    Two seas for one great diversity: Checklist of the marine heterobranchia (Mollusca; Gastropoda) from the Salento Peninsula (South-East Italy)

    The Salento peninsula is a portion of the Italian mainland separating two distinct Mediterranean basins, the Ionian and the Adriatic seas. Several authors have studied the marine Heterobranchia (Mollusca, Gastropoda) fauna composition living in the Ionian Sea, but to date further knowledge regarding this interesting group of mollusks is still needed. Recent studies have corroborated the peculiarity of the Mediterranean Sea showing high levels of endemism and cryptic diversity. On the other hand, marine sea slugs have been revealed to be important indicators of the marine ecosystem's health, due to their species-specific diet that consist of a vast variety of sessile and benthic invertebrates. A baseline study of the marine Heterobranchia diversity is therefore a necessary step to reveal the hidden diversity and to monitor the possible presence of alien species. The present study shows results from approximately 600 scientific dives carried out during a nine-year period in all of the main submarine habitats of the studied area, while accounting for the marine Heterobranchia from both the Ionian and Adriatic Seas. With this contribution, the list of marine Heterobranchia inhabiting the Salento Peninsula rises to 160. Furthermore, it also reports, for the first time, the presence of one alien species and three new records for Italian waters. Ecological notes and geographical distribution for each added species are provided together with animal iconography, consisting mainly of in situ photographs, for species identification

    Leptin receptor expression and in vitro leptin actions on prostaglandin release and nitric oxide synthase activity in the rabbit oviduct

    In this study, we have examined the presence and the distribution of receptors for leptin (Ob-R) in the oviduct of rabbits, and the effects of leptin on the release of prostaglandin (PG) F2alpha and PGE2 and on the activity of nitric oxide (NO) synthase (NOS) by oviducts cultured in vitro. Rabbits were killed during the follicular phase and the oviducts were incubated in vitro with leptin, PGF2alpha, PGE2, NO donor and inhibitors of NOS and cyclo-oxigenase (COX). Using immunohistochemistry, Ob-R-like positive reaction was observed only in the cytoplasm of secretory cells, having stronger intensity in the infundibulum and ampulla tracts than in the isthmus. Both leptin and NO donor inhibited PGE2 release, whereas they enhanced PGF2alpha release; NOS inhibitor alone or with leptin increased PGE2 and decreased PGF2alpha production; NOS activity was enhanced by leptin, while PGs did not affect this enzyme. This study suggests that the oviduct could be a potential target for endocrine regulation by leptin, whose circulating levels may act as a metabolic signal modulating oviductal PG release through mediation of the NOS/NO system

    Role of endothelin-1 system in the luteolytic process of pseudopregnant rabbits

    The aim of this study was to better understand the role of the endothelin-1 (ET-1) system in the process of controlling the corpora lutea (CL) life span in rabbits. ET-1 (10 mug iv) administration at d 9 and 12 of pseudopregnancy induced a functional luteolysis within 24 h of injection, but it was ineffective at both d 4 and 6. Pretreatments with Bosentan, a dual ETA/ETB receptor antagonist, or cyclooxygenase ( COX) inhibitor blocked the luteolytic action of ET-1 but not that induced by prostaglandin F-2alpha (PGF(2alpha)). In CL cultured in vitro, ET-1 increased (P less than or equal to 0.01) both PGF(2alpha) production and luteal nitric oxide synthase activity but decreased (P less than or equal to 0.01) progesterone release. Addition of ETA receptor antagonist BQ123 or COX inhibitor blocked the ET-1 luteolytic effects. Positive staining for ET-1 receptors was localized in ovarian blood vessels, granulosa cells of large follicles, and luteal cells. Immunoblot analysis of ET-1 receptor protein revealed a strong band of approximately 48 kDa in d-9 CL. Up to d 6 of pseudopregnancy, ET-1 mRNA abundance in CL was poorly expressed but then increased (P less than or equal to 0.01) at d 9 and 13. ETA-receptor transcript increased (P less than or equal to 0.01) at d 6, remained at the same level up to d 13, and then declined to the lowest (P less than or equal to 0.01) levels at d 22. ETB-receptor mRNA increased (P less than or equal to 0.01) throughout the late-luteal stage from d 13 up to d 18. Our data suggest that the luteolytic action of ET-1 may be a result of PGF(2alpha) synthesis from both luteal and accessory cells, via the COX pathways

