Evaluation of four different strategies to characterize plasma membrane proteins from banana roots

Plasma membrane proteins constitute a very important class of proteins. They are involved in the transmission of external signals to the interior of the cell and selective transport of water, nutrients and ions across the plasma membrane. However, the study of plasma membrane proteins is challenging because of their poor solubility in aqueous media and low relative abundance. In this work, we evaluated four different strategies for the characterization of plasma membrane proteins from banana roots: (i) the aqueous-polymer two-phase system technique (ATPS) coupled to gelelectrophoresis (gel-based), and (ii) ATPS coupled to LC-MS/MS (gel free), (iii) a microsomal fraction and (iv) a full proteome, both coupled to LC-MS/ MS. Our results show that the gel-based strategy is useful for protein visualization but has major limitations in terms of time reproducibility and efficiency. From the gel-free strategies, the microsomal-based strategy allowed the highest number of plasma membrane proteins to be identified, followed by the full proteome strategy and by the ATPS based strategy. The high yield of plasma membrane proteins provided by the microsomal fraction can be explained by the enrichment of membrane proteins in this fraction and the high throughput of the gel-free approach combined with the usage of a fast high-resolution mass spectrometer for the identification of proteins

Characterizing fruit ripening in plantain and Cavendish bananas: A proteomics approach

The fruit physiology of banana cultivars other than Cavendish is poorly understood. To study the ripening process, samples were taken daily from plantain and Cavendish bananas and the ripening stages were determined. We present data from the green to the fully mature stage. By analyzing the protein abundances during ripening we provide some new insights into the ripening process and how plantains fruits are different. Multivariate analysis of the proteins was performed correlated to the starch dynamics. A drop in sucrose synthase and a rise of acid invertase during ripening indicated a change in the balance of the sucrose fate. During ripening, sugars may no longer be available for respiration since they are stored in the vacuoles, making citrate the preferred respiratory substrate. We found significant cultivar specific differences in granule-bound starch synthase, alpha- and beta amylases and cell wall invertase when comparing the protein content at the same ripening stage. This corroborates the difference in starch content/structure between both banana types. Differences in small heat shock proteins and in the cell wall-modifying enzyme xyloglucan endotransglucosylase/hydrolase support respectively the presumed higher carotenoid content and the firmer fruit structure of plantains

O impacto da auditoria interna na gestão municipal: o caso dos municípios portugueses

