    Cytogenetic characterization of Partamona cupira (Hymenoptera, Apidae) by fluorochromes

    Four colonies of the stingless bee Partamona cupira (Hymenoptera: Apidae) were cytogenetically analyzed using conventional staining and the fluorochromes CMA3 e DAPI. The females have 2n = 34 chromosomes (2K = 32 M¯+2 A¯). Some females, however, presented an additional large B acrocentric chromosome, to a total of 2n = 35. Chromosome B and the chromosomal pairs 2, 9 and 10 showed CMA 3+ bands, indicating an excess of CG base-pairs. A clear association was verified between the P. helleri B chromosome SCAR marker and the presence of a B chromosome in P. cupira. The data obtained suggests that B chromosomes in P. helleri and P. cupira share a common origin

    Dysfunctional LAT2 Amino Acid Transporter Is Associated With Cataract in Mouse and Humans.

    Cataract, the loss of ocular lens transparency, accounts for ∼50% of worldwide blindness and has been associated with water and solute transport dysfunction across lens cellular barriers. We show that neutral amino acid antiporter LAT2 (Slc7a8) and uniporter TAT1 (Slc16a10) are expressed on mouse ciliary epithelium and LAT2 also in lens epithelium. Correspondingly, deletion of LAT2 induced a dramatic decrease in lens essential amino acid levels that was modulated by TAT1 defect. Interestingly, the absence of LAT2 led to increased incidence of cataract in mice, in particular in older females, and a synergistic effect was observed with simultaneous lack of TAT1. Screening SLC7A8 in patients diagnosed with congenital or age-related cataract yielded one homozygous single nucleotide deletion segregating in a family with congenital cataract. Expressed in HeLa cells, this LAT2 mutation did not support amino acid uptake. Heterozygous LAT2 variants were also found in patients with cataract some of which showed a reduced transport function when expressed in HeLa cells. Whether heterozygous LAT2 variants may contribute to the pathology of cataract needs to be further investigated. Overall, our results suggest that defects of amino acid transporter LAT2 are implicated in cataract formation, a situation that may be aggravated by TAT1 defects

    The role of growth factors on renal tubular cells submitted to hypoxia and deprived of glucose

    Background. in acute renal failure (ARF) renal tubular cell death and detachment can be induced by necrotic and apoptotic mechanisms. Several studies have demonstrated some benefits of the use of growth factors in experimental models of ARF. Methods. MDCK cells were cultured in a glucose-free medium for 24 h and were submitted to hypoxia (pO(2) around 35 mmHg) for additional 24 h. To evaluate the possible protective role of growth factors, EGF, IGF-I or HGF were added to the medium (20 ng mL). LDH release, viability (acridine orange and ethidium bromide dyes) and quantification of apoptotic cells (Hoechst 33342 dye fluorescence) were determined. Results. in the injury group, an increase on LDH release (60% vs. 3%) and on number of apoptotic cells (22% vs. 0.2%) which was associated with a reduced cell viability (61% vs. 94%) when compared with controls. Only HGF, not EGF or IGF-I, was able to protect cells from injury. HGF caused a significant reduction on LDH release (30%) and on number of apoptotic cells (5%), with an increase on viability cellular (79%). Conclusions.     OBJETIVO: avaliar os correlatos perceptivo-auditivos (ajustes supraglóticos) e acústicos (formantes) da qualidade vocal de indivíduos disfônicos com alterações glóticas, atendidos em ambulatório de fonoaudiologia de uma instituição hospitalar da cidade de São Paulo. MÉTODOS: o grupo estudado foi representado por amostras de voz de 20 indivíduos disfônicos com idades variadas entre 30 e 65 anos do sexo feminino (atendidos em um hospital da cidade de São Paulo) e julgamentos perceptivo-auditivos da qualidade vocal por amostras avaliadas previamente por um grupo de juízes em atividade clínica. As amostras foram exploradas quanto à freqüência dos três primeiros formantes (F1, F2 e F3). Os valores foram considerados de forma qualitativa (comparados aos dados da análise perceptivo-auditiva) e quantitativa (teste Qui-quadrado). RESULTADOS: os ajustes de qualidade vocal mais freqüentes foram laringe baixa, mandíbula fechada, lábios arredondados, corpo de língua abaixado, corpo de língua retraído, labiodentalização. Diferenças estatisticamente significantes foram encontradas para os valores de freqüência de F1 para os vocábulos "cata" (864 Hz) e "gata" (814 Hz), p=0 e F3 para os vocábulos "cata" (2598 Hz) e "gata" (2660 Hz), p=0,17 dos indivíduos do grupo estudado em relação aos valores de referência para o Português-Brasileiro. CONCLUSÃO: os ajustes de qualidade vocal identificados encontraram correspondência com a diminuição dos valores de F1 e F3 indicando tendências de ajustes do trato vocal.<br>PURPOSE: to evaluate auditory perception (supraglottic adjustments) and acoustic (formants) of vocal quality in dysphonic individuals with glotic disorders, attended in a clinic a hospital in the city of São Paulo. METHODS: the studied was represented by voice samples taken form from 20 dysphonic people aged between 30 and 65 of the female gender (attended in a hospital in the city of Sao Paulo) and the auditory perception judgment of the vocal quality was represented by samples previously evaluated by active judges in clinic activity. The samples were explored as for the frequency of the first three formants (F1, F2 and F3). The values were considered as for their quality (compared to the data of auditory perception analysis) and their quantity (Qui-square test) RESULTS: the more frequent adjustments of vocal quality were low larynx, closed jaw, lip rounding, lowered tongue body, retracted tongue body, labiodentalization. Statistically meaningful differences were found for the frequency value of F1 for the syllables "cata" (864 Hz) and "gata" (814Hz), p=0 and F3 for the syllables "cata" (2598 Hz) and "gata"(2660 Hz), p=0.17 for the subjects of the group studied with relation to the values of reference for Portuguese-Brazilian language. CONCLUSION: the identified adjustments of vocal quality met correspondence with the reducing of values of F1 and F3 indicating trends for vocal treat adjustments

