    The doctoral thesis aims to analyse the recent establishment of the Single Resolution Mechanism (SRM) which constitutes the second pillar of the European Banking Union, entrusted by the European legislator with the mission to ensure an orderly resolution of failing banks and groups, with minimum impact on the real economy and public finances of the participating Member States. The objective of this research is to highlight the main consequences arising from the establishment of such a mechanism within the EU framework. To this purpose, attention is paid, on the one hand, to the compatibility between the SRM regulation and the EU subsidiarity principle and, on the other hand, to the main problems related to the field of judicial protection. Chapter I recalls the historical-legal evolutionary process leading to the establishment of the Banking Union. Chapter II focuses on the analysis of the main provisions of the SRM regulation, with particular regard to the organizational and functional aspects of the entire mechanism; to the relationship between the SRB and the NRAs; to the decision-making process and, finally, to the powers they have been assigned with by both the SRM regulation and the BRRD. Once highlighted the SRM \u201cworking context\u201d, analysis have been conducted in order to study the SRM regulation through the \u201clegal filter\u201d of the subsidiarity principle (Chapter III). In this regard, the latter has been connected to the provision of Article 114 TFEU and the agencification process, which, according to the doctrinal point of view, can be regarded as one of the latest concrete expression of the subsidiarity principle, if not even a way of legitimizing such a principle. On the basis of the the relevant case law, together with the specific and unique features of the banking resolution field and the powers the SRB has been entrusted with, conclusions have been drawn about the compatibility of the SRM regulation with the subsidiarity principle, although not traditionally understood. Moreover, the main purpose being also to get a preliminary understanding of the judicial protection\u2019s effectiveness within the SRM, Chapter IV is devoted to identifying and illustrating the issues and challenges to be faced by those subjects which will be affected by resolution decisions. From this perspective, analysis have been carried out having regard to the allocation of competences outlined in the SRM regulation, namely a complex administrative (and therefore judicial) machinery, most of the time involving interrelations between the European and the national level. Finally, Chapter V attempts to summarize the main blocks of the research and to draw general conclusion on the SRM-related topics

    Improvement of an emotional lexicon for the evaluation of beers

    Emotional response has been the subject of many studies during the last years. Many studies have shown the importance of using consumers to generate emotional lexicons. Chaya et al. (2015) developed a consumer defined (CD) lexicon to assess emotional response elicited by beer products. Shortly after, van Zyl and Meiselman (2015) presented a procedure to ensure that emotional lists were fully composed by emotions. The present research was developed to improve and test the lexicon developed by Chaya et al. (2015) following the approach proposed by van Zyl and Meiselman (2015). The proposed procedure allowed an easy filtering of terms for the study of emotional response. As a consequence, the test was shorter, clearer, and easier to understand and to complete by consumers. The improved emotional lexicon of beer favoured 1) the efficiency of the research in terms of discrimination among samples, 2) the simplicity of use by the consumers

    Sobre la base de estudios morfológicos, anatómicos y filogenéticos de la subfamilia Allioideae, se presenta una nueva circunscripción de la tribu Leucocoryneae que comprende seis géneros americanos:Beauverdia, Ipheion, Leucocoryne, Nothoscordum, Tristagma y Zoellnerallium. Se incluye una sinopsis de la tribu Leucocoryneae con su descripción (enmendada), una clave para diferenciar las tribus y una clave de los géneros de Leucocoryneae, sus descripciones y distribución geográfica.Based on morphological, anatomical, and phylogenetic studies within subfamily Allioideae, a new circumscription of tribe Leucocoryneae is presented including six American genera: Beauverdia, Ipheion, Leucocoryne, Nothoscordum, Tristagma and Zoellnerallium.     Role of sphingolipids in the biogenesis and biological activity of extracellular vesicles

    Extracellular vesicles (EVs) are membrane vesicles released by both eukaryotic and prokaryotic cells; they not only serve physiological functions, such as disposal of cellular components, but also play pathophysiologic roles in inflammatory and degenerative diseases. Common molecular mechanisms for EV biogenesis are evident in different cell biological contexts across eukaryotic phyla, and inhibition of this biogenesis may provide an avenue for therapeutic research. The involvement of sphingolipids (SLs) and their enzymes on EV biogenesis and release has not received much attention in current research. Here, we review how SLs participate in EV biogenesis by shaping membrane curvature and how they contribute to EV action in target cells. First, we describe how acid and neutral SMases, by generating the constitutive SL, ceramide, facilitate biogenesis of EVs at the plasma membrane and inside the endocytic compartment. We then discuss the involvement of other SLs, such as sphingosine-1-phosphate and galactosyl-sphingosine, in EV formation and cargo sorting. Last, we look ahead at some biological effects of EVs mediated by changes in SL levels in recipient cells

    Ostreopsis sp. is a toxic marine benthic dinoflagellate that causes high biomass blooms, posing a threat to human health, marine biota and aquaculture activities, and negatively impacting coastal seawater quality. Species-specific identification and enumeration is fundamental because it can allow the implementation of all the necessary preventive measures to properly manage Ostreopsis spp. bloom events in recreational waters and aquaculture farms. The aim of this study was to apply a rapid and sensitive qPCR method to quantify Ostreopsis cf. ovata abundance in environmental samples collected from Mediterranean coastal sites and to develop site-specific environmental standard curves. Similar PCR efficiencies of plasmid and environmental standard curves allowed us to estimate the LSU rDNA copy number per cell. Moreover, we assessed the effectiveness of mitochondrial COI and cob genes as alternative molecular markers to ribosomal genes in qPCR assays for Ostreopsis spp. quantification

