact site, theoretically enablingL’LORRI to detect an optical flash in the unlikely event it is brighter than Didymos itself. L’LORRI observations during phases 4 through 8 are designed to monitor the temporal and spatial evolution of ejecta associated with the impact event, but ejecta don’t leave the central pixel during Lucy’s observing period unless their speed is greater than about 2 m/s

    BRF1 accelerates prostate tumourigenesis and perturbs immune infiltration

    BRF1 is a rate-limiting factor for RNA Polymerase III-mediated transcription and is elevated in numerous cancers. Here, we report that elevated levels of BRF1 associate with poor prognosis in human prostate cancer. In vitro studies in human prostate cancer cell lines demonstrated that transient overexpression of BRF1 increased cell proliferation whereas the transient downregulation of BRF1 reduced proliferation and mediated cell cycle arrest. Consistent with our clinical observations, BRF1 overexpression in a Pten-deficient mouse (Pten BRF1 ) prostate cancer model accelerated prostate carcinogenesis and shortened survival. In Pten BRF1 tumours, immune and inflammatory processes were altered, with reduced tumoral infiltration of neutrophils and CD4 positive T cells, which can be explained by decreased levels of complement factor D (CFD) and C7 components of the complement cascade, an innate immune pathway that influences the adaptive immune response. We tested if the secretome was involved in BRF1-driven tumorigenesis. Unbiased proteomic analysis on BRF1-overexpresing PC3 cells confirmed reduced levels of CFD in the secretome, implicating the complement system in prostate carcinogenesis. We further identify that expression of C7 significantly correlates with expression of CD4 and has the potential to alter clinical outcome in human prostate cancer, where low levels of C7 associate with poorer prognosis

    VHA-19 Is Essential in Caenorhabditis elegans Oocytes for Embryogenesis and Is Involved in Trafficking in Oocytes

    There is an urgent need to develop new drugs against parasitic nematodes, which are a significant burden on human health and agriculture. Information about the function of essential nematode-specific genes provides insight to key nematode-specific processes that could be targeted with drugs. We have characterized the function of a novel, nematode-specific Caenorhabditis elegans protein, VHA-19, and show that VHA-19 is essential in the germline and, specifically, the oocytes, for the completion of embryogenesis. VHA-19 is also involved in trafficking the oocyte receptor RME-2 to the oocyte plasma membrane and is essential for osmoregulation in the embryo, probably because VHA-19 is required for proper eggshell formation via exocytosis of cortical granules or other essential components of the eggshell. VHA-19 may also have a role in cytokinesis, either directly or as an indirect effect of its role in osmoregulation. Critically, VHA-19 is expressed in the excretory cell in both larvae and adults, suggesting that it may have a role in osmoregulation in C. elegans more generally, probably in trafficking or secretion pathways.
