    The partisan ties of lobbying firms

    This article examines lobbying firms as intermediaries between organized interests and legislators in the United States. It states a partisan theory of legislative subsidy in which lobbying firms are institutions with relatively stable partisan identities. Firms generate greater revenues when their clients believe that firms’ partisan ties are valued highly by members of Congress. It hypothesizes that firms that have partisan ties to the majority party receive greater revenues than do firms that do not have such ties, as well as that partisan ties with the House majority party lead to greater financial returns than do partisan ties to the Senate majority party. These hypotheses are tested using data available under the Lobbying Disclosure Act from 2008 to 2016. Panel regression analysis indicates that firms receive financial benefits when they have partisan ties with the majority party in the House but not necessarily with the Senate majority party, while controlling for firm-level covariates (number of clients, diversity, and organizational characteristics). A difference-in-differences analysis establishes that Democratically aligned lobbying firms experienced financial losses when the Republican Party reclaimed the House in 2011, but there were no significant differences between Republican and Democratic firms when the Republicans reclaimed the Senate in 2015

    Query Expansion for Survey Question Retrieval in the Social Sciences

    In recent years, the importance of research data and the need to archive and to share it in the scientific community have increased enormously. This introduces a whole new set of challenges for digital libraries. In the social sciences typical research data sets consist of surveys and questionnaires. In this paper we focus on the use case of social science survey question reuse and on mechanisms to support users in the query formulation for data sets. We describe and evaluate thesaurus- and co-occurrence-based approaches for query expansion to improve retrieval quality in digital libraries and research data archives. The challenge here is to translate the information need and the underlying sociological phenomena into proper queries. As we can show retrieval quality can be improved by adding related terms to the queries. In a direct comparison automatically expanded queries using extracted co-occurring terms can provide better results than queries manually reformulated by a domain expert and better results than a keyword-based BM25 baseline.Comment: to appear in Proceedings of 19th International Conference on Theory and Practice of Digital Libraries 2015 (TPDL 2015

    Packing fraction of trimodal spheres with small size ratio: An analytical expression

    In previous papers analytical expressions were derived and validated for the packing fraction of bimodal hard spheres with small size ratio, applicable to ordered (crystalline) [H. J. H. Brouwers, Phys. Rev. E 76, 041304 (2007);H. J. H. Brouwers, Phys. Rev. E 78, 011303 (2008)] and disordered (random) packings [H. J. H. Brouwers, Phys. Rev. E 87, 032202 (2013)]. In the present paper the underlying statistical approach, based on counting the occurrences of uneven pairs, i.e., the fraction of contacts between unequal spheres, is applied to trimodal discretely sized spheres. The packing of such ternary packings can be described by the same type of closed-form equation as the bimodal case. This equation contains the mean volume of the spheres and of the elementary cluster formed by these spheres; for crystalline arrangements this corresponds to the unit cell volume. The obtained compact analytical expression is compared with empirical packing data concerning random close packing of spheres, taken from the literature, comprising ternary binomial and geometric packings; good agreement is obtained.     Isolation of Mouse Cerebral Microvasculature for Molecular and Single-Cell Analysis

    Brain microvasculature forms a specialized structure, the blood-brain barrier (BBB), to maintain homeostasis and integrity of the central nervous system (CNS). The BBB dysfunction is emerging as a critical contributor to multiple neurological disorders, including stroke, traumatic brain injury, autoimmune multiple sclerosis, and neurodegenerative diseases. The brain microvasculature exhibits highly cellular and regional heterogeneity to accommodate dynamic changes of microenvironment during homeostasis and diseases. Thus, investigating the underlying mechanisms that contribute to molecular or cellular changes of the BBB is a significant challenge. Here, we describe an optimized protocol to purify microvessels from the mouse cerebral cortex using mechanical homogenization and density-gradient centrifugation, while maintaining the structural integrity and functional activity of the BBB. We show that the isolated microvessel fragments consist of BBB cell populations, including endothelial cells, astrocyte end-feet, pericytes, as well as tight junction proteins that seal endothelial cells. Furthermore, we describe the procedures to generate single-cell suspensions from isolated microvessel fragments. We demonstrate that cells in the single-cell suspensions are highly viable and suitable for single-cell RNA-sequencing analysis. This protocol does not require transgenic mice and cell sorting equipment to isolate fluorescence-labeled endothelial cells. The optimized procedures can be applied to different disease models to generate viable cells for single-cell analysis to uncover transcriptional or epigenetic landscapes of BBB component cells

