    Hysteroscopic findings and intrauterine pathology treatment in Mexican infertile women

    Background: Hysteroscopy is a minimally invasive approach in gynecologic surgery and one of the main procedures performed on women undergoing fertility treatments. Intrauterine pathology negatively affects fertility by decreasing endometrial receptivity and embryo implantation success, and its prevalence has been reported between 19% and 62%. The aim of our study was to describe the hysteroscopic findings, prevalence of intrauterine pathology, the instruments used for the treatment of structural lesions in Mexican infertile women; and to compare the relationship of positive findings with the type of infertility. Methods: This was a descriptive, cross-sectional study conducted at fertility clinic at a private hospital. Results: We evaluated 191 hysteroscopies; the mean age of women was 35.5+3.2 years and the mean time of infertility 5.7+3.2 years. Primary infertility was the most prevalent (79.1%). In 118 cases (61.8%), uterine cavity abnormalities were diagnosed, the most frequent findings were: polyps (n=51, 26.7%), endometritis (n=30, 15.7%), fibroids (n=15, 7.6 %), synechiae (n=12, 6.5%), and müllerian anomalies (n=10, 5.3%). For structural pathology treatment, cold scissors and bipolar energy were used in 65.5% and 34.5%, respectively. Conclusions: Overall intrauterine pathology prevalence in our study population was 61.8%. Cold scissors and bipolar energy were used for structural lesions treatment. When comparing the relationship of hysteroscopic findings, no statistically significant difference was found in the presence of positive findings, with the type of infertility.

    Designed Coiled-Coil Peptides Inhibit the Type Three Secretion System of Enteropathogenic Escherichia coli

    BACKGROUND: Enteropathogenic E. coli (EPEC) and enterohemorrhagic E. coli (EHEC) are two categories of E. coli strains associated with human disease. A major virulence factor of both pathotypes is the expression of a type three secretion system (TTSS), responsible for their ability to adhere to gut mucosa causing a characteristic attaching and effacing lesion (A/E). The TTSS translocates effector proteins directly into the host cell that subvert mammalian cell biochemistry. METHODS/PRINCIPAL FINDINGS: We examined synthetic peptides designed to inhibit the TTSS. CoilA and CoilB peptides, both representing coiled-coil regions of the translocator protein EspA, and CoilD peptide, corresponding to a coiled-coil region of the needle protein EscF, were effective in inhibiting the TTSS dependent hemolysis of red blood cells by the EPEC E2348/69 strain. CoilA and CoilB peptides also reduced the formation of actin pedestals by the same strain in HEp-2 cells and impaired the TTSS-mediated protein translocation into the epithelial cell. Interestingly, CoilA and CoilB were able to block EspA assembly, destabilizing the TTSS and thereby Tir translocation. This blockage of EspA polymerization by CoilA or CoilB peptides, also inhibited the correct delivery of EspB and EspD as detected by immunoblotting. Interestingly, electron microscopy of bacteria incubated with the CoilA peptide showed a reduction of the length of EspA filaments. CONCLUSIONS: Our data indicate that coiled-coil peptides can prevent the assembly and thus the functionality of the TTSS apparatus and suggest that these peptides could provide an attractive tool to block EPEC and EHEC pathogenesis

    Effects of cytomegalovirus infection in human neural precursor cells depend on their differentiation state

    © 2015, Journal of NeuroVirology, Inc. Cytomegalovirus (CMV) is the most common cause of congenital infection in developed countries and a major cause of neurological disability in children. Although CMV can affect multiple organs, the most important sequelae of intrauterine infection are related to lesions of the central nervous system. However, little is known about the pathogenesis and the cellular events responsible for neuronal damage in infants with congenital infection. Some studies have demonstrated that neural precursor cells (NPCs) show the greatest susceptibility to CMV infection in the developing brain. We sought to establish an in vitro model of CMV infection of the developing brain in order to analyze the cellular events associated with invasion by this virus. To this end, we employed two cell lines as a permanent source of NPC, avoiding the continuous use of human fetal tissue, the human SK-N-MC neuroblastoma cell line, and an immortalized cell line of human fetal neural origin, hNS-1. We also investigated the effect of the differentiation stage in relation to the susceptibility of these cell lines by comparing the neuroblastoma cell line with the multipotent cell line hNS-1. We found that the effects of the virus were more severe in the neuroblastoma cell line. Additionally, we induced hNS-1 to differentiate and evaluated the effect of CMV in these differentiated cells. Like SK-N-MC cells, hNS-1-differentiated cells were also susceptible to infection. Viability of differentiated hNS-1 cells decreased after CMV infection in contrast to undifferentiated cells. In addition, differentiated hNS-1 cells showed an extensive cytopathic effect whereas the effect was scarce in undifferentiated cells.     Life Project for Adolescents: A Concept Analysis

    It is essential to identify protective factors during adolescence due to its high incidence on risk behaviors. One of these factors is to have a life project that influences adolescent decision-making. The concept of life project has mainly been linked to teen pregnancy, depression and suicide; however, some authors agree that the concept is not clearly defined. Therefore, the objective of this paper is to define the concept of adolescent life project, using the methodology developed by Walker and Avant for concept analysis. The following steps were followed are: select a concept, determining the purpose of analysis, identifying all uses of the concept, determining the defining attributes, identifying a model, a borderline and a contrary cases, identifying history and consequences of the concept, and finally defining empirical referents. After the analysis, the final definition of the concept of adolescent life project includes the set of desires, future plans and actions necessary to accomplish it that influence the adolescent decisions. Knowing the importance of this concept when working with adolescents may guide development more effective interventions

