Sperm DNA damage in relation to lipid peroxidation following freezing-thawing of boar semen
This study investigated the relationships between lipid peroxidation (LPO) and sperm DNA damage following freezing-thawing of boar semen in different extenders. The comet assay was used to measure the extent of sperm DNA damage in a cryoprotectant-free extender or in cryoprotectant-based extenders after single and repeated freezing and thawing. As well as an analysis of sperm motion characteristics, mitochondrial function, membrane integrity, and lipid peroxidation (LPO) were assessed simultaneously with the measurements of sperm DNA damage. Consistent positive significant correlations were found between sperm DNA damage and LPO after freezing-thawing. Comet assay measurements showed that cryo-induced sperm DNA damage was more marked in the cryoprotectant-free extender, irrespective of freezing cycle. The frequency of sperm cells with damaged DNA increased with repeated freezing and thawing in the cryoprotectant-based extenders. Except for sperm DNA damage, there were no consistent associations between post-thaw sperm LPO and sperm quality characteristics. It could be suggested that the increased LPO of membrane phospholipids is associated with higher susceptibility of boar spermatozoa to cryo-induced DNA damage.Keywords: Comet assay measurements, cryopreservation, extenders, spermatozo
The effect of minor and major mergers on the evolution of low excitation radio galaxies
We use deep, ÎŒ r lesssim 28 mag arcsecâ2, r-band imaging from the Dark Energy Camera Legacy Survey to search for past, or ongoing, merger activity in a sample of 282 low-excitation radio galaxies (LERGs) at z 4Ï excess of major mergers in the LERGs with M * lesssim 1011 Mâ, with 10 ± 1.5% of these active galactic nuclei involved in such large-scale interactions compared to 3.2 ± 0.4% of control galaxies. This excess of major mergers in LERGs decreases with increasing stellar mass, vanishing by M * > 1011.3 Mâ. These observations show that minor mergers do not fuel LERGs, and are consistent with typical LERGs being powered by accretion of matter from their halo. Where LERGs are associated with major mergers, these objects may evolve into more efficiently accreting active galactic nuclei as the merger progresses and more gas falls on to the central engine
On the Progenitors of Core-Collapse Supernovae
Theory holds that a star born with an initial mass between about 8 and 140
times the mass of the Sun will end its life through the catastrophic
gravitational collapse of its iron core to a neutron star or black hole. This
core collapse process is thought to usually be accompanied by the ejection of
the star's envelope as a supernova. This established theory is now being tested
observationally, with over three dozen core-collapse supernovae having had the
properties of their progenitor stars directly measured through the examination
of high-resolution images taken prior to the explosion. Here I review what has
been learned from these studies and briefly examine the potential impact on
stellar evolution theory, the existence of "failed supernovae", and our

Anthropogenic Space Weather
Anthropogenic effects on the space environment started in the late 19th
century and reached their peak in the 1960s when high-altitude nuclear
explosions were carried out by the USA and the Soviet Union. These explosions
created artificial radiation belts near Earth that resulted in major damages to
several satellites. Another, unexpected impact of the high-altitude nuclear
tests was the electromagnetic pulse (EMP) that can have devastating effects
over a large geographic area (as large as the continental United States). Other
anthropogenic impacts on the space environment include chemical release ex-
periments, high-frequency wave heating of the ionosphere and the interaction of
VLF waves with the radiation belts. This paper reviews the fundamental physical
process behind these phenomena and discusses the observations of their impacts.Comment: 71 pages, 35 figure

Salmonella Typhi Colonization Provokes Extensive Transcriptional Changes Aimed at Evading Host Mucosal Immune Defense During Early Infection of Human Intestinal Tissue
