    This paper focusses on the mosaics of the crypt of the St. Nicola’s Basilica in Bari, a valuable evidence of use and reuse of ancient white and coloured marbles from the Roman world, together with local and imitation stones. The study belongs to a wider research project (MARMORA), about ancient marbles employed in the Apulia Cultural Heritage, and aims to improve knowledge and preserve these artworks, in order to enhance their valorisation and enjoyment. Therefore, firstly a high definition survey of mosaic floors was performed and after, characterization and provenance studies of stone tesserae, recognition of geometrical motifs and stylistic influence were carried out.     The gustin (CA6) gene polymorphism, rs2274333 (A/G), as a mechanistic link between PROP tasting and fungiform taste papilla density and maintenance

    Taste sensitivity to PROP varies greatly among individuals and is associated with polymorphisms in the bitter receptor gene TAS2R38, and with differences in fungiform papilla density on the anterior tongue surface. Recently we showed that the PROP non-taster phenotype is strongly associated with the G variant of polymorphism rs2274333 (A/G) of the gene that controls the salivary trophic factor, gustin. The aims of this study were 1) to investigate the role of gustin gene polymorphism rs2274333 (A/G), in PROP sensitivity and fungiform papilla density and morphology, and 2) to investigate the effect of this gustin gene polymorphism on cell proliferation and metabolic activity. Sixty-four subjects were genotyped for both genes by PCR techniques, their PROP sensitivity was assessed by scaling and threshold methods, and their fungiform papilla density, diameter and morphology were determined. In vitro experiments examined cell proliferation and metabolic activity, following treatment with saliva of individuals with and without the gustin gene mutation, and with isolated protein, in the two iso-forms. Gustin and TAS2R38 genotypes were associated with PROP threshold (p=0.0001 and p=0.0042), but bitterness intensity was mostly determined by TAS2R38 genotypes (p<0.000001). Fungiform papillae densities were associated with both genotypes (p<0.014) (with a stronger effect for gustin; p=0.0006), but papilla morphology was a function of gustin alone (p<0.0012). Treatment of isolated cells with saliva from individuals with the AA form of gustin or direct application of the active iso-form of gustin protein increased cell proliferation and metabolic activity (p<0.0135). These novel findings suggest that the rs2274333 polymorphism of the gustin gene affects PROP sensitivity by acting on fungiform papilla development and maintenance, and could provide the first mechanistic explanation for why PROP super-tasters are more responsive to a broad range of oral stimul

    Quotas regulation is necessary but not sufficient to mitigate the impact of SCUBA diving in a highly visited marine protected area

    When effectively managed, Marine Protected Areas (MPAs) can produce wide ecosystem benefits that can foster, directly and indirectly, local economies. Tourism is one of the sectors mainly benefited by the effect of conservation. SCUBA diving represents an important tourism activity, especially in the context of MPAs, where it is one of the few activities often fostered rather than limited, for its capacity to integrate environmental and socio-economic sustainability. However, SCUBA diving can also produce negative impacts on the environment when tourism frequentation exceeds a sustainable threshold, these potentially generating negative effects on the sector itself. In this study, we (1) investigated the impact of SCUBA diving in one of the most frequented diving areas of the Mediterranean Sea (Cabo de Palos - Islas Hormigas marine reserve), and (2) assessed the potential benefits over time related to the adoption of a regulation change for the diving activity (i.e., formally adoption of diving quotas). Specifically, we compared demographic (density of alive and dead colonies) and morphometric (height, width and complexity) characteristics of the false coral (Myriapora truncata) between dived and fully protected (non-dived) locations over four diving seasons (one before and three after the change in diving quotas). The density of alive colonies of the false coral was, on average, six times lower in dived locations compared to controls, highlighting a clear impact of SCUBA diving (consistent over time). Colonies were also significantly smaller in dived locations. The diving quotas produced a significant reduction of the ratio dead/total colonies in the dived locations soon after their adoption, but these benefits disappeared over the following years, possibly due to a gradual decline in operators' and divers' observance and concern, rather than an increasing number of dives. This suggests that the adoption of effective regulations is crucial for the environmental sustainability of diving tourism in protected areas and can provide positive effects, but an effort is needed to ensure that compliance is consistent over time, and that low-impact diving practices are adopted by this important recreational sector

