    Metaphoric coherence: Distinguishing verbal metaphor from `anomaly\u27

    Theories and computational models of metaphor comprehension generally circumvent the question of metaphor versus “anomaly” in favor of a treatment of metaphor versus literal language. Making the distinction between metaphoric and “anomalous” expressions is subject to wide variation in judgment, yet humans agree that some potentially metaphoric expressions are much more comprehensible than others. In the context of a program which interprets simple isolated sentences that are potential instances of cross‐modal and other verbal metaphor, I consider some possible coherence criteria which must be satisfied for an expression to be “conceivable” metaphorically. Metaphoric constraints on object nominals are represented as abstracted or extended along with the invariant structural components of the verb meaning in a metaphor. This approach distinguishes what is preserved in metaphoric extension from that which is “violated”, thus referring to both “similarity” and “dissimilarity” views of metaphor. The role and potential limits of represented abstracted properties and constraints is discussed as they relate to the recognition of incoherent semantic combinations and the rejection or adjustment of metaphoric interpretations

    Currently, there exists a relatively large number of guidelines and recommendations which deal with the fatigue assessment of welded structures. In these guidelines, different approaches are proposed to assess the service life of welded structures under multi-axial loading conditions. Aim of this paper is the evaluation of the quality of these approaches. Experimental fatigue data using multiaxial loading conditions from the literature were collected establishing a target data set. An evaluation of the data was performed using three guidelines (IIW-recommendations, FKM guideline and Eurocode 3) as well as a critical plane approach. All evaluations have been performed for the nominal, the structural and the effective notch stress approach

    Reference genome-independent assessment of mutation density using restriction enzyme-phased sequencing

    <p>Abstract</p> <p>Background</p> <p>The availability of low cost sequencing has spurred its application to discovery and typing of variation, including variation induced by mutagenesis. Mutation discovery is challenging as it requires a substantial amount of sequencing and analysis to detect very rare changes and distinguish them from noise. Also challenging are the cases when the organism of interest has not been sequenced or is highly divergent from the reference.</p> <p>Results</p> <p>We describe the development of a simple method for reduced representation sequencing. Input DNA was digested with a single restriction enzyme and ligated to Y adapters modified to contain a sequence barcode and to provide a compatible overhang for ligation. We demonstrated the efficiency of this method at SNP discovery using rice and arabidopsis. To test its suitability for the discovery of very rare SNP, one control and three mutagenized rice individuals (1, 5 and 10 mM sodium azide) were used to prepare genomic libraries for Illumina sequencers by ligating barcoded adapters to <it>NlaIII </it>restriction sites. For genome-dependent discovery 15-30 million of 80 base reads per individual were aligned to the reference sequence achieving individual sequencing coverage from 7 to 15×. We identified high-confidence base changes by comparing sequences across individuals and identified instances consistent with mutations, i.e. changes that were found in a single treated individual and were solely GC to AT transitions. For genome-independent discovery 70-mers were extracted from the sequence of the control individual and single-copy sequence was identified by comparing the 70-mers across samples to evaluate copy number and variation. This <it>de novo </it>"genome" was used to align the reads and identify mutations as above. Covering approximately 1/5 of the 380 Mb genome of rice we detected mutation densities ranging from 0.6 to 4 per Mb of diploid DNA depending on the mutagenic treatment.</p> <p>Conclusions</p> <p>The combination of a simple and cost-effective library construction method, with Illumina sequencing, and the use of a bioinformatic pipeline allows practical SNP discovery regardless of whether a genomic reference is available.</p

    The Destruction of Tori in Volume-Preserving Maps

    Invariant tori are prominent features of symplectic and volume preserving maps. From the point of view of chaotic transport the most relevant tori are those that are barriers, and thus have codimension one. For an nn-dimensional volume-preserving map, such tori are prevalent when the map is nearly "integrable," in the sense of having one action and n1n-1 angle variables. As the map is perturbed, numerical studies show that the originally connected image of the frequency map acquires gaps due to resonances and domains of nonconvergence due to chaos. We present examples of a three-dimensional, generalized standard map for which there is a critical perturbation size, ϵc\epsilon_c, above which there are no tori. Numerical investigations to find the "last invariant torus" reveal some similarities to the behavior found by Greene near a critical invariant circle for area preserving maps: the crossing time through the newly destroyed torus appears to have a power law singularity at ϵc\epsilon_c, and the local phase space near the critical torus contains many high-order resonances.Comment: laTeX, 16 figure

    An epigenetic blockade of cognitive functions in the neurodegenerating brain

    Cognitive decline is a debilitating feature of most neurodegenerative diseases of the central nervous system, including Alzheimer’s disease [superscript 1]. The causes leading to such impairment are only poorly understood and effective treatments are slow to emerge [superscript 2]. Here we show that cognitive capacities in the neurodegenerating brain are constrained by an epigenetic blockade of gene transcription that is potentially reversible. This blockade is mediated by histone deacetylase 2, which is increased by Alzheimer’s-disease-related neurotoxic insults in vitro, in two mouse models of neurodegeneration and in patients with Alzheimer’s disease. Histone deacetylase 2 associates with and reduces the histone acetylation of genes important for learning and memory, which show a concomitant decrease in expression. Importantly, reversing the build-up of histone deacetylase 2 by short-hairpin-RNA-mediated knockdown unlocks the repression of these genes, reinstates structural and synaptic plasticity, and abolishes neurodegeneration-associated memory impairments.
