Shock accelerated vortex ring
The interaction of a shock wave with a spherical density inhomogeneity leads
to the development of a vortex ring through the impulsive deposition of
baroclinic vorticity. The present fluid dynamics videos display this phenomenon
and were experimentally investigated at the Wisconsin Shock Tube Laboratory's
(WiSTL) 9.2 m, downward firing shock tube. The tube has a square internal
cross-section (0.25 m x 0.25 m) with multiple fused silica windows for optical
access. The spherical soap bubble is generated by means of a pneumatically
retracted injector and released into free-fall 200 ms prior to initial shock
acceleration. The downward moving, M = 2.07 shock wave impulsively accelerates
the bubble and reflects off the tube end wall. The reflected shock wave
re-accelerates the bubble (reshock), which has now developed into a vortex
ring, depositing additional vorticity. In the absence of any flow disturbances,
the flow behind the reflected shock wave is stationary. As a result, any
observed motion of the vortex ring is due to circulation. The shocked vortex

The use of beta-D-glucanase as a substitute for Novozyme 234 in immunofluorescence and protoplasting
Novozym 234 has been used for many years to prepare protoplasts of Aspergillus nidulans and other fungi for transformation. It has also been very useful in immunofluorescence studies for partially digesting walls of fixed hyphae or germlings to allow antibodies to penetrate into the cytoplasm. In recent years, the availability of Novozym 234 has become problematic, and we have searched for combinations of available enzymes that are suitable for protoplasting and immunofluorescence studies in A. nidulans
Alanine-scanning mutagenesis of Aspergillus γ-tubulin yields diverse and novel phenotypes
This is the publisher's version, also available electronically from "www.molbiolcell.org".We have created 41 clustered charged-to-alanine scanning mutations of the mipA, γ-tubulin, gene of Aspergillus nidulans and have created strains carrying these mutations by two-step gene replacement and by a new procedure, heterokaryon gene replacement. Most mutant alleles confer a wild-type phenotype, but others are lethal or conditionally lethal. The conditionally lethal alleles exhibit a variety of phenotypes under restrictive conditions. Most have robust but highly abnormal mitotic spindles and some have abnormal cytoplasmic microtubule arrays. Two alleles appear to have reduced amounts of γ-tubulin at the spindle pole bodies and nucleation of spindle microtubule assembly may be partially inhibited. One allele inhibits germ tube formation. The cold sensitivity of two alleles is strongly suppressed by the antimicrotubule agents benomyl and nocodazole and a third allele is essentially dependent on these compounds for growth. Together our data indicate that γ-tubulin probably carries out functions essential to mitosis and organization of cytoplasmic microtubules in addition to its well-documented role in microtubule nucleation. We have also placed our mutations on a model of the structure of γ-tubulin and these data give a good initial indication of the functionally important regions of the molecule
The OPERA trial : a protocol for the process evaluation of a randomised trial of an exercise intervention for older people in residential and nursing accommodation
Background: The OPERA trial is large cluster randomised trial testing a physical activity intervention to address
depression amongst people living in nursing and residential homes for older people. A process evaluation was
commissioned alongside the trial and we report the protocol for this process evaluation. Challenges included the
cognitive and physical ability of the participants, the need to respect the privacy of all home residents, including
study non-participants, and the physical structure of the homes. Evaluation activity had to be organised around the
structured timetable of homes, leaving limited opportunities for data collection. The aims of this process evaluation
are to provide findings that will assist in the interpretation of the clinical trial results, and to inform potential
implementation of the physical activity intervention on a wider scale.
Methods/design: Quantitative data on recruitment of homes and individuals is being collected. For homes in the
intervention arm, data on dose and fidelity of the intervention delivered; including individual rates of participation
in exercise classes are collected. In the control homes, uptake and delivery of depression awareness training is
monitored. These data will be combined with qualitative data from an in-depth study of a purposive sample of
eight homes (six intervention and two control).
