    Attachment relationships and internalization and externalization problems in a group of adolescents with pathological gambling disorder

    Objective: The evidence accumulated in the relevant literature suggests that the presence and evolution of gambling could be correlated with the internalizing and externalizing problems and with the attachment style. This paper aims at exploring this perspective further. In particular, it analyses how such risk factors interact within the specific context of adolescent gambling disorder. Method: The sample comprises 91 adolescents, 61 male and 30 female, in the 17-22 age range (M = 17.77; SD = 0.98). A structural equation model was used to examine the relationship between the Youth Self-Report latent factors and pathological gambling, and the mode of attachment was assumed to act as a moderator. Results: Our results suggest that in the group characterized by a fearful attachment style there was a positive relationship between somatization and propensity to risk (p = 0.008), whereas in the dismissing attachment group there was a positive relationship between a greater tendency to delinquent behaviour and gambling risk (p = 0.042). Conclusions: The various insecure attachment stylespatterns may contribute in different ways to the development of oppositional-provocative behaviour and problems of conduct in adolescents

    Epigenetic targeting of bromodomain protein BRD4 counteracts cancer cachexia and prolongs survival

    Cancer cachexia is a devastating metabolic syndrome characterized by systemic inflammation and massive muscle and adipose tissue wasting. Although it is responsible for approximately one-third of cancer deaths, no effective therapies are available and the underlying mechanisms have not been fully elucidated. We previously identified the bromodomain and extra-terminal domain (BET) protein BRD4 as an epigenetic regulator of muscle mass. Here we show that the pan-BET inhibitor (+)-JQ1 protects tumor-bearing mice from body weight loss and muscle and adipose tissue wasting. Remarkably, in C26-tumor-bearing mice (+)-JQ1 administration dramatically prolongs survival, without directly affecting tumor growth. By ChIP-seq and ChIP analyses, we unveil that BET proteins directly promote the muscle atrophy program during cachexia. In addition, BET proteins are required to coordinate an IL6-dependent AMPK nuclear signaling pathway converging on FoxO3 transcription factor. Overall, these findings indicate that BET proteins may represent a promising therapeutic target in the management of cancer cachexia

    Lamin A/C sustains PcG protein architecture, maintaining transcriptional repression at target genes

    Beyond its role in providing structure to the nuclear envelope, lamin A/C is involved in transcriptional regulation. However, its cross talk with epigenetic factors--and how this cross talk influences physiological processes--is still unexplored. Key epigenetic regulators of development and differentiation are the Polycomb group (PcG) of proteins, organized in the nucleus as microscopically visible foci. Here, we show that lamin A/C is evolutionarily required for correct PcG protein nuclear compartmentalization. Confocal microscopy supported by new algorithms for image analysis reveals that lamin A/C knock-down leads to PcG protein foci disassembly and PcG protein dispersion. This causes detachment from chromatin and defects in PcG protein-mediated higher-order structures, thereby leading to impaired PcG protein repressive functions. Using myogenic differentiation as a model, we found that reduced levels of lamin A/C at the onset of differentiation led to an anticipation of the myogenic program because of an alteration of PcG protein-mediated transcriptional repression. Collectively, our results indicate that lamin A/C can modulate transcription through the regulation of PcG protein epigenetic factors

    Alexithymia in juvenile primary headache sufferers: a pilot study

    Starting in the 1990s, there has been accumulating evidence of alexithymic characteristics in adult patients with primary headache. Little research has been conducted, however, on the relationship between alexithymia and primary headache in developmental age. In their research on alexithymia in the formative years, the authors identified one of the most promising prospects for research, as discussed here. The aim of this study was to verify whether there is: (a) a link between tension-type headache and alexithymia in childhood and early adolescence; and (b) a correlation between alexithymia in children/preadolescents and their mothers. This study was based on an experimental group of 32 patients (26 females and 6 males, aged from 8 to 15 years, mean 11.2 ± 2.0) suffering from tension-type headache and 32 control subjects (26 females and 6 males, aged from 8 to 15 years, mean 11.8 ± 1.6). Tension-type headache was diagnosed by applying the International Headache Classification (ICHD-II, 2004). The alexithymic construct was measured using an Italian version of the Alexithymia Questionnaire for Children in the case of the juvenile patients and the Toronto Alexithymia Scale (TAS-20) for their mothers. Higher rates of alexithymia were observed in the children/preadolescents in the experimental group (EG) than in the control group; in the EG there was no significant correlation between the alexithymia rates in the children/preadolescents and in their mothers

    Toward a person-follower robot

    In this article we described the attempt to build a robot able to locate and follow an human target moving in a domestic environment. After a brief review of the state of the art in relative location technologies, we described our approach that aims to develop robots provided with simple and robust relative location technologies that do not require to structure the environment and on simple semi-reactive strategies that does not require the use of internal maps and the ability to self-localize. More specifically, the approach is based on a control system able to display and integrate an exploration, obstacle avoidance, and target following behavior and a relative location device based on an signal emitter (placed on the target person) and a directional sensor (placed on the mobile robot)
