    Volatile Chemicals of Adults and Nymphs of the Eucalyptus

    Thaumastocoris peregrinus is an introduced “true bug” that is now a severe pest in Eucalyptus plantations of various Southern Hemisphere countries. The semiochemicals of thaumastocorids are completely unknown. Therefore, volatile chemicals from T. peregrinus nymphs and adults were identified as possible leads for pheromones potentially useful for control. The contents of nymphal exocrine glands, which are shed at molting, were identified from extracts of exuviae. Adults lack functional metathoracic scent glands that are characteristic of most heteropterans; however, both males and females possess a glandular-appearing hold-fast organ that they quickly extrude posteriorly when disturbed. Whole body hexane extracts from males and females were prepared by freezing the insects in a flask so that they extruded the hold-fast organ, and then they were extracted with hexane. Volatiles from nymphal exuviae included benzaldehyde, octanol, (E)-2-octenol, octanoic acid, decanal, and hexanoic acid. Adult volatiles included 3-methylbut-2-en-1-yl butyrate and 3-methylbut-3-en-1-yl butyrate

    Headspace solid-phase microextraction procedure for gas-chromatography analysis of toluene in urine

    Toluene is widely used in industrial and laboratory applications and in many countries is related with social problems of abuse. Unaltered urinary toluene was introduced as a bioindicator of occupational exposure to the solvent, but its analysis presents difficulties due to the low levels of the compound excreted in urine. A gas-chromatography/flame ionization method for toluene in urine is described using headspace solid-phase microextraction and establishing the better conditions for two different extracting phases: polydimethylsiloxane (PDMS) and carboxen-PDMS. The carboxen-PDMS fiber showed lower quantifying limit (12.5 ng mL-1), better extraction efficiency (up to 28.1%) and repeatability (CV < 4.9%) than PDMS coating. The method was applied to analysis of toluene in urine of workers from car repair shops exposed to low solvent levels

    Shortened first-line TB treatment in Brazil: potential cost savings for patients and health services.

    BACKGROUND: Shortened treatment regimens for tuberculosis are under development to improve treatment outcomes and reduce costs. We estimated potential savings from a societal perspective in Brazil following the introduction of a hypothetical four-month regimen for tuberculosis treatment. METHODS: Data were gathered in ten randomly selected health facilities in Rio de Janeiro. Health service costs were estimated using an ingredient approach. Patient costs were estimated from a questionnaire administered to 126 patients. Costs per visits and per case treated were analysed according to the type of therapy: self-administered treatment (SAT), community- and facility-directly observed treatment (community-DOT, facility-DOT). RESULTS: During the last 2 months of treatment, the largest savings could be expected for community-DOT; on average USD 17,351-18,203 and USD 43,660-45,856 (bottom-up and top-down estimates) per clinic. Savings to patients could also be expected as the median (interquartile range) patient-related costs during the two last months were USD 108 (13-291), USD 93 (36-239) and USD 11 (7-126), respectively for SAT, facility-DOT and community-DOT. CONCLUSION: Introducing a four-month regimen may result in significant cost savings for both the health service and patients, especially the poorest. In particular, a community-DOT strategy, including treatment at home, could maximise health services savings while limiting patient costs. Our cost estimates are likely to be conservative because a 4-month regimen could hypothetically increase the proportion of patients cured by reducing the number of patients defaulting and we did not include the possible cost benefits from the subsequent prevention of costs due to downstream transmission averted and rapid clinical improvement with less side effects in the last two months

    Myosteatosis in a systemic inflammation-dependent manner predicts favorable survival outcomes in locally advanced esophageal cancer