    Hiperplasia suprarrenal no clásica, características de laboratorio y respuesta al tratamiento. Estudio de cohorte

    Objetivo: comparar las características de laboratorio y la respuesta al tratamiento en una cohorte de pacientes con hiperplasia suprarrenal congénita no clásica seguidas durante 12 meses. Métodos: se realizó un estudio en una cohorte final de 38 pacientes con hiperplasia suprarrenal no clásica evaluadas entre enero de 2006 hasta diciembre de 2016 y que tuvieron seguimiento a 1 año. Las características de la población de estudio y la respuesta de 17-hidroxiprogesterona (17-OH progesterona), testosterona y dehidroepiandrosterona sulfato (DHEA-S) se informan a los 6 y 12 meses después del tratamiento con dexametasona y etinilestradiol/acetato de ciproterona. Se realizó una regresión lineal múltiple para explicar el cambio de las variables de resultado (17- OH progesterona, testosterona y DHEA-S), ajustado de acuerdo con el índice de masa corporal (IMC) y la edad como covariables. Resultados: la edad promedio de los participantes fue de 25 años, el IMC promedio fue de 28,3 kg/m2. Las características clínicas más prevalentes fueron el acné (52,6 %) y la oligomenorrea (55 %). El hirsutismo se presentó en todos los casos en algún grado (moderado: 31,6 % y grave: 39,5 %, según la escala de Ferriman y Gallwey). El cambio en los niveles de cada una de las hormonas evidenció una disminución significativa de la mediana del rango intercuartílico de 17-OH progesterona, testosterona y DHEA-S, a los 6 y 12 meses (p < 0,05). En la regresión lineal múltiple, los niveles de testosterona presentaron una variación de acuerdo con el IMC (p = 0,04). En el presente estudio, la terapia farmacológica con dexametasona se aplicó a diferentes dosis y todas las pacientes recibieron anticonceptivos con una respuesta clínica y paraclínica adecuada al año de seguimiento. Conclusión: este estudio sugiere que la combinación de etinilestradiol más acetato de ciproterona con dosis variables de dexametasona disminuye el perfil androgénico en las pacientes con hiperplasia suprarrenal no clásica seguidas durante un año

    A beam-beam monitoring detector for the MPD experiment at NICA

    The Multi-Purpose Detector (MPD) is to be installed at the Nuclotron Ion Collider fAcility (NICA) of the Joint Institute for Nuclear Research (JINR). Its main goal is to study the phase diagram of the strongly interacting matter produced in heavy-ion collisions. These studies, while providing insight into the physics of heavy-ion collisions, are relevant for improving our understanding of the evolution of the early Universe and the formation of neutron stars. In order to extend the MPD trigger capabilities, we propose to include a high granularity beam-beam monitoring detector (BE-BE) to provide a level-0 trigger signal with an expected time resolution of 30 ps. This new detector will improve the determination of the reaction plane by the MPD experiment, a key measurement for flow studies that provides physics insight into the early stages of the reaction. In this work, we use simulated Au+Au collisions at NICA energies to show the potential of such a detector to determine the event plane resolution, providing further redundancy to the detectors originally considered for this purpose namely, the Fast Forward Detector (FFD) and the Hadron Calorimeter (HCAL). We also show our results for the time resolution studies of two prototype cells carried out at the T10 beam line at the CERN PS complex.Comment: 16 pages, 12 figures.     Benchmarking of human Y-chromosomal haplogroup classifiers with whole-genome and whole-exome sequence data

    In anthropological, medical, and forensic studies, the nonrecombinant region of the human Y chromosome (NRY) enables accurate reconstruction of pedigree relationships and retrieval of ancestral information. Using high-throughput sequencing (HTS) data, we present a benchmarking analysis of command-line tools for NRY haplogroup classification. The evaluation was performed using paired Illumina data from whole-genome sequencing (WGS) and whole-exome sequencing (WES) experiments from 50 unrelated donors. Additionally, as a validation, we also used paired WGS/WES datasets of 54 individuals from the 1000 Genomes Project. Finally, we evaluated the tools on data from third-generation HTS obtained from a subset of donors and one reference sample. Our results show that WES, despite typically offering less genealogical resolution than WGS, is an effective method for determining the NRY haplogroup. Y-LineageTracker and Yleaf showed the highest accuracy for WGS data, classifying precisely 98% and 96% of the samples, respectively. Yleaf outperforms all benchmarked tools in the WES data, classifying approximately 90% of the samples. Yleaf, Y-LineageTracker, and pathPhynder can correctly classify most samples (88%) sequenced with third-generation HTS. As a result, Yleaf provides the best performance for applications that use WGS and WES. Overall, our study offers researchers with a guide that allows them to select the most appropriate tool to analyze the NRY region using both second- and third-generation HTS data

    ERCP and splenic injury

    Abstractendoscopic Retrograde Colangiopancreatography (eRCP) is an invasive procedure with important complications that occurs in 5%-10% of the cases. The most frequent procedure related complications are: acute pancreatitis, hemorrhage, perforation and infection. an infrequent but potentially life threatening eRCP complication is the splenic injury, with very few cases reported in the literature. We report a patient with cholecholithiasis and biliary pancreatitis who was diagnosed with a subcapsular splenic laceration a few hours after an eRCP. Clinicians should be alerted to this potential post-procedure complication associated with eRCP
