    A serological investigation of caseous lymphadenitis in four flocks of sheep

    A double antibody sandwich ELISA developed by ID-DLO, Lelystad to detect Corynebocterium pseudotuberculosis infection was used on 329 sheep from four pedigree Suffolk flocks in which clinical cases of caseous lymphadenitis (CLA) had occurred. At subsequent necropsy, typical CLA lesions were seen in 133 sheep, and the diagnosis was confirmed on culture. Lesions were most commonly seen in lungs (n = 46), parotid lymph nodes (n = 44), prescapular lymph nodes (n = 38) and mediastinal lymph nodes (n = 31). The sensitivity of the ELISA test for detecting culture-positive sheep was 0.88, while the specificity of the test was 0.55. The antibody ELISA detected 87.5 per cent of sheep that had CLA lesions restricted to internal organs only. It was concluded that the ELISA test has a valuable role in detecting sheep with both clinical and subclinical CLA

    Comparison of the effects of E coli STa with E coli LT, Clostridium difficile toxin A and osmotic burdens on small intestinal fluid transport: additional proof that STa is not a secretory enterotoxin

    Using a recirculation procedure to perfuse anaesthetised rat jejunum, E. coli STa enterotoxin can be shown to inhibit net fluid absorption profoundly, while not causing net fluid secretion, provided fluid measurement is by mass or volume. This observation contradicts many reports of STa causing secretion, implying that the recovered volume technique in the anaesthetised animal over a period of some hours cannot detect secretion because of conjectured or unspecified flaws. Experiments are presented here confirming the viability of the perfusion protocol used in this laboratory but also demonstrate that if secretion were to be occurring, the recovered volume protocol would detect it. It will only return a negative finding, if secretion does not occur. To this end, the effect of two secretory toxins on intestinal fluid movement in a closed loop preparation were studied to demonstrate that the anaesthetic, intestinal preparation or perfusion duration did not hinder the demonstration of net secretion when the intestine was exposed to E. coli LT and C. difficile toxin A.. It is evident that STa itself only reduces net absorption but can appear to be secretory if driving forces such as luminal osmotic pressure or capillary hydrostatic pressure through vasodilatation are introduced, as was likely to have occurred with pithing and theophylline. The recognition that STa is a non-secretory enterotoxin necessarily falsifies several alternative methods that claim to demonstrate secretion. Since STa is not secretory many other substances identified by these methods need also not be secretory and alternative explanations must be found to explain their action. The importance of recognising that action on the small intestine cannot be attributed to a secretory mechanism within the enterocyte adds further weight to the concept that where net secretion does occur, the likely mechanism for it is a combination of increased vasodilatation together with increased hydraulic conductivity

    The recent assertion of [1.] that Fe-EDTA does not catalyze superoxide dismutation is disputed. By directly observing superoxide generated during pulse radiolysis, we have confirmed the results of a previous study ( [2.]) which concluded that Fe-EDTA catalyzed superoxide dismutation. We also demonstrate that the reaction of Fe(II)-EDTA, formed during catalyzed superoxide dismutation, with cytochrome c, the probe molecule in the cytochrome c/xanthine oxidase/xanthine assay system for superoxide dismutase activity, is sufficiently rapid ([12.]) to obscure the weak catalysis of superoxide dismutation by Fe-EDTA.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/24002/1/0000251.pd

    Fast, scalable, Bayesian spike identification for multi-electrode arrays

    We present an algorithm to identify individual neural spikes observed on high-density multi-electrode arrays (MEAs). Our method can distinguish large numbers of distinct neural units, even when spikes overlap, and accounts for intrinsic variability of spikes from each unit. As MEAs grow larger, it is important to find spike-identification methods that are scalable, that is, the computational cost of spike fitting should scale well with the number of units observed. Our algorithm accomplishes this goal, and is fast, because it exploits the spatial locality of each unit and the basic biophysics of extracellular signal propagation. Human intervention is minimized and streamlined via a graphical interface. We illustrate our method on data from a mammalian retina preparation and document its performance on simulated data consisting of spikes added to experimentally measured background noise.     A constraint on antigravity of antimatter from precision spectroscopy of simple atoms

    Consideration of antigravity for antiparticles is an attractive target for various experimental projects. There are a number of theoretical arguments against it but it is not quite clear what kind of experimental data and theoretical suggestions are involved. In this paper we present straightforward arguments against a possibility of antigravity based on a few simple theoretical suggestions and some experimental data. The data are: astrophysical data on rotation of the Solar System in respect to the center of our galaxy and precision spectroscopy data on hydrogen and positronium. The theoretical suggestions for the case of absence of the gravitational field are: equality of electron and positron mass and equality of proton and positron charge. We also assume that QED is correct at the level of accuracy where it is clearly confirmed experimentally

    Control of primary productivity and the significance of photosynthetic bacteria in a meromictic kettle lake.