    Membrane transporters influence biological functions in the ocular lens. Here, we investigate the monocarboxylate transporter 12 (MCT12), also called creatine transporter 2 (CRT2), which is found in the ocular lens and is involved in cataract. As the age-related form affects about half of the population world-wide, understanding relevant pathomechanisms is a prerequisite for exploring non-invasive treatments. We screened the coding exons of the gene SLC16A12 in 877 patients from five cohorts, including Caucasian and Asian ethnicities. A previously identified risk factor, SNP rs3740030, displayed different frequencies in the Asian cohorts but risk could not be established. In 15 patients 13 very rare heterozygous nucleotide substitutions were identified, of which eight led to non-synonymous and four to synonymous amino acid exchanges and one mapped to the canonical splice site in intron 3. Their impact on creatine transport was tested in Xenopus laevis oocytes and human HEK293T cells. Four variants (p.Ser158Pro, p.Gly205Val, p.Pro395Gln and p.Ser453Arg) displayed severe reduction in both model systems, indicating conserved function. Two of these, p.Gly205Val, and p.Ser453Arg, did not localize to the oocyte membrane, suggesting possible impacts on protein interactions for transporter processing. In support, exogenously supplied excess of MCT12's chaperone CD147 in HEK293T cells led to a partial recovery of the defective uptake activity from p.Gly205Val and also from mutant p.Pro395Gln, which did localize to the membrane. Our findings provide first insight in the molecular requirements of creatine transporter, with particular emphasis on rescuing effects by its chaperone CD147, which can provide useful pharmacological information for substrate delivery

    Antioxidantes da dieta como inibidores da nefrotoxicidade induzida pelo antitumoral cisplatina Dietary antioxidants as inhibitors of cisplatin-induced nephrotoxicity

    A cisplatina é uma droga antineoplásica altamente efetiva contra vários tipos de cânceres humanos, tais como tumores do testículo e ovário, câncer da cabeça e pescoço e câncer do pulmão. Entretanto, a nefrotoxicidade é um dos principais efeitos colaterais da terapia com a cisplatina. A gravidade da nefrotoxicidade induzida pela cisplatina está relacionada com a concentração de platina nos rins. As evidências mostram que a nefrotoxicidade induzida pela cisplatina é atribuída ao dano oxidativo resultante da geração de radicais livres, e que a administração de antioxidantes é eficiente na inibição destes efeitos colaterais. Uma abordagem alternativa para proteger os roedores dos efeitos colaterais da cisplatina é o uso de conhecidos antioxidantes da dieta. Alguns estudos têm sido realizados para diminuir a peroxidação lipídica e os efeitos citotóxicos induzidos pela cisplatina, com o emprego de antioxidantes da dieta, tais como, selenito de sódio, vitaminas C e E, curcumina e o carotenóide bixina. Nós sugerimos que aqueles antioxidantes da dieta têm efeito nefroprotetor, e que os mecanismos antioxidantes destes compostos deveriam ser explorados durante a quimioterapia com a cisplatina.<br>Cisplatin is a highly effective antineoplastic drug used against several types of human cancers, such as testicular and ovarian tumors; head and neck; and lung cancer. However, nephrotoxicity is one of the most important side-effects of cisplatin therapy. The severity of cisplatin nephrotoxicity is related to platinum concentration in the kidneys. There is a growing amount of evidence that cisplatin-induced nephrotoxicity is ascribed to oxidative damage resulting from free radical generation and that the administration of antioxidants is efficient in inhibiting these side effects. An alternative approach aiming to protect rodents against cisplatin side-effects is the introduction of known dietary antioxidants. Some studies have been conducted to decrease cisplatin-induced lipid peroxidation and cytotoxic effects by using such dietary antioxidants, including sodium selenite; vitamins C and E; curcumin and the carotenoid bixin. We suggest that these dietary antioxidants have a nephroprotective effect, and that their antioxidant mechanisms should be further explored during cisplatin chemotherapy
