    Rab11-mediated trafficking and human cancers: An updated review

    Many disorders block and subvert basic cellular processes in order to boost their pro-gression. One protein family that is prone to be altered in human cancers is the small GTPase RAB11 family, the master regulator of vesicular trafficking. RAB11 isoforms function as membrane organizers connecting the transport of cargoes towards the plasma membrane with the assembly of autophagic precursors and the generation of cellular protrusions. These processes dramatically impact normal cell physiology and their alteration significantly affects the survival, progression and metastatization as well as the accumulation of toxic materials of cancer cells. In this review, we dis-cuss biological mechanisms ensuring cargo recognition and sorting through a RAB11-dependent pathway, a prerequisite to understand the effect of RAB11 alterations in human cancers

    Canonical solution of a system of long-range interacting rotators on a lattice

    The canonical partition function of a system of rotators (classical X-Y spins) on a lattice, coupled by terms decaying as the inverse of their distance to the power alpha, is analytically computed.     Regeneration of the tropical legume Aeschynomene sensitiva Sw. from root explants

    Regeneration of #Aeschynomene sensitiva Sw. after callogenesis was obtained form small (2-5 mm long) root explants of 30-day-old seedlings aseptically cultivated on Murashige and Skoog medium supplemented with various concentrations of growth regulators. After 4 weeks, the best results were observed with 0.54 microM alpha-naphthaleneacetic acid and 2.22 microM benzyladenine. On this medium, the rate of regeneration depended on seedling age and agar concentration. The highest number of shoots per explant was obtained with small cuttings from 30-day-old seedlings grown on a medium containing 8 g/l of agar. Regeneration success was also dependent on explant size. When longer explants (7-20 mm) were cut from the main root, direct regeneration was obtained in two weeks. These cuttings also generated shoots through callogenesis in four weeks but always in lower quantities than with direct regeneration, whatever the seedling age. Here also, the best regeneration was obtained with cuttings from 30-day-old seedlings maintained on a medium with 8 g/l of agar. Regenerants were rooted on growth-regulator-free Murashige and Skoog medium and then acclimatized in a greenhouse. A better survival to transplantation was observed when plantlets were inoculated with the photosynthetic #Bradyrhizobium strain ORS 278.     Clinical insights into dermatofibrosarcoma protuberans

    Dermatofibrosarcoma protuberans (DFSP) is a rare, locally aggressive cutaneous soft tissue sarcoma, it is the second most common skin sarcoma after Kaposi's sarcoma. The cause of DFSP remains unknown. The case of a 54-year-old female patient with a diagnosis of Dermatofibrosarcoma protuberans is presented, displaying a typical clinical presentation. It is characterized by an initial lesion in the form of a reddish spot on the anterior region of the chest, which showed slow growth until the development of a multinodular and irregular lesion with multiple recurrences. The lesion is resected, confined to the superficial layers of the skin, with 3 cm margins, confirming the histopathological diagnosis of dermatofibrosarcoma protuberans with clear margins. DFSP is an uncommon cutaneous sarcoma that is typically low- to intermediate-grade, and while it has a limited likelihood of metastasis, it exhibits a notable tendency for local recurrence.     How PI3K-derived lipids control cell division

    To succeed in cell division, intense cytoskeletal and membrane remodeling are required to allow accurate chromosome segregation and cytoplasm partitioning. Spatial restriction of the actin dynamics and vesicle trafficking define the cell symmetry and equivalent membrane scission events, respectively. Protein complexes coordinating mitosis are recruited to membrane microdomains characterized by the presence of the phosphatidylinositol lipid members (PtdIns), like PtdIns(3,4,5)P3, PtdIns(4,5)P2 and PtdIns(3)P. These PtdIns represent a minor component of cell membranes, defining membrane domain identity, ultimately controlling cytoskeleton and membrane dynamics during mitosis. The coordinated presence of PtdIns(3,4,5)P3 at the cell poles and PtdIns(4,5)P2 at the cleavage furrow controls the polarity of the actin cytoskeleton leading to symmetrical cell division. In the endosomal compartment, the trafficking of PtdIns(3)P positive vesicles allows the recruitment of the protein machinery required for the abscission

