    In vitro selection of salt-tolerant Ailanthus altissimaSwingle

    Salt-tolerant cell lines of Ailanthus altissima were selected from callus derived protoplasts. Murashige–Skoog (MS) liquid medium incorporated with various concentrations of NaCl was employed to enrich salt-tolerant A. altissima cell lines. Salt-resistant A. altissima cells were transferred on MS solid medium supplemented with 2.5 μM 2,4-dichlorophenoxy acetic acid (2,4-D), 0.5 μM benzyl adenine (BA) and various NaCl concentrations. The callus was cultured on MS medium containing NaCl for 5 months, to determine the survival rate as an index of salt tolerance. The measurement of growth parameters for salt-tolerant cells showed that the selected plant cell lines grew better than the unselected ones at all levels of NaCl tested. The salt-tolerant callus accumulated proline in correlation to the concentration of salts. Media supplemented with BA induced shoot differentiation of salt-resistant A. altissima cells

    Enhanced production of tropane alkaloids in transgenic Scopolia parviflora hairy root cultures over-expressing putrescine N-methyl transferase (PMT) and hyoscyamine-6β-hydroxylase (H6H)

    Scopolia parviflora adventitious roots were metabolically engineered by co-expression of the two gene putrescine N-methyl transferase (PMT) and hyoscyamine-6β-hydroxylase (H6H) cDNAs with the aid of Agrobacterium rhizogenes. The transformed roots developed into morphologically distinct S. parviflora PMT1 (SpPMT1), S. parviflora PMT1 (SpPMT2), and S. parviflora H6H (SpH6H) transgenic hairy root lines. Consequent to the introduction of these key enzyme genes, the production of the alkaloids hyoscyamine and scopolamine was enhanced. Among the transgenic hairy root lines, SpPMT2 line possessed the highest growth index. The treatment of transgenic hairy roots with growth regulators further enhanced the production of scopolamine. Thus, the results suggest that PMT1, PMT2, and H6H genes may not only be involved in the metabolic regulation of alkaloid production but also that these genes may play a role in the root development

    Crystal Structures of the HslVU Peptidase–ATPase Complex Reveal an ATP-Dependent Proteolysis Mechanism

    AbstractBackground: The bacterial heat shock locus HslU ATPase and HslV peptidase together form an ATP-dependent HslVU protease. Bacterial HslVU is a homolog of the eukaryotic 26S proteasome. Crystallographic studies of HslVU should provide an understanding of ATP-dependent protein unfolding, translocation, and proteolysis by this and other ATP-dependent proteases.Results: We present a 3.0 Å resolution crystal structure of HslVU with an HslU hexamer bound at one end of an HslV dodecamer. The structure shows that the central pores of the ATPase and peptidase are next to each other and aligned. The central pore of HslU consists of a GYVG motif, which is conserved among protease-associated ATPases. The binding of one HslU hexamer to one end of an HslV dodecamer in the 3.0 Å resolution structure opens both HslV central pores and induces asymmetric changes in HslV.Conclusions: Analysis of nucleotide binding induced conformational changes in the current and previous HslU structures suggests a protein unfolding–coupled translocation mechanism. In this mechanism, unfolded polypeptides are threaded through the aligned pores of the ATPase and peptidase and translocated into the peptidase central chamber

    Z boson pair production at LHC in a stabilized Randall-Sundrum scenario

    We study the Z boson pair production at LHC in the Randall-Sundrum scenario with the Goldberger-Wise stabilization mechanism. It is shown that comprehensive account of the Kaluza-Klein graviton and radion effects is crucial to probe the model: The KK graviton effects enhance the cross section of ggZZg g \to Z Z on the whole so that the resonance peak of the radion becomes easy to detect, whereas the RS effects on the qqˉZZq\bar{q} \to Z Z process are rather insignificant. The pTp_T and invariant-mass distributions are presented to study the dependence of the RS model parameters. The production of longitudinally polarized Z bosons, to which the SM contributions are suppressed, is mainly due to KK gravitons and the radion, providing one of the most robust methods to signal the RS effects. The 1σ1 \sigma sensitivity bounds on (Λπ,mϕ)(\Lambda_\pi, m_\phi) with k/MPl=0.1k/M_{\rm Pl} =0.1 are also obtained such that the effective weak scale Λπ\Lambda_\pi of order 5 TeV can be experimentally probed.Comment: 28 pages, LaTex file, 18 eps figure

    No one knows what attention is

    In this article, we challenge the usefulness of “attention” as a unitary construct and/or neural system. We point out that the concept has too many meanings to justify a single term, and that “attention” is used to refer to both the explanandum (the set of phenomena in need of explanation) and the explanans (the set of processes doing the explaining). To illustrate these points, we focus our discussion on visual selective attention. It is argued that selectivity in processing has emerged through evolution as a design feature of a complex multi-channel sensorimotor system, which generates selective phenomena of “attention” as one of many by-products. Instead of the traditional analytic approach to attention, we suggest a synthetic approach that starts with well-understood mechanisms that do not need to be dedicated to attention, and yet account for the selectivity phenomena under investigation. We conclude that what would serve scientific progress best would be to drop the term “attention” as a label for a specific functional or neural system and instead focus on behaviorally relevant selection processes and the many systems that implement them.Action Contro

    More and more hadron states are found to be difficult to be accommodated by the quenched quark models which describe baryons as 3-quark states and mesons as antiquark-quark states. Dragging out an antiquark-quark pair from the gluon field in hadrons should be an important excitation mechanism for hadron spectroscopy.     From crystal to amorphopus: a novel route towards unjamming in soft disk packings

    It is presented a numerical study on the unjamming packing fraction of bi- and polydisperse disk packings, which are generated through compression of a monodisperse crystal. In bidisperse systems, a fraction f_+ = 40% up to 80% of the total number of particles have their radii increased by \Delta R, while the rest has their radii decreased by the same amount. Polydisperse packings are prepared by changing all particle radii according to a uniform distribution in the range [-\Delta R,\Delta R]. The results indicate that the critical packing fraction is never larger than the value for the initial monodisperse crystal, \phi = \pi/12, and that the lowest value achieved is approximately the one for random close packing. These results are seen as a consequence of the interplay between the increase in small-small particle contacts and the local crystalline order provided by the large-large particle contacts.Comment: two columns, 14 pages, 12 figures, accepted for publication in Eur.     Bounds on Bipartitiely Shared Entanglement Reduced from Superposed Tripartite Quantum States

    For a tripartite pure state superposed by two individual states, the bipartitely shared entanglement can always be achieved by local measurements of the third party. Consider the different aims of the third party, i.e. maximizing or minimizing the bipartitely shared entanglement, we find bounds on both the possible bipartitely shared entanglement of the superposition state in terms of the corresponding entanglement of the two states being superposed. In particular, by choosing the concurrence as bipartite entanglement measure, we obtain calculable bounds for tripartite (22n)(2\otimes 2\otimes n) -dimensional cases.Comment: 8 pages, 2 figures.
