    A Metagenomic Approach to Characterization of the Vaginal Microbiome Signature in Pregnancy

    While current major national research efforts (i.e., the NIH Human Microbiome Project) will enable comprehensive metagenomic characterization of the adult human microbiota, how and when these diverse microbial communities take up residence in the host and during reproductive life are unexplored at a population level. Because microbial abundance and diversity might differ in pregnancy, we sought to generate comparative metagenomic signatures across gestational age strata. DNA was isolated from the vagina (introitus, posterior fornix, midvagina) and the V5V3 region of bacterial 16S rRNA genes were sequenced (454FLX Titanium platform). Sixty-eight samples from 24 healthy gravidae (18 to 40 confirmed weeks) were compared with 301 non-pregnant controls (60 subjects). Generated sequence data were quality filtered, taxonomically binned, normalized, and organized by phylogeny and into operational taxonomic units (OTU); principal coordinates analysis (PCoA) of the resultant beta diversity measures were used for visualization and analysis in association with sample clinical metadata. Altogether, 1.4 gigabytes of data containing >2.5 million reads (averaging 6,837 sequences/sample of 493 nt in length) were generated for computational analyses. Although gravidae were not excluded by virtue of a posterior fornix pH >4.5 at the time of screening, unique vaginal microbiome signature encompassing several specific OTUs and higher-level clades was nevertheless observed and confirmed using a combination of phylogenetic, non-phylogenetic, supervised, and unsupervised approaches. Both overall diversity and richness were reduced in pregnancy, with dominance of Lactobacillus species (L. iners crispatus, jensenii and johnsonii, and the orders Lactobacillales (and Lactobacillaceae family), Clostridiales, Bacteroidales, and Actinomycetales. This intergroup comparison using rigorous standardized sampling protocols and analytical methodologies provides robust initial evidence that the vaginal microbial 16S rRNA gene catalogue uniquely differs in pregnancy, with variance of taxa across vaginal subsite and gestational age

    Evaluation of alternative preservation treatments (water heat treatment, ultrasounds, thermosonication and UV-C radiation) to improve safety and quality of whole tomato

    Previously optimised postharvest treatments were compared to conventional chlorinated water treatment in terms of their effects on the overall quality of tomato (‘Zinac’) during storage at 10 °C. The treatments in question were water heat treatment (WHT = 40 °C, 30 min), ultrasounds (US = 45 kHz, 80 %, 30 min), thermosonication (TS =40 °C, 30 min, 45 kHz, 80 %) and ultraviolet irradiation (UV-C: 0.97 kJ m−2). The quality factors evaluated were colour, texture, sensorial analysis, mass loss, antioxidant capacity, total phenolic content, peroxidase and pectin methylesterase enzymatic activities, and microbial load reduction. The results demonstrate that all treatments tested preserve tomato quality to some extent during storage at 10 °C. WHT, TS and UV-C proved to be more efficient on minimising colour and texture changes with the additional advantage of microbial load reduction, leading to a shelf life extension when compared to control trials. However, at the end of storage, with exception of WHT samples, the antioxidant activity and phenolic content of treated samples was lower than for control samples. Moreover, sensorial results were well correlated with instrumental colour experimental data.     Sucrose Supply Can Increase Longevity of Broccoli (Brassica-Oleracea) Branchlets Kept At 22-Degrees-C

    Sucrose was supplied several hours after harvest to broccoli branchlets via the transpiration stream in order to increase the amount of sucrose available for respiration and to determine its influence on longevity at 22 degrees C. Calculations based on solution uptake indicated that an 8% (w/v) sucrose solution supplied sufficient substrate for respiration, but the pattern of respiratory decline after harvest was not altered by supply of exogenous sucrose, and yellowing of floret sepals began after 2 days. However, when sucrose was supplied immediately after harvest, yellowing was delayed. Treatment with cytokinin (50 ppm 6-benzylaminopurine), to delay yellowing, had no effect on levels of sucrose in branchlets after 4.5 days, but retarded loss of chlorophyll. Floret tissues appear to sense the decline in sucrose after harvest, the result being induction of senescence as judged by yellowing. 6-benzylaminopurine may block the sensing mechanism

    The growth and productivity of ‘Festival’ strawberry plants growing in a subtropical environment

    Experiments were conducted over 5 years to understand the seasonal phenology of bare-rooted ?Festival? strawberry plants (Fragaria ?ananassa) growing at Nambour in southeastern Queensland, Australia. Yields ranged from 661 to 966 g/plant, and average seasonal fruit fresh weight ranged from 15 to 18 g. The growth of the leaves, crowns, roots, flowers and fruit over time followed a linear or sigmoid pattern. Maximum values of leaf, crown and root dry weight towards the end of the growing season about 190 days after planting were 30, 15 and 7 g/plant, respectively. The rates of leaf and crown growth were lower than those achieved in California under a Mediterranean climate. There were strong relationships between the allocation of dry matter to the leaves, crowns and roots and plant dry weight. Allocation to the leaves, and especially to the crowns and roots, declined as the plants grew. The number of fruit/plant increased initially over time with a decline later in the season. Average fruit fresh weight was generally higher early in the season and then declined as fruit production increased. There were strong relationships between the growth of the whole plant and the growth of the flowers and immature fruit, and leaf expansion, across the growing season and across the 5 different years. These results indicate that seasonal growth and potential productivity were strongly linked to the expansion of the leaves in this environment
