Engineering education towards sustainability
The traditional systems have been studied in an isolated mode, seek to respond to a specific need and deal with a restricted set of variables. The targets established for energy efficient systems and energy sustainability imply that new systems are more comprehensive and combine the various individual systems. This new topology requires a new kind of engineering professionals provided with new skills. New engineering professionals need to have not only a set of deep capabilities in a specific area, but also more comprehensive proficiencies that allow them to understand how to integrate their particular system into a wider functional system.

Sustainability evaluation of a Portuguese “terroir” wine
The challenges of sustainability are transversal to all human activities, and the wine sector has its own role to play in the march for a more sustainable development. The proper definition of the most adequate measures and/or policies must be based on an objective and quantitative evaluation of the sustainability of a product or process. In this work the sustainability of a “terroir” wine is assessed taking into account its life cycle and using the following indicators: carbon and water footprint, material intensity, solid waste generated, worker turnover rate, investment in H&S training and EBITDA. All indicators are expressed per functional unit of 0.75 L of wine. The evaluation used data from the company complemented with data/information from the literature or life cycle inventory databases. To account for climatic variability, data from three consecutive years was used. Average values of 3.51 kgCO2eq and 481.4 L per functional unit were obtained for the carbon and water footprint respectively, both values within the range of values reported in the literature.To Sogrape Vinhos, S.A. for funding Project PP-IJUP2014-SOGRAPE. Chromosomal disorders and male infertility
Male factor infertility is considered a complex disorder with a largely unknown etiology that affects about 7% of men. In general, genetic abnormalities account for 15%-30% of condition and Y chromosome microdeletions are also frequent.
The study, based on our casuistic, aimed at contributing to a better understanding of the genetic causes of infertility.
A group of 410 idiopathic infertile men with non-obstructive azoospermia, oligozoospermia, or unknown semen quality (based on clinical evaluation and/or sperm counts) was retrospectively selected. Conventional karyotype was performed in all samples; Y microdeletion screen was performed in 247 samples.
Forty two abnormal karyotypes (10.2%) were found, indicating an elevated frequency of chromosome abnormalities among the selected infertile men, as compared to that of newborn populations (≈0.4%). This frequency is higher than that reported in most similar studies that pointed to frequencies ranging from 2.2%-14.3%.
Klinefelter´s syndrome was the most common chromosome disorder (4.9%). There were 18 cases with 47,XXY karyotype and 2 cases of mosaicism involving lines 47,XXY and 46,XY. Reciprocal translocations were identified in 10 cases (2.4%), particularly in men with unknown semen quality. Overall, reciprocal translocations have been found in approximately 1% of the infertile men and more commonly in azoospermics than in oligozoospermics. However, this type of association was not found in the present study.
On the other hand, Y microdeletions were identified in 16/247 cases (6.5%), more frequently in azoospermics (13.3%, corresponding to 8/60 azoospermics). Among these 8 cases, 7 presented deletions at the AZFc region.
The marked presence of chromosomal abnormalities and Y microdeletions enphasizes the relevance of studying both factors in infertile men to improve genetic counseling, to allow the development of appropriate therapies, and to expand the knowledge about the ethiology of male infertility
Control of bovine brucellosis from persistently infected holdings using RB51 vaccination with test-and-slaughter : a comparative case report from a high incidence area in Portugal
Bovine brucellosis due to Brucella abortus infection causes significant reproduc-tive and production losses in cattle and is a
major zoonosis. Eradication of this disease has proved difficult to achieve in Portugal where it still occurs in some regions
despite an ongoing national eradication programme. In 2004, the Alent-ejo region, a major cattle producing area, reported
one of the highest levels of bovine brucellosis in the country, especially in one divisional area. In that area, bovine brucellosis
was particularly problematic in a holding of ten herds, the largest extensive cattle unit in the country, which remained infected
despite an extensive test-and-slaughter programme and depopulation of five herds. A 5-year programme of RB51 vaccination
with biannual test-and-slaughter was thus implemented in 2004. The apparent animal seroprevalence decreased from 19%
(646/3,400) to 3% (88/2930) on the third herd-level test and remained below 0.8% (27/3324) after the fourth test. After the
tenth test, the holding had a prevalence of 0.1% (2/2332) and only one herd remained positive with a within-herd prevalence
of 1.1% (2/177). The results were compared to all other herds (n = 10) in the divisional area that were also persistently
infected but were subject only to test-and-slaughter before being depopulated. In these herds, the strategy of test-andslaughter
