    Comparison of Fuzzy Clustering Methods and Their Applications to Geophysics Data

    Fuzzy clustering algorithms are helpful when there exists a dataset with subgroupings of points having indistinct boundaries and overlap between the clusters. Traditional methods have been extensively studied and used on real-world data, but require users to have some knowledge of the outcome a priori in order to determine howmany clusters to look for. Additionally, iterative algorithms choose the optimal number of clusters based on one of several performance measures. In this study, the authors compare the performance of three algorithms (fuzzy c-means, Gustafson-Kessel, and an iterative version of Gustafson-Kessel) when clustering a traditional data set as well as real-world geophysics data that were collected from an archaeological site in Wyoming. Areas of interest in the were identified using a crisp cutoff value as well as a fuzzy α-cut to determine which provided better elimination of noise and non-relevant points. Results indicate that the α-cut method eliminates more noise than the crisp cutoff values and that the iterative version of the fuzzy clustering algorithm is able to select an optimum number of subclusters within a point set (in both the traditional and real-world data), leading to proper indication of regions of interest for further expert analysis

    The First Two Years of Electromagnetic Follow-Up with Advanced LIGO and Virgo

    We anticipate the first direct detections of gravitational waves (GWs) with Advanced LIGO and Virgo later this decade. Though this groundbreaking technical achievement will be its own reward, a still greater prize could be observations of compact binary mergers in both gravitational and electromagnetic channels simultaneously. During Advanced LIGO and Virgo's first two years of operation, 2015 through 2016, we expect the global GW detector array to improve in sensitivity and livetime and expand from two to three detectors. We model the detection rate and the sky localization accuracy for binary neutron star (BNS) mergers across this transition. We have analyzed a large, astrophysically motivated source population using real-time detection and sky localization codes and higher-latency parameter estimation codes that have been expressly built for operation in the Advanced LIGO/Virgo era. We show that for most BNS events the rapid sky localization, available about a minute after a detection, is as accurate as the full parameter estimation. We demonstrate that Advanced Virgo will play an important role in sky localization, even though it is anticipated to come online with only one-third as much sensitivity as the Advanced LIGO detectors. We find that the median 90% confidence region shrinks from ~500 square degrees in 2015 to ~200 square degrees in 2016.     Parameter estimation on gravitational waves from neutron-star binaries with spinning components

    Inspiraling binary neutron stars are expected to be one of the most significant sources of gravitational-wave signals for the new generation of advanced ground-based detectors. We investigate how well we could hope to measure properties of these binaries using the Advanced LIGO detectors, which began operation in September 2015. We study an astrophysically motivated population of sources (binary components with masses 1.2 M⊙1.2~\mathrm{M}_\odot--1.6 M⊙1.6~\mathrm{M}_\odot and spins of less than 0.050.05) using the full LIGO analysis pipeline. While this simulated population covers the observed range of potential binary neutron-star sources, we do not exclude the possibility of sources with parameters outside these ranges; given the existing uncertainty in distributions of mass and spin, it is critical that analyses account for the full range of possible mass and spin configurations. We find that conservative prior assumptions on neutron-star mass and spin lead to average fractional uncertainties in component masses of ∼16%\sim 16\%, with little constraint on spins (the median 90%90\% upper limit on the spin of the more massive component is ∼0.7\sim 0.7). Stronger prior constraints on neutron-star spins can further constrain mass estimates, but only marginally. However, we find that the sky position and luminosity distance for these sources are not influenced by the inclusion of spin; therefore, if LIGO detects a low-spin population of BNS sources, less computationally expensive results calculated neglecting spin will be sufficient for guiding electromagnetic follow-up.Comment: 10 pages, 9 figure

    With the advent and stunning success of combination antiretroviral therapy (ART) to prolong and improve quality of life for persons with HIV infection, HIV research has been afforded the opportunity to pivot towards studies aimed at finding "a cure." The mere idea that cure of HIV might be possible has energized researchers and the community towards achieving this goal. Funding agencies, both governmental and private, have targeted HIV cure as a high priority; many in the field have responded to these initiatives and the cure research agenda is robust. In this "salon" two editors of Pathogens and Immunity, Michael Lederman and Daniel Douek ask whether curing HIV is a realistic, scalable objective. We start with an overview perspective and have asked a number of prominent HIV researchers to add to the discussion

    A study in high-risk, maximally pretreated patients to determine the potential use of PCSK9 inhibitors at various thresholds of total and LDL cholesterol levels

