    Identification of a Chlorovirus PBCV-1 Protein Involved in Degrading the Host Cell Wall during Virus Infection

    Chloroviruses are unusual among viruses infecting eukaryotic organisms in that they must, like bacteriophages, penetrate a rigid cell wall to initiate infection. Chlorovirus PBCV-1 infects its host, Chlorella variabilis NC64A by specifically binding to and degrading the cell wall of the host at the point of contact by a virus-packaged enzyme(s). However, PBCV-1 does not use any of the five previously characterized virus-encoded polysaccharide degrading enzymes to digest the Chlorella host cell wall during virus entry because none of the enzymes are packaged in the virion. A search for another PBCV-1-encoded and virion-associated protein identified protein A561L. The fourth domain of A561L is a 242 amino acid C-terminal domain, named A561LD4, with cell wall degrading activity. An A561LD4 homolog was present in all 52 genomically sequenced chloroviruses, infecting four different algal hosts. A561LD4 degraded the cell walls of all four chlorovirus hosts, as well as several non-host Chlorella spp. Thus, A561LD4 was not cell-type specific. Finally, we discovered that exposure of highly purified PBCV-1 virions to A561LD4 increased the specific infectivity of PBCV-1 from about 25–30% of the particles forming plaques to almost 50%. We attribute this increase to removal of residual host receptor that attached to newly replicated viruses in the cell lysates

    Lipocalin 2 modulates the cellular response to amyloid beta

    The production, accumulation and aggregation of amyloid beta (Aß) peptides in Alzheimer's disease (AD) are influenced by different modulators. Among these are iron and iron-related proteins, given their ability to modulate the expression of the amyloid precursor protein and to drive Aß aggregation. Herein, we describe that lipocalin 2 (LCN2), a mammalian acute-phase protein involved in iron homeostasis, is highly produced in response to Aß1-42 by choroid plexus epithelial cells and astrocytes, but not by microglia or neurons. Although Aß1-42 stimulation decreases the dehydrogenase activity and survival of wild-type astrocytes, astrocytes lacking the expression of Lcn2 are not affected. This protection results from a lower expression of the proapoptotic gene Bim and a decreased inflammatory response.     Life-Cycle and Genome of OtV5, a Large DNA Virus of the Pelagic Marine Unicellular Green Alga Ostreococcus tauri

    Large DNA viruses are ubiquitous, infecting diverse organisms ranging from algae to man, and have probably evolved from an ancient common ancestor. In aquatic environments, such algal viruses control blooms and shape the evolution of biodiversity in phytoplankton, but little is known about their biological functions. We show that Ostreococcus tauri, the smallest known marine photosynthetic eukaryote, whose genome is completely characterized, is a host for large DNA viruses, and present an analysis of the life-cycle and 186,234 bp long linear genome of OtV5. OtV5 is a lytic phycodnavirus which unexpectedly does not degrade its host chromosomes before the host cell bursts. Analysis of its complete genome sequence confirmed that it lacks expected site-specific endonucleases, and revealed the presence of 16 genes whose predicted functions are novel to this group of viruses. OtV5 carries at least one predicted gene whose protein closely resembles its host counterpart and several other host-like sequences, suggesting that horizontal gene transfers between host and viral genomes may occur frequently on an evolutionary scale. Fifty seven percent of the 268 predicted proteins present no similarities with any known protein in Genbank, underlining the wealth of undiscovered biological diversity present in oceanic viruses, which are estimated to harbour 200Mt of carbon

    There is a need in west central Florida and other winter strawberry (Fragaria xananassa Duchesne) production areas for an early ripening cultivar to replace or be an alternative to \u27Sweetcharlie\u27 (Chandleretal., 1997a). \u27Sweet Charlie\u27 has benefitted the Florida strawberry industry through its relatively high production of fruit early in the season, when market prices are generally high. But the average fruit size is small, and the texture of its fruit is often soft, making shipment and shelf life of \u27Sweet Charlie\u27 problematic. \u27Earlibrite\u27 strawberry has produced high early-season (December through February) yields of large, flavorful fruit at the Univ. of Florida\u27s Gulf Coast Research and Education Center in Dover (GCRECD over) and in three commercial fields in westcentral Florida. It is recommended for trial in areas with mild winter climates. The clone was named ‘Earlibrite’ because of its high early season production of bright red fruit, and to honor Earl Albregts, a soil scientist who spent his entire professional career (1967-96) at GCREC-Dover

    Most of the strawberries (Fragaria xananassa Duchesne) produced in Florida are shipped fresh to locations throughout the eastern United States and Canada (Florida Agricultural Statistics [www.nass.usda.gov/ fl]). Therefore, Florida growers need cultivars that produce fruit that are attractive and flavorful, and maintain these qualities during and after long-distance shipment. \u27Strawberry Festival\u27 strawberry has produced commercially acceptable yields of firm, attractive, and flavorful fruit in trials at the Univ. of Florida\u27s Gulf Coast Research and Education Center in Dover (GCREC-Dover) and in two commercial fields in west central Florida. It is recommended for trial in areas where strawberries are grown in the annual hill plasticulture system. The clone was named ‘Strawberry Festival’ in recognition of the Florida Strawberry Festival, an annual festival in Plant City that celebrates the abundant crop of berries harvested in eastern Hillsborough County during late February and early March
