    Regulation of dendritic spine length in corticopontine layer V pyramidal neurons by autism risk gene β3 integrin

    The relationship between autism spectrum disorder (ASD) and dendritic spine abnormalities is well known, but it is unclear whether the deficits relate to specific neuron types and brain regions most relevant to ASD. Recent genetic studies have identified a convergence of ASD risk genes in deep layer pyramidal neurons of the prefrontal cortex. Here, we use retrograde recombinant adeno-associated viruses to label specifically two major layer V pyramidal neuron types of the medial prefrontal cortex: the commissural neurons, which put the two cerebral hemispheres in direct communication, and the corticopontine neurons, which transmit information outside the cortex. We compare the basal dendritic spines on commissural and corticopontine neurons in WT and KO mice for the ASD risk gene Itgb3, which encodes for the cell adhesion molecule beta 3 integrin selectively enriched in layer V pyramidal neurons. Regardless of the genotype, corticopontine neurons had a higher ratio of stubby to mushroom spines than commissural neurons. beta 3 integrin affected selectively spine length in corticopontine neurons. Ablation of beta 3 integrin resulted in corticopontine neurons lacking long (> 2 mu m) thin dendritic spines. These findings suggest that a deficiency in beta 3 integrin expression compromises specifically immature spines on corticopontine neurons, thereby reducing the cortical territory they can sample. Because corticopontine neurons receive extensive local and long-range excitatory inputs before relaying information outside the cortex, specific alterations in dendritic spines of corticopontine neurons may compromise the computational output of the full cortex, thereby contributing to ASD pathophysiology

    We present a combined theoretical and experimental study of the anisotropy in the optical absorption of V-shaped quantum wires. By means of realistic band structure calculations for these structures, we show that detailed information on the heavy- and light-hole states can be singled out from the anisotropy spectra {\em independently of the electron confinement}, thus allowing accurate valence band spectroscopy.Comment: To be published in Appl.     Polydimethylsiloxane-LiNbO3 surface acoustic wave micropump devices for fluid control into microchannels.

    This paper presents prototypical microfluidic devices made by hybrid microchannels based on piezoelectric LiNbO3 and polydimethylsiloxane. This system enables withdrawing micropumping by acoustic radiation in microchannels. The withdrawing configuration, integrated on chip, is here quantitatively investigated for the first time, and found to be related to the formation and coalescence dynamics of droplets within the microchannel, primed by surface acoustic waves. The growth dynamics of droplets is governed by the water diffusion on LiNbO3, determining the advancement of the fluid front. Observed velocities are up to 2.6 mm s−1 for 30 dBm signals applied to the interdigital transducer, corresponding to tens of nl s−1, and the micropumping dynamics is described by a model taking into account an acoustic power exponentially decaying upon travelling along the microchannel. This straighforward and flexible micropumping approach is particularly promising for the withdrawing of liquids in lab-on-chip devices performing cycling transport of fluids and biochemical reactions

    Age estimation in children by measurement of open apices in teeth: a European formula

    The aim of the present paper was to improve and expand research with a larger number of children from various European countries and to provide a common formula useful for all these countries. Orthopantomographs taken from 2,652 European Caucasian children (1,382 boys, 1,270 girls) aged between 4 and 16 years were analyzed. The children came from Croatia, Germany, Kosovo, Italy, Slovenia, Spain, and the UK. Following the pilot study, subjects’ age was modeled as a function of gender (g), morphological variables (predictors)×5 (second premolar), s (sum of normalized open apices) N0, and the first-order interaction between s and N0. The results showed that all these variables contributed significantly to the fit, so that all were included in the regression model, yielding the following linear regression formula: Age=8.387+ 0.282 g−1.692×5+0.835 N0−0.116 s−0.139 s×N0, where g is a variable, 1 for males and 0 for females. The equation explained 86.1% (R2=0.861) of total deviance. The median of the residuals (=observed age minus predicted age) was −0.114 years, with (RefB.2) interquartile range=1.22 years

    Intracellular trafficking of AMPA receptors is a tightly regulated process which involves several adaptor proteins, and is crucial for the activity of excitatory synapses both in basal conditions and during synaptic plasticity. We found that, in rat hippocampal neurons, an intracellular pool of the tetraspanin TSPAN5 promotes exocytosis of AMPA receptors without affecting their internalisation. TSPAN5 mediates this function by interacting with the adaptor protein complex AP4 and Stargazin and possibly using recycling endosomes as a delivery route. This work highlights TSPAN5 as a new adaptor regulating AMPA receptor trafficking

    Provenance of Ordovician clastic sequences of the San Rafael Block (Central Argentina), with emphasis on the Ponón Trehué Formation

    The Ordovician Ponón Trehué Formation is the only early Palaeozoic sedimentary sequence known to record a primary contact with the Grenvillian-age basement of the Argentinean Cuyania terrane, in its southwards extension named the San Rafael block. Petrographic and geochemical data indicate contributions from a dominantly upper continental crustal component and a subordinated depleted component. Nd isotopes indicate εNd of −4.6, ƒSm/Nd −0.36 and TDM 1.47 Ga in average. Pb-isotope ratios display average values for 206Pb/204Pb, 207Pb/204Pb, and 208Pb/204Pb of 19.15, 15.69 and 38.94 respectively. U–Pb detrital zircon ages from the Ponón Trehué Formation cluster around values of 1.2 Ga, indicating a main derivation from a local basement source (Cerro La Ventana Formation). The Upper Ordovician Pavón Formation records a younger episode of clastic sedimentation within the San Rafael block, and it shows a more complex detrital zircon age population (peaks at 1.1 and 1.4 Ga as well as Palaeoproterozoic and Neoproterozoic detrital grains). Detailed comparison between the two Ordovician clastic units indicates a shift with time in provenance from localized basement to more regional sources. Middle to early Upper Ordovician age is inferred for accretion of the Cuyania terrane to the proto-Andean margin of Gondwana.Centro de Investigaciones Geológica

