    Financing the (Environmental) quality of cities with energy efficiency investments

    The purpose of the paper is to study the financing models in order to drive the large amount of financial resources, already allocated for energy efficiency, to improve the quality of cities. These resources are being deployed worldwide by both public and private financial institutions. Energy efficiency is usually managed at the scale of the buildings, i.e., consumption reduction (heating, lighting, etc.). The study methodology is to review energy-efficiency finance (EEF) models, and assess them using multiple case studies. At the same time, the ownership of cities’ spaces is studied across public-private space management, as an effective methodology to bridge the gap between public and investor finance. The comparative analysis of the case studies suggests a paradigm shift in the definition of energy efficiency, not just in terms of the buildings, but instead also the local urban environment with its feedbacks on the quality of urban living. The practical implications are innovative EEF models, such as those being reviewed, which may be: (1) analytical, to assess the environment at the scale of blocks or neighbourhoods; (2) financial, to fund the specific scale; (3) relating to policy, to support and encourage. In recent years, support for urban regeneration is becoming particularly relevant, given the budget constraints of most public administrations and the conjunctural shortening of private partnerships

    The paper resumes some of the conversations the authors had in three years of research, based on the review of best participatory planning practices worldwide. The case projects are selected and discussed with the protagonists across four leading issues: Simulation, Scenario and Visioning, Government and Governance, and Scale. The case-oriented discussion is a peculiarity of the book , contributing to give shape to future cities or regions. The aim is to build a critical thinking on how urban planning, policy and design issues are faced differently or similarly throughout every cases studied. The book include the description of computer models and media, socio-political experiments and professional practices which help communicating the future effects of different design, policy and planning strategies and schemes with a wide range of aims: from information, through consultation, towards active participation. The cases have confirmed that simulation tools can impact on local government and can drive new forms of “glocal” governance, shaping and implementing future plans and projects at different scale and time span. The following paragraphs will point at some of the constant thoughts the authors had around the selection and editing of the book’s case studied and related issues

    The paper presents the ongoing research work on a software system for supporting the exploration of the numerous and often interrelated factors that can affect the urban design. The present implementation of the system supports the simulation of different urban scenarios in relation to the uniqueness and constraints peculiar to a design and a site. The paper considers our ongoing research work to formally represent the implicit and explicit knowledge used by means of ontologies. The ontology semantic system administrates a set of rules and relations among urban entities. To this aim, we are dealing with different issues: all the factors involved in the urban design process cross various domain knowledge, from different competencies and sources; the knowledge is both semantic and procedural

    PhOTO Zebrafish: A Transgenic Resource for In Vivo Lineage Tracing during Development and Regeneration

    Background: Elucidating the complex cell dynamics (divisions, movement, morphological changes, etc.) underlying embryonic development and adult tissue regeneration requires an efficient means to track cells with high fidelity in space and time. To satisfy this criterion, we developed a transgenic zebrafish line, called PhOTO, that allows photoconvertible optical tracking of nuclear and membrane dynamics in vivo. Methodology: PhOTO zebrafish ubiquitously express targeted blue fluorescent protein (FP) Cerulean and photoconvertible FP Dendra2 fusions, allowing for instantaneous, precise targeting and tracking of any number of cells using Dendra2 photoconversion while simultaneously monitoring global cell behavior and morphology. Expression persists through adulthood, making the PhOTO zebrafish an excellent tool for studying tissue regeneration: after tail fin amputation and photoconversion of a ~100µm stripe along the cut area, marked differences seen in how cells contribute to the new tissue give detailed insight into the dynamic process of regeneration. Photoconverted cells that contributed to the regenerate were separated into three distinct populations corresponding to the extent of cell division 7 days after amputation, and a subset of cells that divided the least were organized into an evenly spaced, linear orientation along the length of the newly regenerating fin. Conclusions/Significance: PhOTO zebrafish have wide applicability for lineage tracing at the systems-level in the early embryo as well as in the adult, making them ideal candidate tools for future research in development, traumatic injury and regeneration, cancer progression, and stem cell behavior

