    Crisis Intervention Team Training and the Protection Motivation Theory

    The problem addressed in this phenomenological study was the lack of documentation that supported the lived experiences of crisis intervention team (CIT)-trained police officers related to their field encounters with persons with mental illnesses. The purpose of the study was to explore the lived experiences of officers among CIT-trained police officers to address the problem. The protection motivation theory was aligned closest with the teachings of CIT training as described by the study participants’ lived experiences. Participants provided the study’s collected data, which was composed of completed questionnaires and transcribed interviews. The empirical theoretical framework method of analysis used was a combination of inductive coding and theme analysis that established the results of this study. Key findings of the study identified a significant amount of frustration expressed in the lived experiences of the CIT-trained police officers. Frustration was experienced by officers who applied the protection motivation theory to ensure the well-being of persons experiencing a mental crisis. There was considerable pushback from the public mental health facilities, which added to the frustration experienced by CIT-trained police officers who attempted to navigate treatment with the limited resources available to help persons in mental crisis. The positive social change produced from this study included recommendations to police leadership and mental health advocates to encourage certain CIT-training-related practices that directly impact CIT field encounters with persons in mental crises. Specialized training may promote improved departmental outcomes such as sustainability of gains for those in crises and enable police officer accountability and reliability

    On the design of the steering and front uprights for a race car

    Our contribution to fulfil the requirements for the mechanical design project was to audit the existing steering components and design an upright assembly for the JCU Motorsports FSAE car. We were required to assess the current status of the project and pick up where the previous team left off. As undergraduate engineers, we have followed the design process outlined in the assessment criteria issued at the commencement of the project. Many hours of work have been put into the design, material selection and loading analysis to come up with what we believe to be the best design given the constraints and timeline. A major guideline governing the design of the upright housing was the need for it to be reliable. This has been heavily factored into the design and is reflected in the analysis of the load cases giving significant room for errors in calculations and accounting for unforeseen circumstances which can cause significant angst for designers unfamiliar with the field. Overall the requirements of this project have been fulfilled to a high standard. We have taken what we have been given and produced a design to be incorporated into the motorsports project

    BBC and the Troubles: 1968 — 1998

    In 1985, Prime Minister Margaret Thatcher declared publicity to be the ‘oxygen’ of terrorism. Speaking from within a climate of domestic terrorism, such a statement draws into question the nature of contemporary media coverage. The British Broadcasting Corporation, existing as a public sector broadcaster, occupies a unique position in the context of 20th and 21st century mass media. The BBC is central to the creation and direction of national and international news agendas, in the formation of worldwide public opinion, and the brand name and reputation hold connotations of honesty, accuracy and impartiality. It can therefore be positioned as a ‘a microcosm of some larger system or a whole society' (Gomm et al., 2000, p.99). Yet, the historical visual output of the organisation in relation to domestic terrorism emanating from the environment of the Troubles — a significant period in social, cultural, political, and media history — has never been subject to rigorous academic scrutiny. Grounded in the field of media and cultural studies, and drawing upon extensive archival research, this thesis investigates the representation of domestic terror by the BBC in news and documentary format over the three-decade period of 1968-1998 through two interpretive modes of textual analysis: content analysis and semiotics. Throughout, the representation of events is contextualised in relation to media theory, with the words and pictures broadcast by the BBC analysed. The framing of acts of terror as image events is considered, as well as the visual aesthetic, codes, and values, of news reports. Ultimately, this work argues that BBC coverage of the Troubles has clear and identifiable patterns and symbols. Initial outbreaks of violence, where no corresponding representational referents existed, trended towards the vivid and graphic. Gradually, however, there was an overt movement away from this form; with the notable exception of moments where a method of perception created a disjuncture to established means, coverage was dominated by generic media templates, the rhetoric of euphemism, a concerted lack of contextualisation, and empty symbolism of the absent image

    Genetic diversity, structure and gene flow of migratory barren-ground caribou (Rangifer tarandus groenlandicus) in Canada

    Migratory barren-ground caribou (Rangifer tarandus groenlandicus) provide an opportunity to examine the genetic population structure of a migratory large mammal whose movements and distribution, in some instances, have not been heavily influenced by human activities that result in habitat loss or fragmentation. These caribou have likely reached large effective population sizes since their rapid radiation during the early Holocene despite cyclic changes in abundance. Migratory barren-ground caribou are managed as discrete subpopulations. We investigated genetic variation among those subpopulations to determine the patterns of genetic diversity within and among them, and the implications for long-term persistence of caribou. We identified three distinct genetic clusters across the Canadian arctic tundra: the first cluster consisted of all fully-continental migratory barren-ground subpopulations; the second cluster was the Dolphin and Union caribou; and the third cluster was caribou from Southampton Island. The Southampton Island caribou are especially genetically distinct from the other barren-ground type caribou. Gene flow among subpopulations varied across the range. Occasional gene flow across the sea-ice is likely the reason for high levels of genetic variation in the Dolphin and Union subpopulation, which experienced very low numbers in the past. These results suggest that for most migratory caribou subpopulations, connectivity among subpopulations plays an important role in maintaining natural genetic diversity. Our analyses provide insight into the levels of microsatellite genetic diversity and patterns of gene flow that may be common to large subpopulations that historically had a continuous distribution across a large continental range. These data can also be used as a benchmark to compare the effects of habitat fragmentation and bottlenecks on other large caribou populations