    The spectroscopic and functional properties of the single Met80Ala and double Tyr67His/Met80Ala mutants of human cytochrome c have been investigated in their ferric and ferrous forms, and in the presence of different ligands, in order to clarify the reciprocal effect of these two residues in regulating the access of exogenous molecules into the heme pocket. In the ferric state, both mutants display an aquo high spin and a low spin species. The latter corresponds to an OH- ligand in Met80Ala but to a His in the double mutant. The existence of these two species is also reflected in the functional behavior of the mutants. The observation that (i) a significant peroxidase activity is present in the Met80Ala mutants, (ii) the substitution of the Tyr67 by His leads to only a slight increase of the peroxidase activity in the Tyr67His/Met80Ala double mutant with respect to wild type, while the Tyr67His mutant behaves as wild type, as previously reported, suggests that the peroxidase activity of cytochrome c is linked to an overall conformational change of the heme pocket and not only to the disappearance of the Fe-Met80 bond. Therefore, in human cytochrome c there is an interplay between the two residues at positions 67 and 80 that affects the conformation of the distal side of the heme pocket, and thus the sixth coordination of the hem

    Cardoon meal as alternative protein source to soybean meal for limousine bulls fattening period: Effects on growth performances and meat quality traits

    SIMPLE SUMMARY: Beef cattle feeding strategies are commonly based on soybean meal utilization as a fundamental protein source. This feed, though, might have negative environmental impacts on the major areas of production and is becoming very expensive. Cardoon (Cynara cardunculus L.) is a resilient crop which represents a good opportunity in reclaiming and remediating unutilized lands. Cardoon seeds are rich in oil, which is extracted for industrial purposes, and the related by-products (press cake and meal) are characterized by high protein content and essential fatty acids. The aim of this study was to evaluate cardoon meal as a protein source during the Limousine bulls’ fattening period, in order to study a suitable alternative to develop and create low-input and low-emission feeding strategies. The results obtained in terms of growth performances showed no statistical difference between bulls fed the by control diet (containing soybean meal as main protein source) and animals fed by the experimental diet, where soybean meal was partially replaced by one containing cardoon meal. Meat quality traits were measured, and no differences between the groups were found. Hence, these by-products could be considered as a valuable solution in Limousine bulls’ fattening periods and could be used to represent a key factor to improve cattle-feeding sustainability. ABSTRACT: Soybean meal is the most important protein source in beef cattle feeding. The research of alternative protein sources to replace soy use, avoiding negative effects on in vivo performance and on the product’s quality, is an important issue. In this context, cardoon represents a non-OGM resilient crop that can be cultivated in marginal lands for extracting its seed oil (utilized for biodiesel and biodegradable bioplastic production) and whose and the residual meal from its seed oil (utilized for biodiesel and biodegradable bioplastic production) could be a suitable by-product for animal feeding, due to its fairly high protein content. The aim of this study was to evaluate the feasibility of using cardoon meal as an innovative protein source during the Limousine bulls’ fattening period. Thirty-two bulls were divided into two groups and fed with a diet containing soybean meal (SG) or partially replacing soybean meal with cardoon meal as a protein source (CG), respectively. The feeding trial lasted about 11 months. Growth performances and meat physical–chemical traits were evaluated. No statistical differences in feed efficiency, average daily gain, or in the main meat quality indicators, as well as in fatty acid profiles were found among the groups. Therefore, cardoon meal could be considered as an alternative to soybean meal in fattening Limousine bulls in order to enhance the sustainability of the farming system