    Intramedullary non-specific inflammatory lesion of thoracic spine: A case report

    <p>Abstract</p> <p>Background</p> <p>There are several non-neoplastic lesions which mimick intramedullary spinal cord neoplasm in their radiographic and clinical presentation. These can be classified as either infectious (TB, fungal, bacterial, parasytic, syphilis, CMV, HSV) and non-infectious (sarcoid, MS, myelitis, ADEM, SLE) inflammatory lesions, idiopathic necrotizing myelopathy, unusual vascular lesions and radiation myelopathy. Although biopsy may be indicated in many cases, an erroneous diagnosis of intramedullary neoplasm can often be eliminated pre-operatively.</p> <p>Case description</p> <p>the authors report a very rare case of intramedullary non-specific inflammatory lesion of unknown origin, without signs of infection or demyelinization, in a woman who showed no other evidence of systemic disease.</p> <p>Conclusions</p> <p>Intramedullary lesions that mimick a tumor can be various and difficult to interpret. Preoperative MRI does not allow a certain diagnosis because these lesions have a very similar signal intensity pattern. Specific tests for infective pathologies are useful for diagnosis, but histological examination is essential for establishing a certain diagnosis. In our case the final histological examination and the specific tests that we performed have not cleared our doubts regarding the nature of the lesion that remains controversial.</p

    The Tumor Suppressor PRDM5 Regulates Wnt Signaling at Early Stages of Zebrafish Development

    PRDM genes are a family of transcriptional regulators that modulate cellular processes such as differentiation, cell growth and apoptosis. Some family members are involved in tissue or organ maturation, and are differentially expressed in specific phases of embryonic development. PRDM5 is a recently identified family member that functions as a transcriptional repressor and behaves as a putative tumor suppressor in different types of cancer. Using gene expression profiling, we found that transcriptional targets of PRDM5 in human U2OS cells include critical genes involved in developmental processes, and specifically in regulating wnt signaling. We therefore assessed PRDM5 function in vivo by performing loss-of-function and gain-of-function experiments in zebrafish embryos. Depletion of prdm5 resulted in impairment of morphogenetic movements during gastrulation and increased the occurrence of the masterblind phenotype in axin+/− embryos, characterized by the loss of eyes and telencephalon. Overexpression of PRDM5 mRNA had opposite effects on the development of anterior neural structures, and resulted in embryos with a shorter body axis due to posterior truncation, a bigger head and abnormal somites. In situ hybridization experiments aimed at analyzing the integrity of wnt pathways during gastrulation at the level of the prechordal plate revealed inhibition of non canonical PCP wnt signaling in embryos overexpressing PRDM5, and over-activation of wnt/β-catenin signaling in embryos lacking Prdm5. Our data demonstrate that PRDM5 regulates the expression of components of both canonical and non canonical wnt pathways and negatively modulates wnt signaling in vivo

    The Role of Glypicans in Wnt Inhibitory Factor-1 Activity and the Structural Basis of Wif1's Effects on Wnt and Hedgehog Signaling

    Proper assignment of cellular fates relies on correct interpretation of Wnt and Hedgehog (Hh) signals. Members of the Wnt Inhibitory Factor-1 (WIF1) family are secreted modulators of these extracellular signaling pathways. Vertebrate WIF1 binds Wnts and inhibits their signaling, but its Drosophila melanogaster ortholog Shifted (Shf) binds Hh and extends the range of Hh activity in the developing D. melanogaster wing. Shf activity is thought to depend on reinforcing interactions between Hh and glypican HSPGs. Using zebrafish embryos and the heterologous system provided by D. melanogaster wing, we report on the contribution of glypican HSPGs to the Wnt-inhibiting activity of zebrafish Wif1 and on the protein domains responsible for the differences in Wif1 and Shf specificity. We show that Wif1 strengthens interactions between Wnt and glypicans, modulating the biphasic action of glypicans towards Wnt inhibition; conversely, glypicans and the glypican-binding “EGF-like” domains of Wif1 are required for Wif1's full Wnt-inhibiting activity. Chimeric constructs between Wif1 and Shf were used to investigate their specificities for Wnt and Hh signaling. Full Wnt inhibition required the “WIF” domain of Wif1, and the HSPG-binding EGF-like domains of either Wif1 or Shf. Full promotion of Hh signaling requires both the EGF-like domains of Shf and the WIF domains of either Wif1 or Shf. That the Wif1 WIF domain can increase the Hh promoting activity of Shf's EGF domains suggests it is capable of interacting with Hh. In fact, full-length Wif1 affected distribution and signaling of Hh in D. melanogaster, albeit weakly, suggesting a possible role for Wif1 as a modulator of vertebrate Hh signaling