    Historical silk: a novel method to evaluate degumming with non-invasive infrared spectroscopy and spectral deconvolution

    : To correctly manage a collection of historical silks, it is important to detect if the yarn has been originally subjected to degumming. This process is generally applied to eliminate sericin; the obtained fiber is named soft silk, in contrast with hard silk which is unprocessed. The distinction between hard and soft silk gives both historical information and useful indications for informed conservation. With this aim, 32 samples of silk textiles from traditional Japanese samurai armors (15th-20th century) were characterized in a non-invasive way. ATR-FTIR spectroscopy has been previously used to detect hard silk, but data interpretation is challenging. To overcome this difficulty, an innovative analytical protocol based on external reflection FTIR (ER-FTIR) spectroscopy was employed, coupled with spectral deconvolution and multivariate data analysis. The ER-FTIR technique is rapid, portable, and widely employed in the cultural heritage field, but rarely applied to the study of textiles. The ER-FTIR band assignment for silk was discussed for the first time. Then, the evaluation of the OH stretching signals allowed for a reliable distinction between hard and soft silk. Such an innovative point of view, which exploits a "weakness" of FTIR spectroscopy-the strong absorption from water molecules-to indirectly obtain the results, can have industrial applications too

    Long-term prognosis of epilepsy, prognostic patterns and drug resistance: a population-based study

    BACKGROUND AND PURPOSE: Seizures in most people with epilepsy remit but prognostic markers are poorly understood. There is also little information on the long-term outcome of people who fail to achieve seizure control despite the use of two antiepileptic drugs (drug resistance). METHODS: People with a validated diagnosis of epilepsy in whom two antiepileptic drugs had failed were identified from primary care records. All were registered with one of 123 family physicians in an area of northern Italy. Remission (uninterrupted seizure freedom lasting 2 years or longer) and prognostic patterns (early remission, late remission, remission followed by relapse, no remission) were determined. RESULTS: In all, 747 individuals (381 men), aged 11 months to 94 years, were followed for 11 045.5 person-years. 428 (59%) were seizure-free. The probability of achieving 2-year remission was 18% at treatment start, 34% at 2 years, 45% at 5, 52% at 10 and 67% at 20 years (terminal remission, 60%). Epilepsy syndrome and drug resistance were the only independent predictors of 2- and 5-year remission. Early remission was seen in 101 people (19%), late remission in 175 (33%), remission followed by relapse in 85 (16%) and no remission in 166 (32%). Treatment response was the only variable associated with differing prognostic patterns. CONCLUSION: The long-term prognosis of epilepsy is favourable in most cases. Early seizure remission is not invariably followed by terminal remission and seizure outcome varies according to well-defined patterns. Prolonged seizure remission and prognostic patterns can be predicted by broad syndromic categories and the failure of two antiepileptic drugs

    Glioblastoma multiforme: a multidisciplinary approach to overcome chemoresistance and find new therapeutic strategies

    Objectives: Glioblastoma multiforme is the most frequent malignant brain tumor. Patients die within 15 months after diagnosis. The failure of current therapies is ascribed to a subpopulation of cells with stem-like properties, called glioma stem cells (GSCs). The aim of this study is to develop new effective therapies. Moreover, we want to better characterize the orthotopic xenograft model established by GSCs injection into NOD/SCID mice. Materials and methods: We tested Temolomide and Valproic acid treatments, alone and in combination, on seven GSC lines by MTT assay and we sequenced p53. Moreover, we characterized our xenograft model investigating the expression of stemness and differentiation markers by immunohistochemistry on FFPE tissues and by immunofluorescence on the correspondent cell line. Finally, we performed aCGH on the DNA extracted from the cell line and from FFPE tissues. Results: GSCs were resistant to Temozolomide and slightly sensitive to Valproic acid. The two drugs exerted a synergistic effect when combined performing a pre-conditioning with Valproic acid. Furthermore, several cell lines carry p53 mutations. IF and IHC showed a perfect correspondence for stemness markers expression, but discordant data for the others. aCGH analysis evidenced numerous alterations specific for the ex vivo sample, suggesting the presence of an in vivo clonal selection. Discussion: This work shows the importance of murine microenvironment in GSCs phenotype in vivo and suggests the possibility to use our combined treatment for therapeutic purposes. Conclusions: Orthotopic models from GSCs and in vitro grown cell lines represent good models for the development of GSC-targeted therapies

    Miniaturized NIR Spectrometers in a Nutshell: Shining Light over Sources of Variance

    The increasing portability and accessibility of miniaturized NIR spectrometers are promoting the spread of in-field and online applications. Alongside the successful outcomes, there are also several problems related to the acquisition strategies for each instrument and to experimental factors that can influence the collected signals. An insightful investigation of such factors is necessary and could lead to advancements in experimental set-up and data modelling. This work aimed to identify variation sources when using miniaturized NIR sensors and to propose a methodology to investigate such sources based on a multivariate method (ANOVA-Simultaneous Component Analysis) that considers the effects and interactions between them. Five different spectrometers were chosen for their different spectroscopic range and technical characteristics, and samples of worldwide interest were chosen as the case study. Comparing various portable sensors is interesting since results could significantly vary in the same application, justifying the idea that this kind of spectrometer is not to be treated as a general class of instruments
