    A preliminary analysis of genome structure and composition in Gossypium hirsutum

    <p>Abstract</p> <p>Background</p> <p>Upland cotton has the highest yield, and accounts for > 95% of world cotton production. Decoding upland cotton genomes will undoubtedly provide the ultimate reference and resource for structural, functional, and evolutionary studies of the species. Here, we employed GeneTrek and BAC tagging information approaches to predict the general composition and structure of the allotetraploid cotton genome.</p> <p>Results</p> <p>142 BAC sequences from <it>Gossypium hirsutum </it>cv. Maxxa were downloaded <url>http://www.ncbi.nlm.nih.gov</url> and confirmed. These BAC sequence analysis revealed that the tetraploid cotton genome contains over 70,000 candidate genes with duplicated gene copies in homoeologous A- and D-subgenome regions. Gene distribution is uneven, with gene-rich and gene-free regions of the genome. Twenty-one percent of the 142 BACs lacked genes. BAC gene density ranged from 0 to 33.2 per 100 kb, whereas most gene islands contained only one gene with an average of 1.5 genes per island. Retro-elements were found to be a major component, first an enriched LTR/gypsy and second LTR/copia. Most LTR retrotransposons were truncated and in nested structures. In addition, 166 polymorphic loci amplified with SSRs developed from 70 BAC clones were tagged on our backbone genetic map. Seventy-five percent (125/166) of the polymorphic loci were tagged on the D-subgenome. By comprehensively analyzing the molecular size of amplified products among tetraploid <it>G. hirsutum </it>cv. Maxxa, acc. TM-1, and <it>G. barbadense </it>cv. Hai7124, and diploid <it>G. herbaceum </it>var. <it>africanum </it>and <it>G. raimondii</it>, 37 BACs, 12 from the A- and 25 from the D-subgenome, were further anchored to their corresponding subgenome chromosomes. After a large amount of genes sequence comparison from different subgenome BACs, the result showed that introns might have no contribution to different subgenome size in <it>Gossypium</it>.</p> <p>Conclusion</p> <p>This study provides us with the first glimpse of cotton genome complexity and serves as a foundation for tetraploid cotton whole genomesequencing in the future.</p

    Construction and experimental validation of a signature for predicting prognosis and immune infiltration analysis of glioma based on disulfidptosis-related lncRNAs

    BackgroundsDisulfidptosis, a newly discovered mechanism of programmed cell death, is believed to have a unique role in elucidating cancer progression and guiding cancer therapy strategies. However, no studies have yet explored this mechanism in glioma.MethodsWe downloaded data on glioma patients from online databases to address this gap. Subsequently, we identified disulfidptosis-related genes from published literature and verified the associated lncRNAs.ResultsThrough univariate, multivariate, and least absolute shrinkage and selection operator (LASSO) regression algorithms analyses, we identified 10 lncRNAs. These were then utilized to construct prognostic prediction models, culminating in a risk-scoring signature. Reliability and validity tests demonstrated that the model effectively discerns glioma patients’ prognosis outcomes. We also analyzed the relationship between the risk score and immune characteristics, and identified several drugs that may be effective for high-risk patients. In vitro experiments revealed that LINC02525 could enhances glioma cells’ migration and invasion capacities. Additionally, knocking down LINC02525 was observed to promote glioma cell disulfidptosis.ConclusionThis study delves into disulfidptosis-related lncRNAs in glioma, offering novel insights into glioma therapeutic strategies

    Ethyne Reducing Metal-Organic Frameworks to Control Fabrications of Core/shell Nanoparticles as Catalysts

    An approach using cobalt metal-organic frameworks (Co-MOF) as precursors is established for the fabrication of cobalt nanoparticles in porous carbon shells (core/shell Co@C). Chemical vapor deposition of ethyne is used for controlling the reduction of cobalt nanoclusters in the MOF and the spontaneous formation of the porous carbon shells. The metallic cobalt cores formed are up to 4 - 6 nm with the crystal phase varying between hexagonally-close-packed (hcp) and face-centre-packed (fcc). The porous carbon shells change from amorphous to graphene with the ethyne deposition temperature increasing from 400 to 600 oC. The core/shell Co@C nanoparticles exhibit high catalytic activity in selectively converting syngas (CTY: 254.1 - 312.1 μmolCO·gCo-1·s-1) into hydrocarbons (4.0 - 5.2 gHC·g-cat-1·h-1) at 260 oC. As well as the crystal size and phase, the coordination numbers of the cobalt to oxygen and to other cobalt atoms on the surface of the cobalt nanoparticles, and the permeability of the porous carbon shell have been related to the catalytic performance in FTS reactions