Discussion: Although process evaluations are increasingly funded alongside trials, it is still rare to see the findings
published, and even rarer to see the protocol for such an evaluation published. Process evaluations have the
potential to assist in interpreting and understanding trial results as well as informing future roll-outs of
interventions. If such evaluations are funded they should also be reported and reviewed in a similar way to the
trial outcome evaluation
Antigen depot is not required for alum adjuvanticity
Alum adjuvants have been in continuous clinical use for more than 80 yr. While the prevailing theory has been that depot formation and the associated slow release of antigen and/or inflammation are responsible for alum enhancement of antigen presentation and subsequent T- and B-cell responses, this has never been formally proven. To examine antigen persistence, we used the chimeric fluorescent protein EαGFP, which allows assessment of antigen presentation in situ, using the Y-Ae antibody. We demonstrate that alum and/or CpG adjuvants induced similar uptake of antigen, and in all cases, GFP signal did not persist beyond 24 h in draining lymph node antigen-presenting cells. Antigen presentation was first detectable on B cells within 6–12 h of antigen administration, followed by conventional dendritic cells (DCs) at 12–24 h, then finally plasmacytoid DCs at 48 h or later. Again, alum and/or CpG adjuvants did not have an effect on the magnitude or sequence of this response; furthermore, they induced similar antigen-specific T-cell activation in vivo. Notably, removal of the injection site and associated alum depot, as early as 2 h after administration, had no appreciable effect on antigen-specific T- and B-cell responses. This study clearly rules out a role for depot formation in alum adjuvant activity
The relationship between different types of dissociation and psychosis-like experiences in a non-clinical sample
This study investigates whether detachment-type dissociation, compartmentalisation-type dissociation or absorption was most strongly associated with psychosis-like experiences in the general population. Healthy participants (N=215) were tested with the Dissociative Experiences Scale (DES, for detachment-related dissociative experiences); the Harvard Group Scale of Hypnotic Susceptibility (HGSHS:A, for dissociative compartmentalisation); the Tellegen Absorption Scale (TAS, for non-clinical ‘functional’ dissociative experience); and two measures of psychotic-like experiences, the 21-item Peters et al. Delusions Inventory (PDI-21) and the Cardiff Anomalous Perceptions Scale (CAPS). In multiple regression analyses, DES and TAS but not HGSHS: A scores were found to be significantly associated with PDI-21 and CAPS overall scores. A post-hoc hierarchical cluster analysis checking for cluster overlap between DES and CAPS items, and the TAS and CAPS items showed no overlap between items on the DES and CAPS and minimal overlap between TAS and CAPS items, suggesting the scales measure statistically distinct phenomena. These results show that detachment-type dissociation and absorption, but not compartmentalisation-type dissociation are significantly associated with psychosis-like experiences in a non-clinical population
Roads in Rainforest: best practice guidelines for planning, design and management
These Guidelines were developed as a framework for understanding the primary ecological issues to be addressed in the planning, design and management of roads in rainforest environments. This manual has been created to assist in reducing the main impacts of roads, which are primarily:
• Habitat loss and fragmentation;
• Reduction in habitat quality;
• Edge effects;
• Reduced animal movements, resulting in restricted genetic flows and diminished survival rates in some species;
• Population reduction through road kill of wildlife species;
• Erosion, sedimentation and pollution;
• Impacts on scenic quality;
• Disturbance from vehicular noise, headlights and movement;
• Facilitation of the spread of exotic pests,
• weeds and diseases; and
• Direct mortality from road construction activities.
Cumulatively, these impacts threaten the biodiversity and integrity of ecosystem processes throughout Queensland’s tropical forests. It is therefore imperative that current and future road infrastructure be designed with consideration of these factors within the context of the natural environment.
This document provides a set of principles and supporting guidelines for implementing best practice planning, design and management for ecologically sustainable roads within rainforests
throughout Queensland. The intention of these Guidelines is to describe the particular aspects
of rainforest environments that are unique and thus require particular consideration when
designing and constructing roads in these habitats.
Therefore, these Guidelines were specifically developed to:
• Inform planners, engineers and managers about the key ecological elements (Principles) to consider when building roads in rainforest environments;
• Identify the most important issues to consider when planning, designing and implementing plans for road infrastructure so that the impacts to the primary ecological elements of rainforests are minimised. Each major issue is clarified within its own ‘Guideline’; and
• Provide a set of steps and checklists within each Guideline to ensure that the core Principles are maintained throughout the different phases of planning, design and construction
Interventions to reduce the risk of surgically transmitted Creutzfeldt–Jakob disease: a cost-effective modelling review
Background
Creutzfeldt–Jakob disease is a fatal neurological disease caused by abnormal infectious proteins called prions. Prions that are present on surgical instruments cannot be completely deactivated; therefore, patients who are subsequently operated on using these instruments may become infected. This can result in surgically transmitted Creutzfeldt–Jakob disease.
Objective
To update literature reviews, consultation with experts and economic modelling published in 2006, and to provide the cost-effectiveness of strategies to reduce the risk of surgically transmitted Creutzfeldt–Jakob disease.
Methods
Eight systematic reviews were undertaken for clinical parameters. One review of cost-effectiveness was undertaken. Electronic databases including MEDLINE and EMBASE were searched from 2005 to 2017. Expert elicitation sessions were undertaken. An advisory committee, convened by the National Institute for Health and Care Excellence to produce guidance, provided an additional source of information. A mathematical model was updated focusing on brain and posterior eye surgery and neuroendoscopy. The model simulated both patients and instrument sets. Assuming that there were potentially 15 cases of surgically transmitted Creutzfeldt–Jakob disease between 2005 and 2018, approximate Bayesian computation was used to obtain samples from the posterior distribution of the model parameters to generate results. Heuristics were used to improve computational efficiency. The modelling conformed to the National Institute for Health and Care Excellence reference case. The strategies evaluated included neither keeping instruments moist nor prohibiting set migration; ensuring that instruments were kept moist; prohibiting instrument migration between sets; and employing single-use instruments. Threshold analyses were undertaken to establish prices at which single-use sets or completely effective decontamination solutions would be cost-effective.
