Distribution of siderophile and other trace elements in melt rock at the Chicxulub impact structure
Recent isotopic and mineralogical studies have demonstrated a temporal and chemical link between the Chicxulub multiring impact basin and ejecta at the Cretaceous-Tertiary boundary. A fundamental problem yet to be resolved, however, is identification of the projectile responsible for this cataclysmic event. Drill core samples of impact melt rock from the Chichxulub structure contain Ir and Os abundances and Re-Os isotopic ratios indicating the presence of up to approx. 3 percent meteoritic material. We have used a technique involving microdrilling and high sensitivity instrumental neutron activation analysis (INAA) in conjunction with electron microprobe analysis to characterize further the distribution of siderophile and other trace elements among phases within the C1-N10 melt rock
Sensitivity of bile acid transport by organic anion-transporting polypeptides to intracellular pH
AbstractWe investigated the influence of intracellular pH (pHi) on [14C]-glycocholate (GC) uptake by human hepatoblastoma HepG2 cells that express sodium-independent (mainly OATP-A and OATP-8), but not sodium-dependent, GC transporters. Replacement of extracellular sodium by choline (Chol) stimulated GC uptake but did not affect GC efflux from loaded cells. Amiloride or NaCl replacement by tetraethylammonium chloride (TeACl) or sucrose also increased GC uptake. All stimulating circumstances decreased pHi. By contrast, adding to the medium ammonium or imidazole, which increased pHi, had no effect on GC uptake. In Chinese hamster ovary (CHO) cells expressing rat Oatp1, acidification of pHi had the opposite effect on GC uptake, that is, this was reduced. Changes in extracellular pH (pHo) between 7.40 and 7.00 had no effect on GC uptake at pHi 7.30 or 7.45 when pHo<pHi. However, GC uptake was inhibited at pHo 7.40 and 7.80 when pHo>pHi. Inhibition was not proportional to the pHo−pHi difference. Intracellular acidification decreased Vmax, but had no effect on Km. In sum, sodium-independent GC transport can be affected by intracellular acidification, possibly due both to modifications in the driving forces and to the particular response to protonation of carrier proteins involved in this process
The AMS-02 RICH Imager Prototype - In-Beam Tests with 20 GeV/c per Nucleon Ions -
A prototype of the AMS Cherenkov imager (RICH) has been tested at CERN by
means of a low intensity 20 GeV/c per nucleon ion beam obtained by
fragmentation of a primary beam of Pb ions. Data have been collected with a
single beam setting, over the range of nuclear charges 2<Z<~45 in various beam
conditions and using different radiators. The charge Z and velocity beta

The AMS-RICH velocity and charge reconstruction
The AMS detector, to be installed on the International Space Station,
includes a Ring Imaging Cerenkov detector with two different radiators, silica
aerogel (n=1.05) and sodium fluoride (n=1.334). This detector is designed to
provide very precise measurements of velocity and electric charge in a wide
range of cosmic nuclei energies and atomic numbers. The detector geometry, in
particular the presence of a reflector for acceptance purposes, leads to
complex Cerenkov patterns detected in a pixelized photomultiplier matrix. The
results of different reconstruction methods applied to test beam data as well
as to simulated samples are presented. To ensure nominal performances
throughout the flight, several detector parameters have to be carefully
monitored. The algorithms developed to fulfill these requirements are
presented. The velocity and charge measurements provided by the RICH detector
endow the AMS spectrometer with precise particle identification capabilities in
a wide energy range. The expected performances on light isotope separation are

The Ring Imaging Cherenkov detector (RICH) of the AMS experiment
The Alpha Magnetic Spectrometer (AMS) experiment to be installed on the
International Space Station (ISS) will be equipped with a proximity focusing
Ring Imaging Cherenkov (RICH) detector for measuring the electric charge and
velocity of the charged cosmic particles. A RICH prototype consisting of 96
photomultiplier units, including a piece of the conical reflector, was built
and its performance evaluated with ion beam data. Preliminary results of the
in-beam tests performed with ion fragments resulting from collisions of a 158
GeV/c/nuc primary beam of Indium ions (CERN SPS) on a Pb target are reported.
The collected data included tests to the final front-end electronics and to
different aerogel radiators. Cherenkov rings for a large range of charged
nuclei and with reflected photons were observed. The data analysis confirms the
design goals. Charge separation up to Fe and velocity resolution of the order

