    Intra- and interspecies interactions between prion proteins and effects of mutations and polymorphisms

    Recently, crystallization of the prion protein in a dimeric form was reported. Here we show that native soluble homogenous FLAG-tagged prion proteins from hamster, man and cattle expressed in the baculovirus system are predominantly dimeric. The PrP/PrP interaction was confirmed in Semliki Forest virus-RNA transfected BHK cells co-expressing FLAG- and oligohistidine-tagged human PrP. The yeast two-hybrid system identified the octarepeat region and the C-terminal structured domain (aa90-aa230) of PrP as PrP/PrP interaction domains. Additional octarepeats identified in patients suffering from fCJD reduced (wtPrP versus PrP+90R) and completely abolished (PrP+90R versus PrP+90R) the PrP/PrP interaction in the yeast two-hybrid system. In contrast, the Met/Val polymorphism (aa129), the GSS mutation Pro102Leu and the FFI mutation Asp178Asn did not affect PrP/PrP interactions. Proof of interactions between human or sheep and bovine PrP, and sheep and human PrP, as well as lack of interactions between human or bovine PrP and hamster PrP suggest that interspecies PrP interaction studies in the yeast two-hybrid system may serve as a rapid pre-assay to investigate species barriers in prion diseases

    Satellite to Ground-based LIDAR Comparisons using MPLNET Data Products

    The Micro-Pulse Lidar Network (MPLNET) is a network of ground-based lidar systems that provide continuous long-term observations of aerosol and cloud properties at approximately 10 different locations around the globe. Each site in the network uses an elastic scattering lidar co-located with a sunphotometer to provide data products of aerosol optical physical properties. Data products from sites are available on a next-day basis from the MPLNET website. Expansion of the network is based on partnering with research groups interested in joining MPLNET. Results have contributed to a variety of studies including aerosol transport studies and satellite calibration and validation efforts. One of the key motivations for MPLNET is to contribute towards the calibration and validation of satellite-based lidars such as GLAS/ICESAT and CALIPSO. MPLNET is able to provide comparison to several of the key aerosol and cloud CALIPSO data products including: layer height and thickness, optical depth, backscatter and extinction profiles, and the extinction-to-backscatter ratio

    Elevated frequency of abnormalities in barn swallows from Chernobyl

    Ever since the Chernobyl accident in 1986, that contaminated vast areas in surrounding countries with radiation, abnormalities and birth defects have been reported in human populations. Recently, several studies suggested that the elevated frequency of such abnormalities can be attributed to poverty and stress in affected human populations. Here, we present long-term results for a free-living population of barn swallows, Hirundo rustica, demonstrating the presence of 11 morphological abnormalities in populations around Chernobyl, but much less frequently in an uncontaminated Ukrainian control population and three more distant control populations. The presence of these abnormalities in barn swallows is associated with reduced viability. These findings demonstrate a link between morphological abnormalities and radiation in an animal population that cannot be attributed to poverty and stress. The most parsimonious hypothesis for abnormalities in animal and human populations alike is that the effects are caused by the same underlying cause, viz. radiation derived from the Chernobyl accident

    Electronic excitations and the tunneling spectra of metallic nanograins

    Tunneling-induced electronic excitations in a metallic nanograin are classified in terms of {\em generations}: subspaces of excitations containing a specific number of electron-hole pairs. This yields a hierarchy of populated excited states of the nanograin that strongly depends on (a) the available electronic energy levels; and (b) the ratio between the electronic relaxation rate within the nano-grain and the bottleneck rate for tunneling transitions. To study the response of the electronic energy level structure of the nanograin to the excitations, and its signature in the tunneling spectrum, we propose a microscopic mean-field theory. Two main features emerge when considering an Al nanograin coated with Al oxide: (i) The electronic energy response fluctuates strongly in the presence of disorder, from level to level and excitation to excitation. Such fluctuations produce a dramatic sample dependence of the tunneling spectra. On the other hand, for excitations that are energetically accessible at low applied bias voltages, the magnitude of the response, reflected in the renormalization of the single-electron energy levels, is smaller than the average spacing between energy levels. (ii) If the tunneling and electronic relaxation time scales are such as to admit a significant non-equilibrium population of the excited nanoparticle states, it should be possible to realize much higher spectral densities of resonances than have been observed to date in such devices. These resonances arise from tunneling into ground-state and excited electronic energy levels, as well as from charge fluctuations present during tunneling.Comment: Submitted to the Physical Review

    Specific Heat Study of the Magnetic Superconductor HoNi2B2C

    The complex magnetic transitions and superconductivity of HoNi2B2C were studied via the dependence of the heat capacity on temperature and in-plane field angle. We provide an extended, comprehensive magnetic phase diagram for B // [100] and B // [110] based on the thermodynamic measurements. Three magnetic transitions and the superconducting transition were clearly observed. The 5.2 K transition (T_{N}) shows a hysteresis with temperature, indicating the first order nature of the transition at B=0 T. The 6 K transition (T_{M}), namely the onset of the long-range ordering, displays a dramatic in-plane anisotropy: T_{M} increases with increasing magnetic field for B // [100] while it decreases with increasing field for B // [110]. The anomalous anisotropy in T_{M} indicates that the transition is related to the a-axis spiral structure. The 5.5 K transition (T^{*}) shows similar behavior to the 5.2 K transition, i.e., a small in-plane anisotropy and scaling with Ising model. This last transition is ascribed to the change from a^{*} dominant phase to c^{*} dominant phase.Comment: 9 pages, 11 figure

    Using job strain and organizational justice models to predict multiple forms of employee performance behaviours among Australian policing personnel

    The overall purpose of this investigation was to examine the relationship between stress-related working conditions and three forms of employee performance behaviours: in-role behaviours, citizenship behaviours directed at other individuals and citizenship behaviours directed at the organization. The potentially stressful working conditions were based on the job strain model (incorporating job demands, job control and social support) as well as organizational justice theory. A sample of Australian-based police officers (n = 640) took part in this study and the data were collected via a mail-out survey. Multiple regression analyses were undertaken to assess both the strength and the nature of the relationships between the working conditions and employee performance and these analyses included tests for additive, interactional and curvilinear effects. The overall results indicated that a significant proportion of the explained variance in all three outcome measures was attributed to the additive effects of demand, control and support. The level of variance associated with the organizational justice dimensions was relatively small, although there were signs that specific dimensions of justice may provide unique insights into the relationship between job stressors and employee performance. The implications of these and other notable findings are discussed.<br /
