    Space Time Defects as a Source of Curvature and Torsion

    Space time is described as a continuum four-dimensional medium similar to ordinary elastic continua. Exploiting the analogy internal stress states are considered. The internal ''stress'' is originated by the presence of defects. The defects are described according to the typical Volterra process. The case of a point defect in an otherwise isotropic four-dimensional medium is discussed showing that the resulting metric tensor corresponds to an expanding (or contracting) universe filled up with a non-zero energy-momentum density.Comment: Presentation at the Sixth Friedman seminar in Cargese 28/6-3/7/2004.     Regulated mitochondrial DNA replication during oocyte maturation is essential for successful porcine embryonic development.

    Cellular ATP is mainly generated through mitochondrial oxidative phosphorylation, which is dependent on mitochondrial DNA (mtDNA). We have previously demonstrated the importance of oocyte mtDNA for porcine and human fertilization. However, the role of nuclear-encoded mitochondrial replication factors during oocyte and embryo development is not yet understood. We have analyzed two key factors, mitochondrial transcription factor A (TFAM) and polymerase gamma (POLG), to determine their role in oocyte and early embryo development. Competent and incompetent oocytes, as determined by brilliant cresyl blue (BCB) dye, were assessed intermittently during the maturation process for TFAM and POLG mRNA using real-time RT-PCR, for TFAM and POLG protein using immunocytochemistry, and for mtDNA copy number using real-time PCR. Analysis was also carried out following treatment of maturing oocytes with the mtDNA replication inhibitor, 2',3'-dideoxycytidine (ddC). Following in vitro fertilization, preimplantation embryos were also analyzed. Despite increased levels of TFAM and POLG mRNA and protein at the four-cell stage, no increase in mtDNA copy number was observed in early preimplantation development. To compensate for this, mtDNA appeared to be replicated during oocyte maturation. However, significant differences in nuclear-encoded regulatory protein expression were observed between BCB(+) and BCB(-) oocytes and between untreated oocytes and those treated with ddC. These changes resulted in delayed mtDNA replication, which correlated to reduced fertilization and embryonic development. We therefore conclude that adherence to the regulation of the timing of mtDNA replication during oocyte maturation is essential for successful embryonic development

    Video Evidence That London Infants Can Resettle Themselves Back to Sleep After Waking in the Night, as well as Sleep for Long Periods, by 3 Months of Age

    Objective: Most infants become settled at night by 3 months of age, whereas infants not settled by 5 months are likely to have long-term sleep-waking problems. We assessed whether normal infant development in the first 3 months involves increasing sleep-period length or the ability to resettle autonomously after waking in the night. Methods: One hundred one infants were assessed at 5 weeks and 3 months of age using nighttime infrared video recordings and parental questionnaires. Results: The clearest development was in sleep length; 45% of infants slept continuously for 5 hours or more at night at 3 months compared with 10% at 5 weeks. In addition, around a quarter of infants woke and resettled themselves back to sleep in the night at each age. Autonomous resettling at 5 weeks predicted prolonged sleeping at 3 months suggesting it may be a developmental precursor. Infants reported by parents to sleep for a period of 5 hours or more included infants who resettled themselves and those with long sleeps. Three-month olds fed solely breast milk were as likely to self-resettle or have long sleep bouts as infants fed formula or mixed breast and formula milk. Conclusions: Infants are capable of resettling themselves back to sleep in the first 3 months of age; both autonomous resettling and prolonged sleeping are involved in “sleeping through the night” at an early age. Findings indicate the need for physiological studies of how arousal, waking, and resettling develop into sustained sleeping and of how environmental factors support these endogenous and behavioral processes

    Near-ionization-threshold emission in atomic gases driven by intense sub-cycle pulses

    We study theoretically the dipole radiation of a hydrogen atom driven by an intense sub-cycle pulse. The time-dependent Schr\"odinger equation for the system is solved by ab initio calculation to obtain the dipole response. Remarkably, a narrowband emission lasting longer than the driving pulse appears at a frequency just above the ionization threshold. An additional calculation using the strong field approximation also recovers this emission, which suggests that it corresponds to the oscillation of nearly-bound electrons that behave similarly to Rydberg electrons. The predicted phenomenon is unique to ultrashort driving pulses but not specific to any particular atomic structure.Comment: 8 pages, 2 figure
