    Cierto es que el saber de la Educación Física y sus áreas afines como el deporte, la recreación y la actividad física, entre otros, pasa por distintos lugares que se constituyen en escenarios para la comprensión del devenir del campo, así como para la proyección de lo que ha de ser en términos de producción investigativa y desarrollo teórico de las disciplinas. Tales escenarios, desde el saber y el quehacer docente, intentan responder a las necesidades de la sociedad actual, en ocasiones tan voluble, compleja y efímera, resaltando con ello la tarea de los académicos del área. Asimismo, configuran nodos de discusión que, si bien es cierto han sido abordados hace tiempo, se encuentran a la orden del día como reinterpretaciones, resignificaciones e innovaciones teóricas con base investigativa; es así como la competencia deportiva da lugar a nuevos cuestionamientos sobre los métodos de entrenamiento, las respuestas fisiológicas a la carga o el lugar del entrenamiento en el desarrollo multidimensional de los deportistas

    Single-Center Retrospective Analysis in Sixty-Four Patients

    Objectives. Portal vein embolization (PVE) stimulates hypertrophy of the future liver remnant (FLR) and improves the safety of extended hepatectomy. This study evaluated the efficacy of PVE, performed with PVA and coils, in relation to its effect on FLR volume and ratio. Secondary endpoints were the assessment of PVE complications, accomplishment of liver surgery, and patient outcome after hepatectomy. Materials and Methods. All patients who underwent PVE before planned major hepatectomy between 2013 and 2017 were retrospectively analyzed, comprising a total of 64 patients. Baseline patient clinical characteristics, imaging records, liver volumetric changes, complications, and outcomes were analyzed. Results. There were 45 men and 19 women with a mean age of 64 years. Colorectal liver metastasis was the most frequent liver tumor. The majority of patients (n = 53) had a right PVE. FLR increased from a mean value of 484 ml ± 242 to 654 ml ± 287 p<0.001 after PVE. Two major complications were experienced after PVE: 1 case of left hepatic artery branch laceration and 1 case of hemoperitoneum and hemothorax. A total of 44 (69%) patients underwent liver surgery. Twenty-one patients were not taken to surgery due to disease progression (n = 18), liver insufficiency (n = 1), and insufficient FLR volume (n = 1), and one patient declined surgery (n = 1). Conclusions. PVE with PVA and coils was accomplished safely and promoted a high FLR hypertrophy yield, enabling most of our patients to be submitted to the potentially curative treatment of liver tumor resection.publishersversionpublishe

    Centrosomes are organelles consisting of two structurally and functionally distinct centrioles, with the mother centriole having complex distal (DA) and subdistal appendages (SDA). Despite their importance, how appendages are assembled and maintained remains unclear. This study investigated human TBCCD1, a centrosomal protein essential for centrosome positioning, to uncover its localization and role at centrioles. We found that TBCCD1 localizes at both proximal and distal regions of the two centrioles, forming a complex structure spanning from SDA to DA and extending inside and outside the centriole lumen. TBCCD1 depletion caused centrosome mispositioning, which was partially rescued by taxol, and the loss of microtubules (MTs) anchored to centrosomes. TBCCD1 depletion also reduced levels of SDA proteins involved in MT anchoring such as Centriolin/CEP110, Ninein, and CEP170. Additionally, TBCCD1 was essential for the correct positioning of motile cilia basal bodies and associated structures in Paramecium. This study reveals that TBCCD1 is an evolutionarily conserved protein essential for centriole and basal body localization and appendage assembly and maintenance.     MFGE8 does not influence chorio-retinal homeostasis or choroidal neovascularization in vivo

    Purpose: Milk fat globule-epidermal growth factor-factor VIII (MFGE8) is necessary for diurnal outer segment phagocytosis and promotes VEGF-dependent neovascularization. The prevalence of two single nucleotide polymorphisms (SNP) in MFGE8 was studied in two exsudative or “wet” Age-related Macular Degeneration (AMD) groups and two corresponding control groups. We studied the effect of MFGE8 deficiency on retinal homeostasis with age and on choroidal neovascularization (CNV) in mice. Methods: The distribution of the SNP (rs4945 and rs1878326) of MFGE8 was analyzed in two groups of patients with “wet” AMD and their age-matched controls from Germany and France. MFGE8-expressing cells were identified in Mfge8+/− mice expressing ß-galactosidase. Aged Mfge8+/− and Mfge8−/− mice were studied by funduscopy, histology, electron microscopy, scanning electron microscopy of vascular corrosion casts of the choroid, and after laser-induced CNV. Results: rs1878326 was associated with AMD in the French and German group. The Mfge8 promoter is highly active in photoreceptors but not in retinal pigment epithelium cells. Mfge8−/− mice did not differ from controls in terms of fundus appearance, photoreceptor cell layers, choroidal architecture or laser-induced CNV. In contrast, the Bruch's membrane (BM) was slightly but significantly thicker in Mfge8−/− mice as compared to controls. Conclusions: Despite a reproducible minor increase of rs1878326 in AMD patients and a very modest increase in BM in Mfge8−/− mice, our data suggests that MFGE8 dysfunction does not play a critical role in the pathogenesis of AMD