    Increased adiposity and its attendant metabolic features as well as systemic inflammation have been associated with prognosis in locally advanced esophageal cancer (LAEC). However, whether myosteatosis and its combination with systemic inflammatory markers are associated with prognosis of esophageal cancer is unknown. Our study aimed to investigate the influence of myosteatosis and its association with systemic inflammation on progression-free survival (PFS) and overall survival (OS) in LAEC patients treated with definitive chemoradiotherapy (dCRT). We retrospectively gathered information on 123 patients with LAEC submitted to dCRT at the University of Campinas Hospital. Computed tomography (CT) images at the level of L3 were analyzed to assess muscularity and adiposity. Systemic inflammation was mainly measured by calculating the neutrophil-to-lymphocyte ratio (NLR). Median PFS for patients with myosteatosis (n = 72) was 11.0 months vs 4.0 months for patients without myosteatosis (n = 51) (hazard ratio [HR]: 0.53; 95% confidence interval [CI], 0.34-0.83; P = .005). Myosteatosis was also independently associated with a favorable OS. Systemic inflammation (NLR > 2.8) was associated with a worse prognosis. The combination of myosteatosis with systemic inflammation revealed that the subgroup of patients with myosteatosis and without inflammation presented less than half the risk of disease progression (HR: 0.47; 95% CI: 0.26-0.85; P = .013) and death (HR: 0.39; 95% CI, 0.21-0.72; P = .003) compared with patients with inflammation.     The Transcriptome of the Salivary Glands of Amblyomma aureolatum Reveals the Antimicrobial Peptide Microplusin as an Important Factor for the Tick Protection Against Rickettsia rickettsii Infection

    The salivary glands (SG) of ixodid ticks play a pivotal role in blood feeding, producing both the cement and the saliva. The cement is an adhesive substance that helps the attachment of the tick to the host skin, while the saliva contains a rich mixture of antihemostatic, anti-inflammatory, and immunomodulatory substances that allow ticks to properly acquire the blood meal. The tick saliva is also a vehicle used by several pathogens to be transmitted to the vertebrate host, including various bacterial species from the genus Rickettsia. Rickettsia rickettsii is a tick-borne obligate intracellular bacterium that causes the severe Rocky Mountain spotted fever. In Brazil, the dog yellow tick Amblyomma aureolatum is a vector of R. rickettsii. In the current study, the effects of an experimental infection with R. rickettsii on the global gene expression profile of A. aureolatum SG was determined by next-generation RNA sequencing. A total of 260 coding sequences (CDSs) were modulated by infection, among which 161 were upregulated and 99 were downregulated. Regarding CDSs in the immunity category, we highlight one sequence encoding one microplusin-like antimicrobial peptide (AMP) (Ambaur-69859). AMPs are important effectors of the arthropod immune system, which lack the adaptive response of the immune system of vertebrates. The expression of microplusin was confirmed to be significantly upregulated in the SG as well as in the midgut (MG) of infected A. aureolatum by a quantitative polymerase chain reaction preceded by reverse transcription. The knockdown of the microplusin expression by RNA interference caused a significant increase in the prevalence of infected ticks in relation to the control. In addition, a higher rickettsial load of one order of magnitude was recorded in both the MG and SG of ticks that received microplusin-specific dsRNA. No effect of microplusin knockdown was observed on the R. rickettsii transmission to rabbits. Moreover, no significant differences in tick engorgement and oviposition were recorded in ticks that received dsMicroplusin, demonstrating that microplusin knockdown has no effect on tick fitness. Further studies must be performed to determine the mechanism of action of this AMP against R. rickettsii

    Este texto explora a ideia de que a evolução mundial nos próximos anos vai ser marcada pela interacção complexa entre, por um lado as tensões associadas à Globalização da Economia Mundial, e por outro as Incertezas em torno da Fragmentação Geopolítica Mundial. Começa por identificar os grandes processos envolvidos na primeira "força motriz" - uma ampliação e "regionalização" da Economia Mundial; uma dinâmica de Globalização económica; uma competição acesa entre "Modelos de Capitalísmo"; uma mutação tecnológica abrangente, que modifica as estruturas económicas e a posição relativa das economias; e por último uma regulação económica global que procura responder à acumulação de tensões geradas pela interacção dos processos anteriores. Seguidamente identifica alguns processos chave que organizam a segunda" força motriz", como sejam o avanço da democratização, decorrendo em paralelo com a sobreposição de crises profundas em diversos Estados; um processo de fragmentação e "regionalização" em termos geopolíticos e de segurança; uma alteração na relação de forças entre potências, que está ainda numa fase inconclusiva; uma mutação tecnológica militar que pode influenciar decisivamente essa alteração; e a manifestação de dificuldades na regulação estratégica e geopolítica mundial, pela interacção dos processos anteriores e no contexto da ultrapassagem dos mecanismos de regulação típicos da guerra fria. Por último o texto ilustra algumas das interacções que se podem estabelecer entre as dinâmicas das duas "forças motrizes" sem explorar em profundidade o tema
