    An Examination of Pharmacy Faculty Quality of Work Life: Work Satisfaction, Turnover Intentions, and Self-efficacy

    Previous research on pharmacy faculty quality of work life lacks a thorough, well-conceived theoretical foundation, especially in critical areas such as work satisfaction and turnover intentions. The objectives of this study are to develop a comprehensive measure of pharmacy academician work satisfaction and determine the contribution of work life variables toward pharmacy academician work satisfaction, job turnover intentions, and self-efficacies. An e-mail survey sent to pharmacy faculty elicited responses on various single and multi-item measures of quality of work life variables. Factor analytic procedures were used to evaluate the validity and reliability of the work satisfaction scale. Multiple regression procedures were used to identify predictors of the satisfaction, turnover, and self-efficacy constructs. The satisfaction measure demonstrated construct validity, while each of the six domain subscales exhibited internal consistency reliability. Institutional support and intradisciplinary consensus variables were commonly identified as predictors of satisfaction, stress, and self-efficacies for pharmacy faculty. Administrators might consider these results when implementing policies that may impact organizational climate and faculty morale. There is still an opportunity to further examine quality of work life among pharmacy faculty

    Spatially distributed water-balance and meteorological data from the Wolverton catchment, Sequoia National Park, California

    Accurate water-balance measurements in the seasonal, snow-dominated Sierra Nevada are important for forest and downstream water management. However, few sites in the southern Sierra offer detailed records of the spatial and temporal patterns of snowpack and soil-water storage and the fluxes affecting them, i.e., precipitation as rain and snow, snowmelt, evapotranspiration, and runoff. To explore these stores and fluxes we instrumented the Wolverton basin (2180-2750 m) in Sequoia National Park with distributed, continuous sensors. This 2006-2016 record of snow depth, soil moisture and soil temperature, and meteorological data quantifies the hydrologic inputs and storage in a mostly undeveloped catchment. Clustered sensors record lateral differences with regards to aspect and canopy cover at approximately 2250 and 2625 m in elevation, where two meteorological stations are installed. Meteorological stations record air temperature, relative humidity, radiation, precipitation, wind speed and direction, and snow depth.     Enhanced Integrin α4β1-Mediated Adhesion Contributes to a Mobilization Defect of Endothelial Progenitor Cells in Diabetes.

    Diabetes is associated with a deficit of circulating endothelial progenitor cells (EPCs), which has been attributed to their defective mobilization from the bone marrow. The basis for this mobilization defect is not completely understood, and we sought to determine if hyperglycemic conditions enhanced EPC adhesion. We found that culturing EPCs in high glucose media increased adhesion to bone marrow stromal cells. This enhanced adhesion was associated with decreased expression of protein kinase A regulatory subunit 1β (PRKAR1β), activation of protein kinase A (PKA), and phosphorylation of α4-integrin on serine 988. This potentiated adhesion was reversed by treatment with a PKA inhibitor, overexpression of PRKAR1β, or expression of a phosphorylation-defective α4-integrin variant (α4[S988A]). Using a model of type 1 diabetes, we showed that α4(S988A)-expressing mice have more circulating EPCs than their wild-type counterparts. Moreover, diabetic α4(S988A) mice demonstrate enhanced revascularization after hind limb ischemia. Thus, we have identified a novel signaling mechanism activating PKA in diabetes (downregulation of an inhibitory regulatory subunit) that leads to deficits of circulating EPCs and impaired vascular repair, which could be reversed by α4-integrin mutation

    Development of a single-session, transdiagnostic preventive intervention for young adults at risk for emotional disorders

    Cognitive-behavioral prevention programs have demonstrated efficacy in reducing subclinical symptoms of anxiety and depression, and there is some evidence to suggest that they can lower the risk of future disorder onset. However, existing interventions tend to be relatively lengthy and target specific disorders or problem areas, both of which limit their potential for widespread dissemination. To address these limitations, we aimed to develop a single-session, transdiagnostic preventive intervention based on the Unified Protocol for Transdiagnostic Treatment of Emotional Disorders for young adults at risk for developing anxiety and/or depressive disorders within a college setting. Results from this proof-of-concept study indicated that the intervention was viewed as highly satisfactory and acceptable. The intervention also was successful at delivering adaptive emotion management skills in its 2-hr workshop format. Future studies evaluating the efficacy of this novel transdiagnostic, emotion-focused prevention program are warranted.Accepted manuscrip

    Atmosphere-snow transfer function for H2O2: microphysical considerations

    H2O2 analyses of polar ice cores show an increase in concentration from 200 years to the present. In order to quantitatively relate the observed trend in the ice to atmospheric levels, the atmosphere-snow transfer behavior and postdepositional changes must be known. Atmosphere-snow transfer was studied by investigating uptake and release of H2O2 in a series of laboratory column experiments in the temperature range −3°C to −45°C. Experiments consisted of passing H2O2-containing air through a column packed with 200-μm diameter ice spheres and measuring the change in gas phase H2O2 concentration with time. The uptake of H2O2 was a slow process requiring several hours to reach equilibrium. Uptake involved incorporation of H2O2 into the bulk ice as well as surface accumulation. The amount of H2O2 taken up by the ice was greater at the lower temperatures. The sticking coefficient for H2O2 on ice in the same experiments was estimated to be of the order of 0.02 to 0.5. Release of H2O2 from the ice occurred upon passing H2O2-free air through the packed columns, with the time scale for degassing similar to that for uptake. These results suggest that systematic losses of H2O2 from polar snow could occur under similar conditions, when atmospheric concentrations of H2O2 are low, that is, in the winter