did not reduce the prevalence, which remained significantly higher than the vaccinated group (median = 0.48% and
8.5% in vaccinated versus non-vaccinated herds; Wilcoxon rank sum test; P < 0.01). The success of this pilot programme in
continental Portugal pro-vided a valuable case study to the official veterinary services by illustrating the value of RB51
vaccination with parallel testing and improved biosecurity as a comprehensive and sustainable strategy for bovine brucellosis

Age-associated distribution of normal B-cell and plasma cell subsets in peripheral blood
Background: Humoral immunocompetence develops stepwise throughout life and contributes to individual susceptibility to infection, immunodeficiency, autoimmunity, and neoplasia. Immunoglobulin heavy chain (IgH) isotype serum levels can partly explain such age-related differences, but their relationship with the IgH isotype distribution within memory B-cell (MBC) and plasma cell (PCs) compartments remains to be investigated. Objective: We studied the age-related distribution of MBCs and PCs expressing different IgH isotypes in addition to the immature/transitional and naive B-cell compartments. Methods: B-cell and PC subsets and plasma IgH isotype levels were studied in cord blood (n = 19) and peripheral blood (n = 215) from healthy donors aged 0 to 90 years by using flow cytometry and nephelometry, respectively. Results: IgH-switched MBCs expressing IgG1, IgG2, IgG3, IgA1, and IgA2 were already detected in cord blood and newborns at very low counts, whereas CD27+IgM++IgD+ MBCs only became detectable at 1 to 5 months and remained stable until 2 to 4 years, and IgD MBCs peaked at 2 to 4 years, with both populations decreasing thereafter. MBCs expressing IgH isotypes of the second immunoglobulin heavy chain constant region (IGHC) gene block (IgG1, IgG3, and IgA1) peaked later during childhood (2-4 years), whereas MBCs expressing third IGHC gene block immunoglobulin isotypes (IgG2, IgG4, and IgA2) reached maximum values during adulthood. PCs were already detected in newborns, increasing in number until 6 to 11 months for IgM, IgG1, IgG2, IgG3, IgA1, and IgA2; until 2 to 4 years for IgD; and until 5 to 9 years for IgG4 and decreasing thereafter. For most IgH isotypes (except IgD and IgG4), maximum plasma levels were reached after PC and MBC counts peaked. Conclusions: PC counts reach maximum values early in life, followed by MBC counts and plasma IgH isotypes. Importantly, IgH isotypes from different IGHC gene blocks show different patterns, probably reflecting consecutive cycles of IgH isotype switch recombination through life
A Model for the Development of the Rhizobial and Arbuscular Mycorrhizal Symbioses in Legumes and Its Use to Understand the Roles of Ethylene in the Establishment of these two Symbioses
We propose a model depicting the development of nodulation and arbuscular mycorrhizae. Both processes are dissected into many steps, using Pisum sativum L. nodulation mutants as a guideline. For nodulation, we distinguish two main developmental programs, one epidermal and one cortical. Whereas Nod factors alone affect the cortical program, bacteria are required to trigger the epidermal events. We propose that the two programs of the rhizobial symbiosis evolved separately and that, over time, they came to function together. The distinction between these two programs does not exist for arbuscular mycorrhizae development despite events occurring in both root tissues. Mutations that affect both symbioses are restricted to the epidermal program. We propose here sites of action and potential roles for ethylene during the formation of the two symbioses with a specific hypothesis for nodule organogenesis. Assuming the epidermis does not make ethylene, the microsymbionts probably first encounter a regulatory level of ethylene at the epidermis–outermost cortical cell layer interface. Depending on the hormone concentrations there, infection will either progress or be blocked. In the former case, ethylene affects the cortex cytoskeleton, allowing reorganization that facilitates infection; in the latter case, ethylene acts on several enzymes that interfere with infection thread growth, causing it to abort. Throughout this review, the difficulty of generalizing the roles of ethylene is emphasized and numerous examples are given to demonstrate the diversity that exists in plants
Balanço do nitrogênio e fósforo em solo com cultivo orgânico de hortaliças após a incorporação de biomassa de guandu.
Os objetivos deste trabalho foram avaliar os efeitos de faixas de guandu (Cajanus cajan) e da incorporação da biomassa proveniente de sua poda na fertilidade do solo e na produtividade de três hortaliças sob cultivo orgânico. O delineamento usado foi de blocos casualizados completos em esquema de parcelas subsubdivididas com três repetições. As produtividades de beterraba, cenoura e feijão-de-vagem não foram afetadas pelos tratamentos. Nas parcelas onde não houve incorporação da biomassa de guandu, o balanço de nitrogênio no sistema foi negativo, ao passo que com a incorporação, esse balanço foi positivo. Embora tenha ocorrido balanço positivo para o fósforo nas parcelas sem a incorporação de biomassa de guandu, houve um aumento significativo na absorção desse elemento pelas hortaliças quando o material foi incorporado. O sistema de cultivo em aléias de guandu pode representar uma prática vantajosa para os produtores orgânicos, por contribuir na manutenção da fertilidade do solo
