    MarrowQuant Across Aging and Aplasia: A Digital Pathology Workflow for Quantification of Bone Marrow Compartments in Histological Sections.

    The bone marrow (BM) exists heterogeneously as hematopoietic/red or adipocytic/yellow marrow depending on skeletal location, age, and physiological condition. Mouse models and patients undergoing radio/chemotherapy or suffering acute BM failure endure rapid adipocytic conversion of the marrow microenvironment, the so-called "red-to-yellow" transition. Following hematopoietic recovery, such as upon BM transplantation, a "yellow-to-red" transition occurs and functional hematopoiesis is restored. Gold Standards to estimate BM cellular composition are pathologists' assessment of hematopoietic cellularity in hematoxylin and eosin (H&E) stained histological sections as well as volumetric measurements of marrow adiposity with contrast-enhanced micro-computerized tomography (CE-μCT) upon osmium-tetroxide lipid staining. Due to user-dependent variables, reproducibility in longitudinal studies is a challenge for both methods. Here we report the development of a semi-automated image analysis plug-in, MarrowQuant, which employs the open-source software QuPath, to systematically quantify multiple bone components in H&E sections in an unbiased manner. MarrowQuant discerns and quantifies the areas occupied by bone, adipocyte ghosts, hematopoietic cells, and the interstitial/microvascular compartment. A separate feature, AdipoQuant, fragments adipocyte ghosts in H&E-stained sections of extramedullary adipose tissue to render adipocyte area and size distribution. Quantification of BM hematopoietic cellularity with MarrowQuant lies within the range of scoring by four independent pathologists, while quantification of the total adipocyte area in whole bone sections compares with volumetric measurements. Employing our tool, we were able to develop a standardized map of BM hematopoietic cellularity and adiposity in mid-sections of murine C57BL/6 bones in homeostatic conditions, including quantification of the highly predictable red-to-yellow transitions in the proximal section of the caudal tail and in the proximal-to-distal tibia. Additionally, we present a comparative skeletal map induced by lethal irradiation, with longitudinal quantification of the "red-to-yellow-to-red" transition over 2 months in C57BL/6 femurs and tibiae. We find that, following BM transplantation, BM adiposity inversely correlates with kinetics of hematopoietic recovery and that a proximal to distal gradient is conserved. Analysis of in vivo recovery through magnetic resonance imaging (MRI) reveals comparable kinetics. On human trephine biopsies MarrowQuant successfully recognizes the BM compartments, opening avenues for its application in experimental, or clinical contexts that require standardized human BM evaluation

    Searching for R-Parity Violation at Run-II of the Tevatron

    We present an outlook for possible discovery of supersymmetry with broken R-parity at Run II of the Tevatron. We first present a review of the literature and an update of the experimental bounds. In turn we then discuss the following processes: 1. Resonant slepton production followed by R-parity violating decay, (a) via LQDcLQD^c and (b) via LLEcLLE^c. 2. How to distinguish resonant slepton production from ZZ' or WW' production. 3. Resonant slepton production followed by the decay to neutralino LSP, which decays via LQDcLQD^c. 4. Resonant stop production followed by the decay to a chargino, which cascades to the neutralino LSP. 5. Gluino pair production followed by the cascade decay to charm squarks which decay directly via L1Q2D1cL_1Q_2D^c_1. 6. Squark pair production followed by the cascade decay to the neutralino LSP which decays via L1Q2D1cL_1Q_2D^c_1. 7.     Pluronic® F127 Thermoresponsive Viscum album Hydrogel: Physicochemical Features and Cellular In Vitro Evaluation