Results
A total of 169 papers were identified for the clinical review. The evidence from published literature was not deemed sufficiently strong to take precedence over the distributions obtained from expert elicitation. Forty-eight papers were identified in the review of cost-effectiveness. The previous modelling structure was revised to add the possibility of misclassifying surgically transmitted Creutzfeldt–Jakob disease as another neurodegenerative disease, and assuming that all patients were susceptible to infection. Keeping instruments moist was estimated to reduce the risk of surgically transmitted Creutzfeldt–Jakob disease cases and associated costs. Based on probabilistic sensitivity analyses, keeping instruments moist was estimated to on average result in 2.36 (range 0–47) surgically transmitted Creutzfeldt–Jakob disease cases (across England) caused by infection occurring between 2019 and 2023. Prohibiting set migration or employing single-use instruments reduced the estimated risk of surgically transmitted Creutzfeldt–Jakob disease cases further, but at considerable cost. The estimated costs per quality-adjusted life-year gained of these strategies in addition to keeping instruments moist were in excess of £1M. It was estimated that single-use instrument sets (currently £350–500) or completely effective cleaning solutions would need to cost approximately £12 per patient to be cost-effective using a £30,000 per quality-adjusted life-year gained value.
Limitations
As no direct published evidence to implicate surgery as a cause of Creutzfeldt–Jakob disease has been found since 2005, the estimations of potential cases from elicitation are still speculative. A particular source of uncertainty was in the number of potential surgically transmitted Creutzfeldt–Jakob disease cases that may have occurred between 2005 and 2018.
Conclusions
Keeping instruments moist is estimated to reduce the risk of surgically transmitted Creutzfeldt–Jakob disease cases and associated costs. Further surgical management strategies can reduce the risks of surgically transmitted Creutzfeldt–Jakob disease but have considerable associated costs.

Academic motherhood and fieldwork: Juggling time, emotions and competing demands
The idea and practice of going ‘into the field’ to conduct research and gather data is a deeply rooted aspect of Geography as a discipline. For global North Development Geographers, amongst others, this usually entails travelling to, and spending periods of time in, often far-flung parts of the global South. Forging a successful academic career as a Development Geographer in the UK, is therefore to some extent predicated on mobility. This paper aims to critically engage with the gendered aspects of this expected mobility, focusing on the challenges and time constraints that are apparent when conducting overseas fieldwork as a mother, unaccompanied by her children. The paper emphasises the emotion work that is entailed in balancing the competing demands of overseas fieldwork and mothering, and begins to think through the implications of these challenges in terms of the types of knowledge we produce, as well as in relation to gender equality within the academy
Hunting for answers: Linking lectures with the real world using a mobile treasure hunt app
Plants underpin our society providing food, fuel, medicines, clean air and water, positive mental health, and are central to biodiversity conservation. Despite this importance and an increasing need for people with plant-identification skills, many societies are becoming increasingly ignorant to the species with which they interact. To benefit both our undergraduates and the society they will enter, we applied mobile technology to improve plant identification and appreciation , while providing opportunities to practice transferable team work and verbal communication skills. Encouraging 'plant vision' will improve conservation efforts while increasing personal connections with green spaces, leading to mental health improvements for society. Summary • Despite the importance of plants to human civilization, many societies are becoming increasingly ignorant to the plants that inhabit their surrounding environment. A phenomenon known as 'plant blindness'. To address plant blindness in undergraduate students we designed an outdoor activity using a mobile phone app. Our aims were to identify the level of 'plant blindness' in our students; investigate engagement with the app and activity ; determine if we can raise awareness of links between lecture content and real world scenarios; and assess the student experience as a result of the activity in large classes. • The app chosen was ActionBound. Students were asked to find and photograph local examples of four plant families, along with identifying physiological benefits of features covered in lectures. Two different first year classes were exposed to this activity-Plant Science and Life on Earth. • The Plant Science students (60% success rate for three families; 55 students) were less plant blind than Life on Earth students (less than 44% success rate in any of the four families; 200 students). Students engaged well with the activity with all groups submitting sensible attempts at the responses. Most students reported that the activity increased links to lecture material and all but one student reported positive experiences. • Our students found the treasure hunt learning environment is a fun way to engage with the plant topics covered in lectures. In future iterations, we will more explicitly explain the links to potential careers and will address some of the logistical challenges faced in this first cohort.