    PURPOSE OF THE STUDY: Statins and ezetimibe reduce low-density lipoprotein cholesterol (LDL-c) and cardiovascular disease (CVD) risk. Proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitors lower LDL-c by 50%-70% and might be useful in refractory patients. The National Institute for Health and Care Excellence (NICE) technology appraisal guidance (TAG) recommends use of these drugs in secondary prevention and familial hypercholesterolaemia (FH) at differing LDL-c thresholds. We have estimated the proportion of patients in whom this third-line drug might be useful. STUDY DESIGN: We used data from a lipid-lowering audit programme to study 72 with FH and/or CVD of 271 patients referred over 12 months who failed to achieve target total cholesterol (TC) and LDL-c levels. All 72 patients were treated with ezetimibe, and 69 cases also received statins. We used LDL-c thresholds 1.5-5.5 mmol/L to estimate how many of these refractory patients could benefit from PCSK9 inhibitors. RESULTS: In 72 patients, TC and LDL-c targets were not met by 64 and 53 patients, respectively. We judged using the NICE TAG that only one patient (1.4% ezetimibe requiring and 0.4% total referrals) required a PCSK9 inhibitor. CONCLUSIONS: We determined that the proportion of patients eligible for a PCSK9 inhibitor at various TC and LDL-c levels is modest. This may reflect the use of all available statins in UK lipid clinics often at non-daily frequency. We suggest that cost-effective use of PCSK9 inhibitors requires prescribing being restricted to clinicians working in specialised lipid clinics

    Parameter Estimation for Binary Neutron-star Coalescences with Realistic Noise during the Advanced LIGO Era

    Advanced ground-based gravitational-wave (GW) detectors begin operation imminently. Their intended goal is not only to make the first direct detection of GWs, but also to make inferences about the source systems. Binary neutron-star mergers are among the most promising sources. We investigate the performance of the parameter-estimation (PE) pipeline that will be used during the first observing run of the Advanced Laser Interferometer Gravitational-wave Observatory (aLIGO) in 2015: we concentrate on the ability to reconstruct the source location on the sky, but also consider the ability to measure masses and the distance. Accurate, rapid sky localization is necessary to alert electromagnetic (EM) observatories so that they can perform follow-up searches for counterpart transient events. We consider PE accuracy in the presence of non-stationary, non-Gaussian noise. We find that the character of the noise makes negligible difference to the PE performance at a given signal-to-noise ratio. The source luminosity distance can only be poorly constrained, since the median 90% (50%) credible interval scaled with respect to the true distance is 0.85 (0.38). However, the chirp mass is well measured. Our chirp-mass estimates are subject to systematic error because we used gravitational-waveform templates without component spin to carry out inference on signals with moderate spins, but the total error is typically less than 10^(-3) M_☉. The median 90% (50%) credible region for sky localization is ~ 600 deg^2 (~150 deg^2), with 3% (30%) of detected events localized within 100 deg^2. Early aLIGO, with only two detectors, will have a sky-localization accuracy for binary neutron stars of hundreds of square degrees; this makes EM follow-up challenging, but not impossible

    A Rare Variant in ERF (rs144812092) Predisposes to Prostate and Bladder Cancers in an Extended Pedigree

    Simple SummaryHere we applied a powerful predisposition candidate gene identification strategy to identify rare variants shared by two related bladder cancer cases who were members of pedigrees exhibiting a significant excess of bladder cancers. We sequenced the exomes of pairs of related bladder cancer cases belonging to high-risk bladder cancer pedigrees to identify rare, shared variants shared as candidates for predisposition. A rare, shared variant in ERF was also found to show significant association with bladder cancer risk in an independent population, was present in other prostate cancer-affected members in the pedigree, and showed evidence for altering the function of the associated protein. This evidence supports ERF (ETS2 Repressor Factor) as a bladder and prostate cancer predisposition gene.Pairs of related bladder cancer cases who belong to pedigrees with an excess of bladder cancer were sequenced to identify rare, shared variants as candidate predisposition variants. Candidate variants were tested for association with bladder cancer risk. A validated variant was assayed for segregation to other related cancer cases, and the predicted protein structure of this variant was analyzed. This study of affected bladder cancer relative pairs from high-risk pedigrees identified 152 bladder cancer predisposition candidate variants. One variant in ERF (ETS Repressing Factor) was significantly associated with bladder cancer risk in an independent population, was observed to segregate with bladder and prostate cancer in relatives, and showed evidence for altering the function of the associated protein. This finding of a rare variant in ERF that is strongly associated with bladder and prostate cancer risk in an extended pedigree both validates ERF as a cancer predisposition gene and shows the continuing value of analyzing affected members of high-risk pedigrees to identify and validate rare cancer predisposition variants