    Variation characteristics of stress distribution in the subchondral bone of the knee joint of judo athletes with long-term stress changes

    ObjectiveTo investigate the distribution of bone density in the subchondral bone tissue of the knee joint due to the mechanical stress load generated by judo, the bone tissue volume of different densities and the bone remodeling characteristics of the subchondral bone of the knee joint.MethodsCT imaging data of the knee joint were collected from 15 healthy individuals as controls and 15 elite judo athletes. Firstly, they were processed by the CTOAM technique, and secondly, the distribution pattern of high-density areas of the knee joint was localized using nine anatomical regions. In addition, three tomographic images were selected in the sagittal, coronal, and axial 2D image windows to observe the distribution of different densities of bone tissue. Finally, the percentage of bone tissue volume (%BTV) and bone remodeling trend of bone tissues with different densities were determined.ResultsIn this study, high-density areas were found in the 4th, 5th, and 6th regions of the articular surface of the distal femur and the 1st, 2nd, 3rd, 4th, 5th, 6th, 7th and 8th regions of the tibial plateau in judo athletes; the distribution of high-density areas on the articular surface of the distal femur in control subjects was similar with judo athletes, and high-density areas were mainly found in the 4th and 5th regions of the tibial plateau. The %BTV of low (401-500HU in the distal femur; 301-400 HU and 401-500HU in the tibial plateau), moderate, and high bone density was higher in judo athletes than in controls in the subchondral bone of the distal femur and tibial plateau (P&lt; 0.05).ConclusionThe history of compressive stresses, struck stresses, soft tissue tension and pull, self-gravity and intra-articular stress loading generated by the lower limb exercise technique of judo leads to specific forms of stress distribution and bone tissue remodeling in the subchondral bone tissue within the distal femur and tibia plateau

    Das Optical Neume Recognition Project (ONRP) hat die digitale Kodierung von musikalischen Notationszeichen aus dem Jahr um 1000 zum Ziel – ein ambitioniertes Vorhaben, das die Projektmitglieder veranlasste, verschiedenste methodische Ansätze zu evaluieren. Die Optical Music Recognition-Software soll eine linienlose Notation aus einem der ältesten erhaltenen Quellen mit Notationszeichen, dem Antiphonar Hartker aus der Benediktinerabtei St. Gallen (Schweiz), welches heute in zwei Bänden in der Stiftsbibliothek in St. Gallen aufbewahrt wird, erfassen. Aufgrund der handgeschriebenen, linienlosen Notation stellt dieser Gregorianische Gesang den Forscher vor viele Herausforderungen. Das Werk umfasst über 300 verschiedene Neumenzeichen und ihre Notation, die mit Hilfe der Music Encoding Initiative (MEI) erfasst und beschrieben werden sollen. Der folgende Artikel beschreibt den Prozess der Adaptierung, um die MEI auf die Notation von Neumen ohne Notenlinien anzuwenden. Beschrieben werden Eigenschaften der Neumennotation, um zu verdeutlichen, wo die Herausforderungen dieser Arbeit liegen sowie die Funktionsweise des Classifiers, einer Art digitalen Neumenwörterbuchs

    Application of Computer Technology in Optimal Design of Overall Structure of Special Machinery

    With the transformation and upgrading of my country’s industrial structure, the level of manufacturing automation has gradually improved. According to research, the design of mechanical products is mostly completed by improvement or innovation on the basis of existing design knowledge. Knowledge reuse is a technique to ensure the maximization of design resource utilization by reusing design knowledge. This article applies knowledge reuse technology to the development and design of mechanical products. By integrating the technical logic of the functional analysis system with the development of quality functions, the transformation of customer demand information and product function design is realized, and the task of the product design plan analysis phase is completed. This paper uses the finite element analysis software ANSYS to explore a new nonlinear finite element modeling method and conducts simulation experiments. At the same time, this paper improves the genetic algorithm, which effectively improves the optimization efficiency and completes the parameter optimization under multiobjective and multistructure conditions. From the experimental results, it takes 328.64 seconds for the basic genetic algorithm to search for the optimal solution of the complex problem. The improved time is shortened to 86.31 seconds, and the efficiency is increased by 73.74%. This shows that the improved genetic algorithm has better robustness and can find the optimal solution in a shorter calculation time. For complex problems such as the optimization of the overall structure of special machinery, the improved genetic algorithm obviously helps to improve the optimization efficiency and improves the effectiveness and pertinence of product design schemes