Nos últimos tempos, a investigação em gestão pública tem evoluído significativamente. No entanto, apesar dessa evolução, são ainda várias as questões que permanecem sem resposta na investigação em Administração Pública. O estudo do impacto e da importância da atividade da auditoria interna na gestão municipal é disso exemplo. Este estudo teve como principal objetivo perceber em que medida as informações veiculadas pela atividade de auditoria interna influenciam a tomada de decisão dos órgãos executivos. Foram aplicados dois inquéritos por questionário. Um aos responsáveis pela auditoria interna e outro aos responsáveis do órgão executivo dos 278 municípios de Portugal continental. Os resultados obtidos permitem concluir que os respondentes têm o mesmo entendimento quanto ao impacto e à importância da atividade de auditoria interna na gestão municipal. Apesar de algumas das variáveis independentes não estarem diretamente associadas à variável dependente, observa-se que a auditoria é importante na tomada de decisões, quer pelos executivos como pelos demais utilizadores da informação produzida pelos auditores internos.Este estudo foi realizado no Centro de Investigação em Ciência Política (UID/CPO/00758/2013), Universidade do Minho e apoiado pela Fundação para a Ciência e a Tecnologia e pelo Ministério da Educação e Ciência através de fundos nacionais. No entanto, apesar dessa evolução são ainda várias as questões que permanecem sem resposta na investigação em Administração Pública. O estudo do impacto e da importância da atividade da auditoria interna na gestão municipal é exemplo disso. Este estudo tem como principal objetivo perceber em que medida, e de que maneira as informações veiculadas pela atividade de auditoria interna influenciam na tomada de decisão pelos órgãos executivos. Para a realização deste estudo, foram aplicados dois inquéritos por questionário. A amostra foi constituída pelos responsáveis pela auditoria interna e pelos responsáveis do órgão executivo dos 278 municípios existentes no continente. Neste estudo foi realizada uma análise descritiva e inferencial de forma a verificar as relações existentes entre as variáveis em análise. Os resultados obtidos permitem concluir que o grupo dos respondentes têm o mesmo entendimento quanto ao impacto e à importância da atividade de auditoria interna na gestão municipal. Apesar de algumas das variáveis independentes (solicitação, avaliação e o grau de importância) não estarem diretamente associadas com a influência das informações veiculadas pela atividade de auditoria interna no processo de tomada de decisão pelos órgãos executivos, são reconhecidas vantagens à auditoria das contas municipais quer pelos executivos como pelos demais utilizadores da informação produzidas pelos auditores internos.En los últimos tiempos, la investigación en gestión pública ha evolucionado significativamente. Sin embargo, a pesar de esta evolución son todavía varias las cuestiones que permanecen sin respuesta en la investigación en Administración Pública. El estudio del impacto y de la importancia de la actividad de la auditoría interna en la gestión municipal es un ejemplo de ello. Este estudio tiene como principal objetivo percibir en qué medida, y de qué manera las informaciones transmitidas por la actividad de auditoría interna influyen en la toma de decisión por los órganos ejecutivos. Para la realización de este estudio, se aplicaron dos encuestas por cuestionario. La muestra fue constituida por los responsables de la auditoría interna y por los responsables del órgano ejecutivo de los 278 municipios existentes en el continente. En este estudio se realizó un análisis descriptivo e inferencial para verificar las relaciones existentes entre las variables en análisis. Los resultados obtenidos permiten concluir que el grupo de los encuestados tiene el mismo entendimiento en cuanto al impacto ya la importancia de la actividad de auditoría interna en la gestión municipal. A pesar de que algunas de las variables independientes (solicitud, evaluación y el grado de importancia) no están directamente asociadas con la influencia de las informaciones transmitidas por la actividad de auditoría interna en el proceso de toma de decisión por los órganos ejecutivos, se reconocen ventajas a la auditoría de las cuentas municipales, por los ejecutivos como por los demás usuarios de la información producidos por los auditores internos.In recent times, public management research has evolved significantly. However, despite this evolution are still several questions that remain unanswered in public administration research. The study of the impact and importance of the internal audit activity on municipal management is an example of this. This study has as main objective to understand to what extent, and in what way the information conveyed by the internal audit activity influences in the decision making by the executive organs. For the accomplishment of this study, two questionnaire surveys were applied. The sample consisted of those responsible for the internal audit and the heads of the executive board members of the 278 municipalities on the continent. In this study, a descriptive and inferential analysis was performed to verify the existing relationships between the variables under analysis. The results obtained allow us to conclude that the group of respondents have the same understanding regarding the impact and importance of the internal audit activity in municipal management. Although some of the independent variables (request, evaluation and degree of importance) are not directly associated with the influence of the information conveyed by the internal audit activity in the decision-making process by the executive board members, the audit of municipal accounts is recognized executives and other users of information produced by internal auditors

Pemodelan dan Simulasi Sistem Proteksi Mikrogrid

Mikrogrid mengintegrasi sumber energi terdistribusi untuk menyediakan sistem yang handal dan ramah lingkungan. Akibat adanya generator dan PV pada semua level sistem mikrogrid dan 3 konfigurasi sistem yang berbeda, yaitu hanya terhubung grid, terhubung grid dan mikrogrid dan islanding, arus gangguan pada sistem akan bervariasi (arus gangguan naik 8.2 kA ketika sistem terhubung dengan grid). Dengan koordinasi rele dan LVCB yang benar, maka sistem proteksi dapat mengamankan pembangkit dan beban. Sehingga dapat menyediakan keandalan sistem yang lebih tinggi. Sistem proteksi yang didesain dimaksudkan untuk dapat bekerja dalam 3 konfigurasi sistem yang berbeda akibat adanya grid dan sumber energy terdistribusi. Sistem proteksi yang digunakan pada tugas akhir ini dilengkapi dengan Directional Over Current Relay yang diuji menggunakan ETAP pada sebuah sistem mikrogrid yang terdiri dari PV dan biomassa. Aplicativo multibase para tablets: análise de uma de suas funcionalidades

O trabalho, apresentado neste artigo, tenta colaborar com a inserção de tablets no contexto escolar, ao trazer a análise do uso de um aplicativo baseado no material Dourado, criado pela educadora italiana Maria Montessori, para auxiliar na aprendizagem de conceitos de bases numéricas e operações aritméticas, por meio de manipulação. É apresentada a descrição da primeira versão do aplicativo e analisada uma de suas funcionalidades, ao ser utilizado por um grupo de estudantes do 6o ano do ensino fundamental. Essa análise está pautada em bases teóricas relacionadas à compreensão dos conceitos matemáticos envolvidos e ao uso de dispositivos touchscreen em Educação Matemática. A experiência apontou algumas potencialidades do aplicativo, entre elas a perspectiva de realização de ações gestuais conscientes pelos estudantes, que foram devidamente registradas e são acessíveis ao professor, contribuindo para a construção de significados matemáticos
