Elastin is Localised to the Interfascicular Matrix of Energy Storing Tendons and Becomes Increasingly Disorganised With Ageing
Tendon is composed of fascicles bound together by the interfascicular matrix (IFM). Energy storing tendons are more elastic and extensible than positional tendons; behaviour provided by specialisation of the IFM to enable repeated interfascicular sliding and recoil. With ageing, the IFM becomes stiffer and less fatigue resistant, potentially explaining why older tendons become more injury-prone. Recent data indicates enrichment of elastin within the IFM, but this has yet to be quantified. We hypothesised that elastin is more prevalent in energy storing than positional tendons, and is mainly localised to the IFM. Further, we hypothesised that elastin becomes disorganised and fragmented, and decreases in amount with ageing, especially in energy storing tendons. Biochemical analyses and immunohistochemical techniques were used to determine elastin content and organisation, in young and old equine energy storing and positional tendons. Supporting the hypothesis, elastin localises to the IFM of energy storing tendons, reducing in quantity and becoming more disorganised with ageing. These changes may contribute to the increased injury risk in aged energy storing tendons. Full understanding of the processes leading to loss of elastin and its disorganisation with ageing may aid in the development of treatments to prevent age related tendinopathy
Effect of fatigue loading on structure and functional behaviour of fascicles from energy-storing tendons
Tendons can broadly be categorized according to their function: those that act purely to position the limb and those that have an additional function as energy stores. Energy-storing tendons undergo many cycles of large deformations during locomotion, and so must be able to extend and recoil efficiently, rapidly and repeatedly. Our previous work has shown rotation in response to applied strain in fascicles from energy-storing tendons, indicating the presence of helical substructures which may provide greater elasticity and recovery. In the current study, we assessed how preconditioning and fatigue loading affect the ability of fascicles from the energy-storing equine superficial digital flexor tendon to extend and recoil. We hypothesized that preconditioned samples would exhibit changes in microstructural strain response, but would retain their ability to recover. We further hypothesized that fatigue loading would result in sample damage, causing further alterations in extension mechanisms and a significant reduction in sample recovery. The results broadly support these hypotheses: preconditioned samples showed some alterations in microstructural strain response, but were able to recover following the removal of load. However, fatigue loaded samples showed visual evidence of damage and exhibited further alterations in extension mechanisms, characterized by decreased rotation in response to applied strain. This was accompanied by increased hysteresis and decreased recovery. These results suggest that fatigue loading results in a compromised helix substructure, reducing the ability of energy-storing tendons to recoil. A decreased ability to recoil may lead to an impaired response to further loading, potentially increasing the likelihood of injury

Scientific possibilities of a solar electric powered rendezvous with comet Encke
The minimum scientific spacecraft instrumentation is considered that is likely to result in as complete an understanding of the composition, structure, and activity of a cometary nucleus as is possible without landing on it. The payload will also give useful results on secondary goals of a better understanding of physical processes in the inner and outer coma. Studies of composition, by means of an actual landing on the surface, details of the internal structure of the nucleus, and sample return were considered beyond the scope of this mission
Understanding and Application of Culturally Diverse Issues Within University Settings
This qualitative study focused on exploring the knowledge and beliefs of university faculty, staff and students regarding: the effects of multi-culturalism and immigration on their professional lives; the influence of cultural issues within the university setting; and, finally, the teaching and learning needs of culturally diverse students. The major findings include the limited knowledge base held by all groups in the sample about multi- culturalism and immigration with subsequent little effect on their professionall lives. However, of the three groups, the student group more readily recognized that they have little preparation for the multicultural interactions that will occur in their future professional practices. Although some faculty members included other teaching experiences and strategies to expose students to diversity, they also admitted to a lack of expertise in this area.Cette eĢtude qualitative explore les