    During 1986 planktonic primary production and controlling factors were investigated in a small (A0 = 11.8 · 103 m2, Zmax = 11.5 m) meromictic kettle lake (Mittlerer Buchensee). Annual phytoplankton productivity was estimated to ca 120 gC · m–2 · a–1 (1,42 tC · lake–1 · a–1). The marked thermal stratification of the lake led to irregular vertical distributions of chlorophylla concentrations (Chla) and, to a minor extent, of photosynthesis (Az). Between the depths of 0 to 6 m low Chla concentrations (< 7 mg · m–3) and comparatively high background light attenuation (kw = 0,525 m–1, 77% of total attenuation due to gelbstoff and abioseston) was found. As a consequence, light absorption by algae was low (mean value 17,4%) and self-shading was absent. Because of the small seasonal variation of Chla concentrations, no significant correlation between Chla and areal photosynthesis (A) was observed. Only in early summer (June–July) biomass appears to influence the vertical distribution of photosynthesis on a bigger scale. Around 8 m depth, low-light adapted algae and phototrophic bacteria formed dense layers. Due to low ambient irradiances, the contribution of these organisms to total primary productivity was small. Primary production and incident irradiance were significantly correlated with each other (r2 = 0.68). Although the maximum assimilation number (Popt) showed a clear dependence upon water temperature (Q10 = 2.31), the latter was of minor importance to areal photosynthesis

    Evaluation of Biopolymer Materials and Synthesis Techniques to Develop a Rod-Shaped Biopolymer Surrogate for Legionella pneumophila

    Biopolymer microparticles have been developed for applications that require biocompatibility and biodegradability, such as drug delivery. In this study, we assessed the production of microparticles using carnauba wax, κ-carrageenan, alginate, and poly (lactic-co-glycolic acid) (PLGA) with the aim of developing a novel, DNA-tracer-loaded, biopolymer surrogate with a size, shape, surface charge, and relative hydrophobicity similar to stationary-phase Legionella pneumophila to mimic the bacteria’s mobility and persistence in engineered water systems. We found that the type and concentration of biopolymer, reaction conditions, and synthesis methods affected the morphology, surface charge, relative hydrophobicity, and DNA tracer loading efficiency of the biopolymer microparticles produced. Carnauba wax, κ-carrageenan, and alginate (Protanal®, and low and medium viscosity) produced highly polydisperse microspheres. In contrast, PLGA and alginate-CaCO3 produced uniform microspheres and rod-shaped microparticles, respectively, with high DNA tracer loading efficiencies (PLGA 70% and alginate-CaCO3 95.2 ± 5.7%) and high reproducibilities. Their synthesis reproducibility was relatively high. The relative hydrophobicity of PLGA microspheres closely matched the cell surface hydrophobicity of L. pneumophila but not the bacterial morphology, whereas the polyelectrolyte layer-by-layer assembly was required to enhance the relative hydrophobicity of alginate-CaCO3 microparticles. Following this surface modification, alginate-CaCO3 microparticles represented the best match to L. pneumophila in size, morphology, surface charge, and relative hydrophobicity. This new biopolymer surrogate has the potential to be used as a mimic to study the mobility and persistence of L. pneumophila in water systems where the use of the pathogen is impractical and unsafe

    Perfectionism and self-conscious emotions in British and Japanese students: Predicting pride and embarrassment after success and failure

    Regarding self-conscious emotions, studies have shown that different forms of perfectionism show different relationships with pride, shame, and embarrassment depending on success and failure. What is unknown is whether these relationships also show cultural variations. Therefore, we conducted a study investigating how self-oriented and socially prescribed perfectionism predicted pride and embarrassment after success and failure comparing 363 British and 352 Japanese students. Students were asked to respond to a set of scenarios where they imagined achieving either perfect (success) or flawed results (failure). In both British and Japanese students, self-oriented perfectionism positively predicted pride after success and embarrassment after failure whereas socially prescribed perfectionism predicted embarrassment after success and failure. Moreover, in Japanese students, socially prescribed perfectionism positively predicted pride after success and self-oriented perfectionism negatively predicted pride after failure. The findings have implications for our understanding of perfectionism indicating that the perfectionism–pride relationship not only varies between perfectionism dimensions, but may also show cultural variations