    A Novel Combined Term Suggestion Service for Domain-Specific Digital Libraries

    Interactive query expansion can assist users during their query formulation process. We conducted a user study with over 4,000 unique visitors and four different design approaches for a search term suggestion service. As a basis for our evaluation we have implemented services which use three different vocabularies: (1) user search terms, (2) terms from a terminology service and (3) thesaurus terms. Additionally, we have created a new combined service which utilizes thesaurus term and terms from a domain-specific search term re-commender. Our results show that the thesaurus-based method clearly is used more often compared to the other single-method implementations. We interpret this as a strong indicator that term suggestion mechanisms should be domain-specific to be close to the user terminology. Our novel combined approach which interconnects a thesaurus service with additional statistical relations out-performed all other implementations. All our observations show that domain-specific vocabulary can support the user in finding alternative concepts and formulating queries.Comment: To be published in Proceedings of Theories and Practice in Digital Libraries (TPDL), 201

    Immunoresponsive gene 1 modulates the severity of brain injury in cerebral ischaemia

    Inflammatory stimuli induce immunoresponsive gene 1 expression that in turn catalyses the production of itaconate through diverting cis-aconitate away from the tricarboxylic acid cycle. The immunoregulatory effect of the immunoresponsive gene 1/itaconate axis has been recently documented in lipopolysaccharide-activated mouse and human macrophages. In addition, dimethyl itaconate, an itaconate derivative, was reported to ameliorate disease severity in the animal models of psoriasis and multiple sclerosis. Currently, whether immunoresponsive gene 1/itaconate axis exerts a modulatory effect in ischaemic stroke remains unexplored. In this study, we investigated whether immunoresponsive gene 1 plays a role in modulating ischaemic brain injury. In addition, the molecular mechanism underlying the protective effects of immunoresponsive gene 1 in ischaemic stroke was elucidated. Our results showed that immunoresponsive gene 1 was highly induced in the ischaemic brain following ischaemic injury. Interestingly, we found that IRG1-/- stroke animals exhibited exacerbated brain injury, displayed with enlarged cerebral infarct, compared to wild-type stroke controls. Furthermore, IRG1-/- stroke animals presented aggravated blood-brain barrier disruption, associated with augmented Evans blue leakage and increased immune cell infiltrates in the ischaemic brain. Moreover, IRG1-/- stroke animals displayed elevated microglia activation, demonstrated with increased CD68, CD86 and Iba1 expression. Further analysis revealed that immunoresponsive gene 1 was induced in microglia after ischaemic stroke, and deficiency in immunoresponsive gene 1 resulted in repressed microglial heme oxygenase-1 expression and exacerbated ischaemic brain injury. Notably, the administration of dimethyl itaconate to compensate for the deficiency of immunoresponsive gene 1/itaconate axis led to enhanced microglial heme oxygenase-1 expression, alleviated ischaemic brain injury, improved motor function and decreased mortality in IRG1-/- stroke animals. In summary, we demonstrate for the first time that the induction of immunoresponsive gene 1 in microglia following ischaemic stroke serves as an endogenous protective mechanism to restrain brain injury through heme oxygenase-1 up-regulation. Thus, our findings suggest that targeting immunoresponsive gene 1 may represent a novel therapeutic approach for the treatment of ischaemic stroke

    A significant proportion of early HCI research was guided by one very clear vision: that the existing theory base in psychology and cognitive science could be developed to yield engineering tools for use in the interdisciplinary context of HCI design. While interface technologies and heuristic methods for behavioral evaluation have rapidly advanced in both capability and breadth of application, progress toward deeper theory has been modest, and some now believe it to be unnecessary. A case is presented for developing new forms of theory, based around generic “systems of interactors.” An overlapping, layered structure of macro- and microtheories could then serve an explanatory role, and could also bind together contributions from the different disciplines. Novel routes to formalizing and applying such theories provide a host of interesting and tractable problems for future basic research in HCI