    Engineered G protein coupled receptors reveal independent regulation of internalization, desensitization and acute signaling

    BACKGROUND: The physiological regulation of G protein-coupled receptors, through desensitization and internalization, modulates the length of the receptor signal and may influence the development of tolerance and dependence in response to chronic drug treatment. To explore the importance of receptor regulation, we engineered a series of G(i)-coupled receptors that differ in signal length, degree of agonist-induced internalization, and ability to induce adenylyl cyclase superactivation. All of these receptors, based on the kappa opioid receptor, were modified to be receptors activated solely by synthetic ligands (RASSLs). This modification allows us to compare receptors that have the same ligands and effectors, but differ only in desensitization and internalization. RESULTS: Removal of phosphorylation sites in the C-terminus of the RASSL resulted in a mutant that was resistant to internalization and less prone to desensitization. Replacement of the C-terminus of the RASSL with the corresponding portion of the mu opioid receptor eliminated the induction of AC superactivation, without disrupting agonist-induced desensitization or internalization. Surprisingly, removal of phosphorylation sites from this chimera resulted in a receptor that is constitutively internalized, even in the absence of agonist. However, the receptor still signals and desensitizes in response to agonist, indicating normal G-protein coupling and partial membrane expression. CONCLUSIONS: These studies reveal that internalization, desensitization and adenylyl cyclase superactivation, all processes that decrease chronic G(i)-receptor signals, are independently regulated. Furthermore, specific mutations can radically alter superactivation or internalization without affecting the efficacy of acute G(i )signaling. These mutant RASSLs will be useful for further elucidating the temporal dynamics of the signaling of G protein-coupled receptors in vitro and in vivo

    Age related diffusion and tractography changes in typically developing pediatric cervical and thoracic spinal cord

    Background and objective: Diffusion tensor imaging (DTI) and diffusion tensor tractography (DTT) are two techniques that can measure white matter integrity of the spinal cord. Recently, DTI indices have been shown to change with age. The purpose of this study is (a) to evaluate the maturational states of the entire pediatric spinal cord using DTI and DTT indices including fractional anisotropy (FA), mean diffusivity (MD), mean length of white matter fiber tracts and tract density and (b) to analyze the DTI and DTT parameters along the entire spinal cord as a function of spinal cord levels and age. Method: A total of 23 typically developing (TD) pediatric subjects ranging in age from 6 to 16 years old (11.94 ± 3.26 (mean ± standard deviation), 13 females and 10 males) were recruited, and scanned using 3.0 T MR scanner. Reduced FOV diffusion tensor images were acquired axially in the same anatomical location prescribed for the T2-weighted images to cover the entire spinal cord (C1-mid L1 levels). To mitigate motion induced artifacts, diffusion directional images were aligned with the reference image (b0) using a rigid body registration algorithm performed by in-house software developed in Matlab (MathWorks, Natick, Massachusetts). Diffusion tensor maps (FA and MD) and streamline deterministic tractography were then generated from the motion corrected DTI dataset. DTI and DTT parameters were calculated by using ROIs drawn to encapsulate the whole cord along the entire spinal cord by an independent board certified neuroradiologist. These indices then were compared between two age groups (age group A = 6–11 years (n = 11) and age group B = 12–16 years (n = 12)) based on similar standards and age definitions used for reporting spinal cord injury in the pediatric population. Standard least squared linear regression based on a restricted maximum likelihood (REML) method was used to evaluate the relationship between age and DTI and DTT parameters. Results: An increase in FA (group A = 0.42 ± 0.097, group B = 0.49 ± 0.116), white matter tract density (group A = 368.01 ± 236.88, group B = 440.13 ± 245.24) and mean length of fiber tracts (group A = 48.16 ± 20.48 mm, group B = 60.28 ± 23.87 mm) and a decrease in MD (group A = 1.06 ± 0.23 × 10−3 mm2/s, group B = 0.82 ± 0.24 × 10−3 mm2/s) were observed with age along the entire spinal cord. Statistically significant increases have been shown in FA (p = 0.004, R2 = 0.57), tract density (p = 0.0004, R2 = 0.58), mean length of fiber tracts (p \u3c 0.001, R2 = 0.5) and a significant decrease has been shown in MD (p = 0.002, R2 = 0.59) between group A and group B. Also, it has been shown DTI and DTT parameters vary along the spinal cord as a function of intervertebral disk and mid-vertebral body level. Conclusion: This study provides an initial understanding of age related changes of DTI values as well as DTT metrics of the spinal cord. The results show significant differences in DTI and DTT parameters which may result from decreasing water content, myelination of fiber tracts, and the thickening diameter of fiber tracts during the maturation process. Consequently, when quantitative DTI and DTT of the spinal cord is undertaken in the pediatric population an age and level matched normative dataset should be used to accurately interpret the quantitative results. © 201