Commensal microorganisms influence a variety of host functions in the gut, including immune response, glucose homeostasis, metabolic pathways and oxidative stress, among others. This study describes how Salmonella Typhi, the pathogen responsible for typhoid fever, uses similar strategies to escape immune defense responses and survive within its human host. To elucidate the early mechanisms of typhoid fever, we performed studies using healthy human intestinal tissue samples and âmini-guts,â organoids grown from intestinal tissue taken from biopsy specimens. We analyzed gene expression changes in human intestinal specimens and bacterial cells both separately and after colonization. Our results showed mechanistic strategies that S. Typhi uses to rearrange the cellular machinery of the host cytoskeleton to successfully invade the intestinal epithelium, promote polarized cytokine release and evade immune system activation by downregulating genes involved in antigen sampling and presentation during infection. This work adds novel information regarding S. Typhi infection pathogenesis in humans, by replicating work shown in traditional cell models, and providing new data that can be applied to future vaccine development strategies
Derivatization of estrogens enhances specificity and sensitivity of analysis of human plasma and serum by liquid chromatography tandem mass spectrometry
AbstractEstrogens circulate at concentrations less than 20pg/mL in men and postmenopausal women, presenting analytical challenges. Quantitation by immunoassay is unreliable at these low concentrations. Liquid chromatography tandem mass spectrometry (LCâMS/MS) offers greater specificity and sometimes greater sensitivity, but ionization of estrogens is inefficient. Introduction of charged moieties may enhance ionization, but many such derivatives of estrogens generate non-specific product ions originating from the âreagentâ group. Therefore an approach generating derivatives with product ions specific to individual estrogens was sought.Estrogens were extracted from human plasma and serum using solid phase extraction and derivatized using 2-fluoro-1-methylpyridinium-p-toluenesulfonate (FMP-TS). Electrospray in positive mode with multiple reaction monitoring using a QTrap 5500 mass spectrometer was used to quantify âFMPâ derivatives of estrogens, following LC separation.Transitions for the FMP derivatives of estrone (E1) and estradiol (E2) were compound specific (m/z 362â238 and m/z 364â128, respectively). The limits of detection and quantitation were 0.2pg on-column and the method was linear from 1â400pg/sample. Measures of intra- and inter-assay variability, precision and accuracy were acceptable (<20%). The derivatives were stable over 24h at 10°C (7â9% degradation). Using this approach, E1 and E2, respectively were detected in human plasma and serum: pre-menopausal female serum (0.5mL) 135â473, 193â722pmol/L; male plasma (1mL) 25â111, 60â180pmol/L and post-menopausal female plasma (2mL), 22â78, 29â50pmol/L.Thus FMP derivatization, in conjunction with LCâMS/MS, is suitable for quantitative analysis of estrogens in low abundance in plasma and serum, offering advantages in specificity over immunoassay and existing MS techniques
Detecting the Dependent Evolution of Biosequences
A probabilistic graphical model is developed in order to detect the dependent evolution between different sites in biological sequences. Given a multiple sequence alignment for each molecule of interest and a phylogenetic tree, the model can predict potential interactions within or between nucleic acids and proteins. Initial validation of the model is carried out using tRNA sequence data. The model is able to accurately identify the secondary structure of tRNA as well as several known tertiary interactions
A Historiometric Examination of Machiavellianism and a New Taxonomy of Leadership
Although researchers have extensively examined the relationship between charismatic leadership and Machiavellianism (Deluga, 2001; Gardner & Avolio, 1995; House & Howell, 1992), there has been a lack of investigation of Machiavellianism in relation to alternative forms of outstanding leadership. Thus, the purpose of this investigation was to examine the relationship between Machiavellianism and a new taxonomy of outstanding leadership comprised of charismatic, ideological, and pragmatic leaders. Using an historiometric approach, raters assessed Machiavellianism via the communications of 120 outstanding leaders in organizations across the domains of business, political, military, and religious institutions. Academic biographies were used to assess twelve general performance measures as well as twelve general controls and five communication specific controls. The results indicated that differing levels of Machiavellianism is evidenced across the differing leader types as well as differing leader orientation. Additionally, Machiavellianism appears negatively related to performance, though less so when type and orientation are taken into account.Yeshttps://us.sagepub.com/en-us/nam/manuscript-submission-guideline