    Status of charcoal canker on oak trees at a site of community importance: case study of the relict castelfidardo forest (Sic area it520008, castelfidardo, an, Italy)

    Oaks are dominant and key tree species in Mediterranean forest ecosystems. However, in recent decades, oak forests have been heavily impacted by oak decline, a worldwide phenomenon exacerbated by climate change. The charcoal disease agent Biscogniauxia mediterranea is involved in the decline of Mediterranean oak formations in a variety of contexts. Here, we investigated the impact and role of B. mediterranea in the decline of oaks in Castelfidardo Forest, a relict wood of the late Holocene and a Site of Community Importance. We established five plots within which we recorded tree positions, any symptoms and signs of decline, association of B. mediterranea to declining trees, and deadwood and associated mycota. Of 471 oaks inspected, 7.0% showed brownish exudates on the stems, 46.9% showed epicormic shoots along the main trunk, and 24.4% showed black carbonaceous stromata on diseased branches and trunks. The decline was most severe for Quercus cerris, which comprised plots #4 and #5, at 50.0% (81/162 trees) and 29.0% (33/114), respectively; then for Quercus robur for plot #3, at 40.0% (38/95); and finally for Quercus pubescens for plots #1 and #2, at 13.7% (7/51) and 12.3% (6/49), respectively. Bark tissues were collected from trees with charcoal cankers and taken to the laboratory for microscopic examination and identification by mycological and molecular methods. This investigation revealed a close association between oaks with pronounced reduction of vitality and incidence of B. mediterranea. Deadwood was equally distributed among the five plots, and was heavily colonized by Basidiomycota. The high incidence of the charcoal canker pathogen B. mediterranea appeared to be related to environmental stresses. However, the absence of silvicultural management, high competition among physiologically mature trees, and the geographic isolation of this residual forest may have predisposed oaks to decline

    Electrically conductive and high temperature resistant superhydrophobic composite films from colloidal graphite

    Electrically conductive and self-cleaning superhydrophobic films (water contact angles >160°, droplet roll off angles <5°) were fabricated by simply solution casting sub-micron polytetrafluoroethylene (Teflon) particle dispersed alcohol-based colloidal graphite solutions. The process is very suitable for forming conductive superhydrophobic coatings on glasses, metals, ceramics and high performance polymers such as polyimide (Kapton®). The solutions were deposited on microscope glass slides and Kapton® films by drop casting. After solvent evaporation under ambient conditions, the coatings were annealed to melt Teflon. Upon melting, Teflon particles fused into one another forming a hydrophobic polymer matrix. The degree of superhydrophobicity and the surface morphology of the coatings together with their electrical conductivity were studied in detail by varying Teflon-to-graphite weight fractions. A number of applications can be envisioned for these coatings such as electrode materials for energy conversion devices, high performance electromagnetic shielding materials, flexible electronic components and heat exchanger surfaces, to name a few

    The Italian document: decisions for intensive care when there is an imbalance between care needs and resources during the COVID-19 pandemic

    Background: In early 2020, the Italian Society of Anesthesia Analgesia Resuscitation and Intensive Care (SIAARTI) published clinical ethics recommendations for the allocation of intensive care during COVID-19 pandemic emergency. Later the Italian National Institute of Health (ISS) invited SIAARTI and the Italian Society of Legal and Insurance Medicine to prepare a draft document for the definition of triage criteria for intensive care during the emergency, to be implemented in case of complete saturation of care resources. Methods: Following formal methods, including two Delphi rounds, a multidisciplinary group with expertise in intensive care, legal medicine and law developed 12 statements addressing: (1) principles and responsibilities; (2) triage; (3) previously expressed wishes; (4) reassessment and shifting to palliative care; (5) collegiality and transparency of decisions. The draft of the statements, with their explanatory comments, underwent a public consultation opened to Italian scientific or technical-professional societies and other stakeholders (i.e., associations of citizens, patients and caregivers; religious communities; industry; public institutions; universities and research institutes). Individual healthcare providers, lay people, or other associations could address their comments by e-mail. Results: Eight stakeholders (including scientific societies, ethics organizations, and a religious community), and 8 individuals (including medical experts, ethicists and an association) participated to the public consultation. The stakeholders’ agreement with statements was on average very high (ranging from 4.1 to 4.9, on a scale from 1—full disagreement to 5—full agreement). The 4 statements concerning triage stated that in case of saturation of care resources, the intensive care triage had to be oriented to ensuring life-sustaining treatments to as many patients as possible who could benefit from them. The decision should follow full assessment of each patient, taking into account comorbidities, previous functional status and frailty, current clinical condition, likely impact of intensive treatment, and the patient's wishes. Age should be considered as part of the global assessment of the patient. Conclusions: Lacking national guidelines, the document is the reference standard for healthcare professionals in case of imbalance between care needs and available resources during a COVID-19 pandemic in Italy, and a point of reference for the medico-legal assessment in cases of dispute