    Viscum album L., popularly known as mistletoe, is well known for its anti-cancer properties, and the pharmaceutical application of hydroalcoholic dry extracts is still limited due to its low solubility in aqueous media, and physicochemical instability. The Pluronic® F127 is an amphiphilic polymer, which permits the solubilization of lipophilic and hydrophilic compounds. In this investigation, physicochemical features of hydrogel containing V. album dry extract (VADE-loaded-hydrogel) were performed by: dynamic light scattering (DLS), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), and transmission electron microscopy (TEM). VADE-loaded-hydrogel presented nanometer-size micelles with volume distribution ranging from 10.58 nm to 246.7 nm, and a polydispersity index of 0.441. The sample thermal analyses (TG and DSC) showed similar decomposition curves; however, the thermal events indicated an increase in thermal stability in relation to the presence of the extract. In addition to these interesting pharmaceutical features, IC50 values of 333.40 µg/mL and >1000 µg/mL were obtained when tumor (SCC-25) and non-tumor (L929) cells were incubated with VADE-loaded-hydrogel, respectively. The optical and ultrastructural cellular analysis confirmed the tumor selectivity since the following alterations were detected only in SCC-25 cells: disorganization of plasmatic membrane; an increase of cytoplasmatic vacuole size; alteration in the cristae mitochondrial shape; and generation of amorphous cellular material. These results emphasize the promising antitumoral potential of VADE-loaded-hydrogel as an herbal drug delivery system via in vitro assays

    O objetivo deste trabalho foi estimar os parâmetros genéticos, as correlações e os ganhos de seleção para a produção de óleo de progenies de híbridos interespecíficos entre caiaué, da origem 'Manicoré', e dendê da origem 'La Mé'. Trinta e nove progênies foram avaliadas, do sexto ao nono ano pós-plantio, quanto a produtividade de cachos de frutos frescos (PROD_FFB), teor de óleo no cacho (OCB) e produtividade de óleo de palma (PROD_OP). Foram estimados os parâmetros genéticos e os ganhos de seleção direta (GDS) e indireta (GIS) para PROD_OP. Altos valores de herdabilidade para a razão CVg/CVe indicaram condições favoráveis para a seleção. Com a seleção de 20% das progênies (seleção em ambos os sexos), as seguintes estimativas foram obtidas: 11,15% de GDS para PROD_OP, 9,1% GIS para OCB e 8,1% GIS para PROD_FFB, respectivamente. A PROD_OP das progênies foi de 6.175, 6.057 e 5.995 kg ha-1 por ano, com GSD e GSI em OCB e PROD_CFF, respectivamente. A seleção restrita a genitores masculinos de dendê LM2T resultou em 5,1% de GSD e na média de 5.800 kg ha-1 por ano para PROD_OP. Ganhos de seleção para PROD_OP podem ser obtidos imediatamente pela seleção restrita de genitores masculinos de dendê e, em médio e longo prazo, pela seleção recorrente recíproca interespecífica entre as populações de caiaué e dendê

    Matéria seca, pH e carboidrados sóluveis das silagens de seis genótipos de sorgo (Sorghum bicolor (L.) Moench).

    Foram utilizados seis genótipos de sorgo (Sorghum bicolor (L.) Moench) de porte alto e colmo seco com o objetivo de determinar as percentagens de Matéria Seca (MS), de Carboidratos Solúveis (CHOS) e o valor de pH. As silagens foram feitas com sorgo colhido no estádio de grão leitoso/pastoso, em silos de laboratório, feitos de PVC. Os híbridos 1 e 2 são as testemunhas comerciais (BRS610 e VOLUMAX, respectivamente), enquanto que os outros quatro são novos materiais desenvolvidos pela EMBRAPA Milho e Sorgo. Foram avaliados oito períodos, sendo que um destes é a forragem antes de ensilar (PO). Os silos foram abertos com 1; 3; 5; 7; 14; 28 e 56 dias de fermentação. que correspondem a P1; P2; P3; P4; P5; P6 e P7, respectivamente. Utilizou-se o teste SNK para comparação entre médias, com um delineamento experimental inteiramente ao acaso. Houve elevação do teor de MS com o processo de ensilagem. Nas silagens os valores de pH oscilaram de 3,69 a 3,98. De modo geral os híbridos de sorgo apresentaram bom padrão de fermentação, com boa classificação, que variou de boa a muito boa, quanto aos parâmetros utilizados