    Allergic Rhinitis and its Associated Co-Morbidities at Bugando Medical Centre in Northwestern Tanzania; A Prospective Review of 190 Cases.

    Allergic rhinitis is one of the commonest atopic diseases which contribute to significant morbidity world wide while its epidemiology in Tanzania remains sparse. There was paucity of information regarding allergic rhinitis in our setting; therefore it was important to conduct this study to describe our experience on allergic rhinitis, associated co-morbidities and treatment outcome in patients attending Bugando Medical Centre. This was descriptive cross-sectional study involving all patients with a clinical diagnosis of allergic rhinitis at Bugando Medical Centre over a three-month period between June 2011 and August 2011. Data was collected using a pre-tested coded questionnaire and analyzed using SPSS statistical computer software version 17.0. A total of 190 patients were studied giving the prevalence of allergic rhinitis 14.7%. The median age of the patients was 8.5 years. The male to female ratio was 1:1. Adenoid hypertrophy, tonsillitis, hypertrophy of inferior turbinate, nasal polyps, otitis media and sinusitis were the most common co-morbidities affecting 92.6% of cases and were the major reason for attending hospital services. Sleep disturbance was common in children with adenoids hypertrophy (χ2 = 28.691, P = 0.000). Allergic conjunctivitis was found in 51.9%. The most common identified triggers were dust, strong perfume odors and cold weather (P < 0.05). Strong perfume odors affect female than males (χ2 = 4.583, P = 0.032). In this study family history of allergic rhinitis was not a significant risk factor (P =0.423). The majority of patients (68.8%) were treated surgically for allergic rhinitis co morbidities. Post operative complication and mortality rates were 2.9% and 1.6% respectively. The overall median duration of hospital stay of in-patients was 3 days (2 - 28 days). Most patients (98.4%) had satisfactory results at discharge. The study shows that allergic rhinitis is common in our settings representing 14.7% of all otorhinolaryngology and commonly affecting children and adolescent. Sufferers seek medical services due to co-morbidities of which combination of surgical and medical treatment was needed. High index of suspicions in diagnosing allergic rhinitis and early treatment is recommended

    Unraveling the role of TBCCD1 protein on cell size control: the regulation of cytoskeleton dynamics and cell junctions

    During their lifetime most cells maintain their size. There is increasing evidence showing that this process may be dynamic and that cells can adapt their size in response to external signals and changes in the environment [1], which strongly suggests that cell size is regulated. Both Hippo and IGF/PI3K/AKT/mTORC1 pathways have been described as being involved in cell size/growth control [1]. Interestingly, these pathways are in a cross-talk with others involved and/or dependent on cellular polarity [2]. Our group characterized a centrosomal protein, TBCCD1 (TBCC domain – containing human protein 1) which, when depleted in human retinal epithelial (RPE–1) cells, leads to an abnormal localization of the centrosome at the cell periphery accompanied by the fragmentation of the Golgi apparatus, resulting in the disruption of the intrinsic cell polarity axis “Nucleus-Centrosome-Golgi Apparatus”. Moreover, TBCCD1 – depleted cells are larger, slower and have a lower efficiency in primary cilia assembly than control cells [3]. We identified the TBCCD1 interactome that showed that most of its partners are involved in cell polarity. Furthermore, most of them participate in the formation/maintenance of cell junctions, which are main regulators of cell polarity in epithelia and are upstream of pathways, like Hippo pathway. We also observed that TBCCD1 overexpression affects tubulin acetylation, which supports our results showing that some of the partners are involved in the regulation of the cytoskeleton dynamics, which may affect cell size. Therefore, it is tempting to hypothesize that the mechanisms involved in the establishment of intrinsic cell polarity may also directly/indirectly participate in the regulation of cell size.info:eu-repo/semantics/publishedVersio