    Worldwide variation of the COL14A1 gene is shaped by genetic drift rather than selective pressure

    Background: The aim of this study is to analyze the worldwide distribution of SNP rs4870723 in COL14A1 gene to check if there are significant genetic differences among different populations and to test if the gene is a trait under selection. Methods: Genomic DNA was extracted from 69 unrelated individuals from Sardinia and genotyped for SNP rs4870723. Data were compared with 26 different populations, clustered in 5 super-populations, from the public 1000 genomes database. Allele frequency and heterozygosity were calculated with Genepop. The Hardy–Weinberg equilibrium and pairwise population differentiation through analysis of molecular variance (AMOVA FST) were determined with Arlequin. Results: Allele frequencies of COL14A1 rs4870723 were compared in 27 populations clustered in 5 super-populations. All populations were in the Hardy–Weinberg equilibrium. In almost all populations, allele C was the most frequent allele, reaching the highest values in East Asia. The 27 populations showed an appreciable structure, with significant differences observed between European, African, and Asian populations. Conclusion: Significant differences were observed in the rs4870723 SNP distribution among the populations studied. However, we found no evidence for a selective pressure. Rather, the differentiation among the populations is likely the result of founder effect, genetic drift, and cultural factors, all events known to establish and maintain genetic diversity between populations

    Influence of the MCT1 rs1049434 on Indirect Muscle Disorders/Injuries in Elite Football Players

    The aim of this study was to investigate the association between MCT1 rs1049434 polymorphism and indirect muscle injuries in elite football players. One hundred and seventy-three male elite Italian football players (age = 19.2 ± 5.3 years) were recruited from a first-league football club participating at the Official National Italian Football Championship (Serie A, Primavera, Allievi, Giovanissimi). The cohort was genotyped for the MCT1 rs1049434 polymorphism, and muscle injuries data were collected during the period of 2009-2014 (five football seasons).Genomic DNA was extracted using a buccal swab, and genotyping was performed using PCR method. Structural-mechanical injuries and functional muscle disorder were included in the acute indirect muscle injury group.Participants with the MCT1 AA (AA = 1.57 ± 3.07, n = 69) genotype exhibit significantly higher injury incidents compared to participants with the TT genotype (TT = 0.09 ± 0.25, n = 22, P = 0.04).The MCT1 rs1049434 polymorphism is associated with the incidence of muscle injuries in elite football players. We anticipate that the knowledge of athletes' genetic predisposition to sports-related injuries might aid in individualizing training programs

    Local support for conservation is associated with perceptions of good governance, social impacts, and ecological effectiveness

    Local support is important for the longevity of conservation initiatives. The literature suggests that perceptions of ecological effectiveness, social impacts, and good governance will influence levels of local support for conservation. This paper examines these relationships using data from a survey of small-scale fishermen in 11 marine protected areas from six countries in the Mediterranean Sea. The survey queried small-scale fishermen regarding perceptions and support for conservation. We constructed composite scores for three categories of perceptions-ecological effectiveness, social impacts, and good governance-and tested the relationship with levels of support using ordinal regression models. While all three factors were positively correlated with support for conservation, perceptions of good governance and social impacts were stronger predictors of increasing support. These findings suggest that employing good governance processes and managing social impacts may be more important than ecological effectiveness for maintaining local support for conservation

    The physiopathological role of Wall Shear Stress (WSS) in intracranial aneurysm development/rupture and the action of contraction/expansion played by shear forces on vessel wall make topological skeleton analysis of the WSS vector field of great interest. Here we present a practical way to analyze WSS topological skeleton through the identification and classification of WSS fixed points and manifolds. The method is based on the calculation of the WSS vector field divergence and Poincarè index, and it is here successfully applied to a dataset computational hemodynamic models of intracranial aneurysms