The RICH detector of the AMS-02 experiment: status and physics prospects
The Alpha Magnetic Spectrometer (AMS), whose final version AMS-02 is to be
installed on the International Space Station (ISS) for at least 3 years, is a
detector designed to measure charged cosmic ray spectra with energies up to the
TeV region and with high energy photon detection capability up to a few hundred
GeV. It is equipped with several subsystems, one of which is a proximity
focusing RICH detector with a dual radiator (aerogel+NaF) that provides
reliable measurements for particle velocity and charge. The assembly and
testing of the AMS RICH is currently being finished and the full AMS detector
is expected to be ready by the end of 2008. The RICH detector of AMS-02 is

Modulation of KV4.3-KChIP2 Channels by IQM-266: Role of DPP6 and KCNE2
The transient outward potassium current (Itof) is generated by the activation of KV4 channels assembled with KChIP2 and other accessory subunits (DPP6 and KCNE2). To test the hypothesis that these subunits modify the channel pharmacology, we analyzed the electrophysiological effects of (3-(2-(3-phenoxyphenyl)acetamido)-2-naphthoic acid) (IQM-266), a new KChIP2 ligand, on the currents generated by KV4.3/KChIP2, KV4.3/KChIP2/DPP6 and KV4.3/KChIP2/KCNE2 channels. CHO cells were transiently transfected with cDNAs codifying for different proteins (KV4.3/KChIP2, KV4.3/KChIP2/DPP6 or KV4.3/KChIP2/KCNE2), and the potassium currents were recorded using the whole-cell patch-clamp technique. IQM-266 decreased the maximum peak of KV4.3/KChIP2, KV4.3/KChIP2/DPP6 and KV4.3/KChIP2/KCNE2 currents, slowing their time course of inactivation in a concentration-, voltage-, time- and use-dependent manner. IQM-266 produced an increase in the charge in KV4.3/KChIP2 channels that was intensified when DPP6 was present and abolished in the presence of KCNE2. IQM-266 induced an activation unblocking effect during the application of trains of pulses to cells expressing KV4.3/KChIP2 and KV4.3/KChIP2/KCNE2, but not in KV4.3/KChIP2/DPP6 channels. Overall, all these results are consistent with a preferential IQM-266 binding to an active closed state of Kv4.3/KChIP2 and Kv4.3/KChIP2/KCNE2 channels, whereas in the presence of DPP6, IQM-266 binds preferentially to an inactivated state. Further Characterization of the Electrogenicity and pH Sensitivity of the Human Organic Anion-Transporting Polypeptides OATP1B1 and OATP1B3
Organic anion-transporting polypeptides (OATPs) are involved in the liver uptake of many endogenous and xenobiotic compounds, such as bile acids and drugs, respectively. Using Xenopus laevis oocytes and Chinese hamster ovary (CHO) cells expressing rat Oatp1a1, human OATP1B1, or OATP1B3, the sensitivity of these transporters to extracellular/intracellular pH (pHo/pHi) and changes in plasma membrane potential (ΔΨ) was investigated. In X. laevis oocytes, nonspecific plasma membrane permeability increased only at pHo below 4.5. Above this value, both using oocytes and CHO cells, extracellular acidification affected differently the specific transport of taurocholic acid (TCA) and estradiol 17β-d-glucuronide (E217βG) by Oatp1a1 (stimulation), OATP1B1 (inhibition), and OATP1B3 (stimulation). Changes in substrate uptake in the presence of valinomycin (K+-ionophore), carbonyl cyanide 3-chlorophenylhydrazone and nigericin (protonophores), and amiloride (Na+/H+-inhibitor) and cation replacement in the medium were studied with fluorescent probes for measuring substrate uptake (cholylglycyl amidofluorescein) and changes in pHi (SNARF-4F) and ΔΨ [DilC1(5)]. The results suggest that activity of these three carriers is sodium/potassium-independent and affected differently by changes in pHo and ΔΨ: Oatp1a1 was confirmed to be an electroneutral anion exchanger, whereas the function of both OATP1B1 and OATP1B3 was markedly affected by the magnitude of ΔΨ. Moreover, electrophysiological measurements revealed the existence of a net anion influx associated to OATP1B1/OATP1B3-mediated transport of TCA, E217βG, and estrone-3-sulfate. Furthermore, a leakage of Na+ through OATP1B1 and OATP1B3, which is not coupled to substrate transport, was found. In conclusion, these results suggest that OATP1B1 and OATP1B3 are electrogenic transporters whose activity may be strongly affected under circumstances of displacement of local pH