connaissances et les convictions des professeurs, du personnel universitaire et des eĢtudiants en ce qui a trait aux effets de l'immigration et de la diversiteĢ culturelle sur leur vie professionnelle, aĢ l'influence de la question culturelle dans l'environnement universitaire, ainsi qu'aux besoins lieĢs aĢ l'apprentissage et la formation des eĢtudiants de cultures diverses. Les reĢsultats essentiels indiquent la pieĢtre connaissance qu'ont ces diffeĢrents milieux des questions touchant la diversiteĢ culturelle, ce qui a pour correĢlat une absence apparente d'influence de cette diversiteĢ sur leur vie professionnelle. Toutefois, sur l'ensemble des trois groupes consulteĢs, les eĢtudiants semblent plus prompts aĢ reconnaiĢtre leur absence de formation en la matieĢre: ils se sentent peu preĢpareĢs aux interactions pluriculturelles inheĢrentes aĢ leur vie professionnelle aĢ venir. Gender Diversity in the UK Construction Industry
For many years, there has been an assumption that the UK construction industry is devoid of gender diversity. Part of this assumption was predicated on the representation of
workers in construction sites, who are predominantly male. However, there are many positions in the construction industry that are not public-facing, and the question is whether the assumption that males fill most of these roles is also true. This issue has existed in the construction industry for many years; it appears that placing the sole onus on construction companies to employ more women may not be producing the desired level of progress. This research aim to establish whether clients can influence gender diversity in the construction industry with an objective of determining the current position of inequality in the construction industry; and existing incentives and frameworks. The research adopted an exploratory approach, with data collected through existing literature and records of 20 major construction companies, followed by interviews with Six individuals working for UK construction companies and clients of varying sizes. The study will awaken the construction industry and how it can initiate new proposals or support schemes that have worked previously, to encourage more
women to join the construction industry. It reveals that clients have an important role to play if the construction industry is to improve on gender diversity through contractual commitments that could be monitored regularly throughout the duration of a project
Mesenchymal stem cell therapies for intervertebral disc degeneration: consideration of the degenerate niche
We have previously reported a synthetic LaponiteĀ® crosslinked pNIPAMācoāDMAc (NPgel) hydrogel, which induces nucleus pulposus (NP) cell differentiation of human MSCs (hMSCs) without the need for additional growth factors. Furthermore NP gel supports integration following injection into the disc and restores mechanical function to the disc. However, translation of this treatment strategy into clinical application is dependent on the survival and differentiation of hMSC to the correct cell phenotype within the degenerate IVD. Here, we investigated the viability and differentiation of hMSCs within NP gel within a catabolic microenvironment.
Human MSCs were encapsulated in NPgel and cultured for 4 weeks under hypoxia (5% O2) with Ā± calcium, ILā1Ī² and TNFĪ± either individually or in combination to mimic the degenerate environment. Cell viability, and cellular phenotype was investigated.
Stem cell viability was maintained within hydrogel systems for the 4 weeks investigated under all degenerate conditions. NP matrix markers: Agg and Col II and NP phenotypic markers: HIFā1Ī±, FOXF1 and PAX1 were expressed within the NPgel cultures and expression was not affected by culture within degenerate conditions. Alizarin red staining demonstrated increased calcium deposition under cultures containing CaCl2 indicating calcification of the matrix. Interestingly MMP's, ADAMTS 4 and Col I expression by hMSCs cultured in NPgel was upregulated by calcium but not by proāinflammatory cytokines ILā1Ī² and TNFĪ±.
Importantly ILā1Ī² and TNFĪ±, regarded as key contributors to disc degeneration, were not shown to affect the NP cell differentiation of MSCs in the NPgel. In agreement with our previous findings, NPgel alone was sufficient to induce NP cell differentiation of MSCs, with expression of both aggrecan and collagen type II, under both standard and degenerate culture conditions; thus could provide a therapeutic option for the repair of the NP during IVD degeneration

We use a new algorithm to find the stress-carrying backbone of ``generic''
site-diluted triangular lattices of up to 10^6 sites. Generic lattices can be
made by randomly displacing the sites of a regular lattice. The percolation
threshold is Pc=0.6975 +/- 0.0003, the correlation length exponent \nu =1.16
+/- 0.03 and the fractal dimension of the backbone Db=1.78 +/- 0.02. The number
of ``critical bonds'' (if you remove them rigidity is lost) on the backbone