    Persistent Spillback of Bovine Tuberculosis From White-Tailed Deer to Cattle in Michigan, USA: Status, Strategies, and Needs

    Free-ranging white-tailed deer (Odocoileus virginianus) are believed to be a self-sustaining reservoir for bovine tuberculosis (bTB) in northeastern Lower Michigan, USA. Although a comprehensive control program is in place and on-farm mitigation strategies to curtail bTB transmission between cattle and deer have been implemented for over a decade, cattle and deer continue to become infected with the disease. Thus, renewed motivation to eradicate bTB is needed if that is truly the goal. Recurrent detection of bTB in cattle in the region is of mounting concern for state and federal agricultural agencies, producers, and wildlife managers. Current on-farm mitigation efforts include fencing and refined cattle feeding and watering practices. Liberal removal of antlerless deer through hunter harvest and disease control permits (DCPs) issued to cattle producers and agency sharp shooters have also been ongoing. Although these strategies have merit and efforts to reduce prevalence in deer and occurrence of positive farms are elevated, additional actions are needed. Heightened management actions to combat bTB in deer could include deer vaccination programs, strategic habitat manipulations to redistribute deer from farms, and precision removal of deer in proximity to high-risk farms. Foundational research to address development and delivery of vaccine to free-ranging deer is complete. Strategic management and habitat manipulation could reduce and disperse local concentrations of deer while better meeting wildlife, forestry, and agricultural goals. The responses of local deer populations to targeted removal of individuals are generally understood and there is potential to reduce deer activity around agricultural operations while allowing them to persist nearby on natural foods. We summarize the history and progress to date, discuss the realized merit of novel management strategies, and suggest options to rid deer and cattle in Michigan of bTB

    Small molecules targeting endocytic uptake and recycling pathways

    Over the past years a growing number of studies highlighted the pivotal role of intracellular trafficking in cell physiology. Among the distinct transport itineraries connecting the endocytic system, both internalization (endocytosis) and recycling (endocytic recycling) pathways were found fundamental to ensure cellular sensing, cell-to-cell communication, cellular division, and collective cell migration in tissue specific-contexts. Consistently, the dysregulation of endocytic trafficking pathways is correlated with several human diseases including both cancers and neurodegeneration. Aimed at suppress specific intracellular trafficking routes involved in disease onset and progression, huge efforts have been made to identify small molecule inhibitors with suitable pharmacological properties for in vivo administration. Here, we review most used drugs and recently discovered small molecules able to block endocytosis and endocytic recycling pathways. We characterize such pharmacological inhibitors by emphasizing their target specificity, molecular affinity, biological activity and efficacy in both in vitro and in vivo experimental models

    Scaling laws for the largest Lyapunov exponent in long-range systems: A random matrix approach

    We investigate the laws that rule the behavior of the largest Lyapunov exponent (LLE) in many particle systems with long range interactions. We consider as a representative system the so-called Hamiltonian alpha-XY model where the adjustable parameter alpha controls the range of the interactions of N ferromagnetic spins in a lattice of dimension d. In previous work the dependence of the LLE with the system size N, for sufficiently high energies, was established through numerical simulations. In the thermodynamic limit, the LLE becomes constant for alpha greater than d whereas it decays as an inverse power law of N for alpha smaller than d. A recent theoretical calculation based on Pettini's geometrization of the dynamics is consistent with these numerical results (M.-C. Firpo and S. Ruffo, cond-mat/0108158). Here we show that the scaling behavior can also be explained by a random matrix approach, in which the tangent mappings that define the Lyapunov exponents are modeled by random simplectic matrices drawn from a suitable ensemble.Comment: 5 pages, no figure
