    Subfractionation of human very low density lipoproteins by heparin-Sepharose affinity chromatography.

    Very low density lipoproteins obtained from normolipidemic subjects were fractionated into subclasses by means of affinity chromatography on a heparin-Sepharose column in the presence of MnCl2. The four subfractions eluted at 0.05, 0.12, 0.20, and 0.38 M NaCl and they differed in chemical composition and apoprotein pattern. The relative amounts of apoB and apoE in subfractions increased with increasing concentrations of the NaCl eluant. Modification of the arginyl residues with 1-2 cyclohexadione demonstrated that arginine plays an important role in determining the elution pattern of VLDL. In vitro studies indicated that only fractions eluted at 0.2 and 0.5 M NaCl compete with LDL for cellular receptors. These data suggest that the various subfractions may represent VLDL at different stages of catabolism

    In the context of allogeneic transplant platforms, human leukocyte antigen (HLA)-haploidentical hematopoietic stem cell transplantation (haplo-HSCT) represents one of the latest and most promising curative strategies for patients affected by high-risk hematologic malignancies. Indeed, this platform ensures a suitable stem cell source immediately available for virtually any patents in need. Moreover, the establishment in recipients of a state of immunologic tolerance toward grafted hematopoietic stem cells (HSCs) remarkably improves the clinical outcome of this transplant procedure in terms of overall and disease free survival. However, the HLA-mismatch between donors and recipients has not been yet fully exploited in order to optimize the Graft vs. Leukemia effect. Furthermore, the efficacy of haplo-HSCT is currently hampered by several life-threatening side effects including the onset of Graft vs. Host Disease (GvHD) and the occurrence of opportunistic viral infections. In this context, the quality and the kinetic of the immune cell reconstitution (IR) certainly play a major role and several experimental efforts have been greatly endorsed to better understand and accelerate the post-transplant recovery of a fully competent immune system in haplo-HSCT. In particular, the IR of innate immune system is receiving a growing interest, as it recovers much earlier than T and B cells and it is able to rapidly exert protective effects against both tumor relapses, GvHD and the onset of life-threatening opportunistic infections. Herein, we review our current knowledge in regard to the kinetic and clinical impact of Natural Killer (NK), \u3b3\u3b4 and Innate lymphoid cells (ILCs) IRs in both allogeneic and haplo-HSCT. The present paper also provides an overview of those new therapeutic strategies currently being implemented to boost the alloreactivity of the above-mentioned innate immune effectors in order to ameliorate the prognosis of patients affected by hematologic malignancies and undergone transplant procedures

    ATR-FTIR chemical screening for adulterants and sugar characterisation in honeys

    The search for improvements in honey analysis is an important topic due to the relevance that adulterations and variations of sugar composition have in this food. Attenuated Total Reflectance Fourier Transform InfraRed spectroscopy (ATR-FTIR) is a reliable and fast analytical instrumentation for analysis of liquids, semi-solids and solids, which is currently used in food chemistry for qualitative and quantitative investigations. This work is aimed to build a fast analytical method, using ATR-FTIR and Partial Least Square (PLS) chemometric tool, to quantitative determination of glucose, fructose and sucrose present in honeys. We analyzed 25 honeys from Trentino and Sicily and a significant spectral range from the whole ATR-FTIR spectra were selected. Then a PLS multivariate calibration model was built by using mixtures of glucose, fructose and sucrose at various concentration. Samples for cross-validation were selected, thereby allowing quantitative evaluation of glucose, fructose and sucrose in all samples. Furthermore, a representative number of honeys were mixed with common adulterant syrups to simulate adulteration procedures and to verify the reliability of the ATR-FTIR technique in recognizing them. Preliminary results revealed not only the quantitative reliability of ATR-FTIR spectroscopy in the evaluation of sugars in honeys, but also its ability in recognizing honey adulteration

    The impact of the first powering cycles on the LHC superconducting dipoles coil geometry has been investigated. Dedicated magnetic measurements have been performed to estimate the changes in the geometric component of the harmonics, caused by the last highest Lorentz force the coil has ever experienced. Several magnets have been powered at increasing current steps while the field quality has been measured to quantify the changes in geometry. The effect of the thermal cycles has been also considered as well as the systematic differences between measurements before and after the quench training. The reconstruction of the coil geometry variations is discussed in terms of continuous modes of deformation as well as single block displacements

    The scent of emotions: A systematic review of human intra- and interspecific chemical communication of emotions.

    The sense of olfaction has been considered of minor importance in human communication. In recent years, evidence has emerged that humans might be influenced by unconscious messages sent through chemosignals in body odors. Data concerning the ability of humans to recognize fear, maybe related to the evolutionary role of these emotions in the fight-or-flight reactions, are well known. To further understand the role of emotional chemosignals in mediating communication in humans and its influence on animal behaviors, we conducted a systematic literature review. Chemosignals derived from axillary odors collected under a variety of emotional stimuli and sad tears in humans affect receivers' social interactions, danger detection and risk-taking behavior, social aspects of eating, and performance under stressing conditions. In addition, beyond the fight-or-flight response, even the body odors of happiness can be perceived by others. Furthermore, human chemosignals can influence behaviors and stressful responses in animals, particularly dogs and horses, which may partially explain their special relationship with humans. Our review highlights the importance of chemosignaling in human intra- and interspecific interactions and suggests the need for further investigations, both in physiological conditions and in patients with psychiatric or neurodegenerative disorders

    Patient controlled pressure support ventilation

    Introduction Pressure support ventilation is mostly used in weaning from mechanical ventilaton in acute respiratory failure. There are no data regarding the optimal level of assistance for each patient in different clinical conditions. We approach a new method that allows patients to set their own PSV level by a remote control connected to the ventilator. Materials and methods In 9 awake intubated patients (age 57 \ub1 17 years, BMI 24 \ub1 3 kg/m2, PaO2/FiO2 283 \ub1 51, Ramsey 1.9 \ub1 0.4) we measured the breathing pattern (VT, RR), the work of breathing (WOB J/min, estimated from a modified Campbell diagram [1]) and the dyspnea sensation using the Borg visual scale [2]. Patients were studied at three fixed levels of PSV (5-15-25 cmH2O). At the end of each step we gave the patient the possibility to change the level of PSV using the remote control. Patients were previously instructed by attendant physician about the use of the remote control. Results See Table and Figure. It appears that increasing the pressure support level, the patient work of breathing decreases while the Borg dyspnea scale shows no significant differences. Figure. Conclusions Patient controlled PSV could be a useful technique in ventilatory management of critically ill awake patients

    Steering the Field Quality in the Production of the Main Quadrupoles of the Large Hadron Collider

    The main issues concerning the field quality in the main quadrupoles of the Large Hadron Collider are presented. We show the trend plots for the focusing strength and multipoles at room temperature covering more than 2/3 of the production. We describe the correction of the coil layout to improve b6 at injection field level. A non-negligible fraction of the quadrupoles has been manufactured with collars featuring a magnetic permeability somewhat higher than the specified limits. We show plots for this anomaly. Field quality correlations to measurements in operational conditions are discussed. The dependence of field quality on cable manufacturer is analyse
