Realist Review: Current Practice and Future Prospects
Realist review has emerged as a specific literature review approach that is concerned with explaining the outcomes of complex intervention programs. We undertook a systematic scoping review to examine the current practice of realist review. A systematic scoping review is a process of mapping the existing evidence base on a particular topic. We identified a growing body of literature using the realist review approach. We selected 54 reviews for our study. These reviews covered a range of topics, including health care, education, management, and public safety. We found that the initial process of exploratory scoping of the literature was described in only 58 per cent of the reviews. The approaches regarding appraisal, analysis, and synthesis of the selected studies were poorly described in most reviews. Overall, there was little uniformity and transparency regarding many methodological issues. Specific methodological guidance may need to be developed if realist reviews are to have a more uniform and transparent approach

Testing soil fertility of Prehispanic terraces at Viejo Sangayaico in the upper Ica catchment of south-central highland Peru
This study presents a pilot geoarchaeological investigation of terraced agricultural systems near San Francisco de Sangayaico, in the upper Ica catchment of the Southern Peruvian Andes. It aims to assess the evidence for soil fertility associated with agricultural strategies practiced throughout the Prehispanic, Spanish colonial and modern occupations in this region. A series of twenty-two test pits were hand excavated through two terraced field systems, and sampled to examine the changes in soil physical and chemical characteristics down-profile and downslope.
This study provides the first geoarchaeological analyses of the agrarian soil system surrounding Viejo Sangayaico in the upper Ica catchment. Results demonstrate that the soil system was much modified prior to the creation of the terrace systems, probably about 900 years ago. This system was characterised by a weakly acidic to slightly calcareous pH, a consistent but low electrical conductivity, reasonable-but- variable phosphorus content, and a loamy soil texture with a component of weathered volcanic tonalite parent material. The shallow terrace soil build-up on the slopes investigated indicates that slope modification was as minimal as possible. Moreover, the relatively low frequencies of organic material and phosphorus suggest that the terraces were not heavily fertilised in the past, making the stability and management of the nutrient-rich topsoil vital.
The results of these excavations and soil fertility analyses are situated within the context of the wider Andean ethno-historic and the archaeological record to address questions regarding how the terraces were built and maintained over time. Agricultural terraces undoubtedly mitigated the effects of slope erosion associated with cultivation. But, the terrace soil features observed at Sangayaico do not appear to be the same as those documented in other geoarchaeological studies of Andean terrace systems. These contrasts may be accounted for by a combination of differing geological substrate and hydrological conditions, as well as variable trajectories in past soil development, erosion factors, manuring/field management practices and crop selection.Funding for the field and laboratory work was provided by the Leverhulme Trust, a private donation, and the Department of Anthropology, Washington State University.This is the author accepted manuscript. Differential cargo mobilisation within Weibel-Palade bodies after transient fusion with the plasma membrane.
Inflammatory chemokines can be selectively released from Weibel-Palade bodies (WPBs) during kiss-and-run exocytosis. Such selectivity may arise from molecular size filtering by the fusion pore, however differential intra-WPB cargo re-mobilisation following fusion-induced structural changes within the WPB may also contribute to this process. To determine whether WPB cargo molecules are differentially re-mobilised, we applied FRAP to residual post-fusion WPB structures formed after transient exocytosis in which some or all of the fluorescent cargo was retained. Transient fusion resulted in WPB collapse from a rod to a spheroid shape accompanied by substantial swelling (>2 times by surface area) and membrane mixing between the WPB and plasma membranes. Post-fusion WPBs supported cumulative WPB exocytosis. To quantify diffusion inside rounded organelles we developed a method of FRAP analysis based on image moments. FRAP analysis showed that von Willebrand factor-EGFP (VWF-EGFP) and the VWF-propolypeptide-EGFP (Pro-EGFP) were immobile in post-fusion WPBs. Because Eotaxin-3-EGFP and ssEGFP (small soluble cargo proteins) were largely depleted from post-fusion WPBs, we studied these molecules in cells preincubated in the weak base NH4Cl which caused WPB alkalinisation and rounding similar to that produced by plasma membrane fusion. In these cells we found a dramatic increase in mobilities of Eotaxin-3-EGFP and ssEGFP that exceeded the resolution of our method (∼ 2.4 µm2/s mean). In contrast, the membrane mobilities of EGFP-CD63 and EGFP-Rab27A in post-fusion WPBs were unchanged, while P-selectin-EGFP acquired mobility. Our data suggest that selective re-mobilisation of chemokines during transient fusion contributes to selective chemokine secretion during transient WPB exocytosis. Selective secretion provides a mechanism to regulate intravascular inflammatory processes with reduced risk of thrombosis
Multichannel social signatures and persistent features of ego networks
The structure of egocentric networks reflects the way people balance their need for strong, emotionally intense relationships and a diversity of weaker ties. Egocentric network structure can be quantified with ’social signatures’, which describe how people distribute their communication effort across the members (alters) of their personal networks. Social signatures based on call data have indicated that people mostly communicate with a few close alters; they also have persistent, distinct signatures. To examine if these results hold for other channels of communication, here we compare social signatures built from call and text message data, and develop a way of constructing mixed social signatures using both channels. We observe that all types of signatures display persistent individual differences that remain stable despite the turnover in individual alters. We also show that call, text, and mixed signatures resemble one another both at the population level and at the level of individuals. The consistency of social signatures across individuals for different channels of communication is surprising because the choice of channel appears to be alter-specific with no clear overall pattern, and ego networks constructed from calls and texts overlap only partially in terms of alters. These results demonstrate individuals vary in how they allocate their communication effort across their personal networks and this variation is persistent over time and across different channels of communication