    The 9-1-1 checkpoint clamp stimulates DNA resection by Dna2-Sgs1 and Exo1

    Single-stranded DNA (ssDNA) at DNA ends is an important regulator of the DNA damage response. Resection, the generation of ssDNA, affects DNA damage checkpoint activation, DNA repair pathway choice, ssDNA-associated mutation and replication fork stability. In eukaryotes, extensive DNA resection requires the nuclease Exo1 and nuclease/helicase pair: Dna2 and Sgs1^(BLM). How Exo1 and Dna2-Sgs1^(BLM) coordinate during resection remains poorly understood. The DNA damage checkpoint clamp (the 9-1-1 complex) has been reported to play an important role in stimulating resection but the exact mechanism remains unclear. Here we show that the human 9-1-1 complex enhances the cleavage of DNA by both DNA2 and EXO1 in vitro, showing that the resection-stimulatory role of the 9-1-1 complex is direct. We also show that in Saccharomyces cerevisiae, the 9-1-1 complex promotes both Dna2-Sgs1 and Exo1-dependent resection in response to uncapped telomeres. Our results suggest that the 9-1-1 complex facilitates resection by recruiting both Dna2-Sgs1 and Exo1 to sites of resection. This activity of the 9-1-1 complex in supporting resection is strongly inhibited by the checkpoint adaptor Rad9^(53BP1). Our results provide important mechanistic insights into how DNA resection is regulated by checkpoint proteins and have implications for genome stability in eukaryotes

    Advanced Space Suit PLSS 2.0 Cooling Loop Evaluation and PLSS 2.5 Recommendations

    From 2012 to 2015 The NASA/JSC AdvSS (Advanced Space Suit) PLSS (Portable Life Support Subsystem) team, with support from UTC Aerospace Systems, performed the build-up, packaging and testing of PLSS 2.0. One aspect of that testing was the evaluation of the long-term health of the water cooling circuit and the interfacing components. Periodic and end-of-test water, residue and hardware analyses provided valuable information on the status of the water cooling circuit, and the approaches that would be necessary to enhance water cooling circuit health in the future. The evaluated data has been consolidated, interpreted and woven into an action plan for the maintenance of water cooling circuit health for the planned FY (fiscal year) 2016 through FY 2018 PLSS 2.5 testing. This paper provides an overview of the PLSS 2.0 water cooling circuit findings and the associated steps to be taken in that regard for the PLSS 2.5

    Refrigerated Stability of Diluted Cisatracurium, Rocuronium, and Vecuronium for skin testing after perioperative anaphylaxis

    Rationale: The purpose of this study is to investigate the stored stability of dilutions of neuromuscular blocking agents (NMBAs), namely cisatracurium, rocuronium, and vecuronium, for skin prick/intradermal testing. Methods: Concentrations of NMBAs were monitored by liquid chromatography-mass spectrometry (LC-MS/MS) for a period of 14 days. Dilutions of NMBAs were prepared in saline by factors of 10x, 100x, 1,000x, 10,000x, and 100,000x as sensitivity of the assay allowed. Diluted drug products were stored in a laboratory refrigerator until sampling. On sampling days, aliquots of each dilution were removed and compared to a freshly prepared set of reference dilutions. Results: The results are measured as beyond use date (BUD) defined as recovery of drug versus the reference (90-110%). Based on the LC-MS/MS data, the BUD for cisatracurium diluted to 10x and 100x is 96 hours. Higher dilutions (1,000x to100,000x) should be used immediately following preparation (within less than 24 hours). Vecuronium at 10x and 100x, also has a BUD of 96 hours, and the 1,000x dilution is stable for 24 hours. The 10,000x dilution should be used immediately. Rocuronium at 10x to 1,000x has a BUD of 48 hours, yet higher dilutions (10,000x and 100,000x) should be used immediately. Conclusions: With increasing dilution factors, the stability of these drugs in saline decreases, increasing deviation between samples and references. The most stable dilutions for each of the drugs tested were 10x and 100x. Stability of these drugs is likely compromised by hydrolysis of the ester bonds in the drug molecules
