    Effects of multiple-dose ponesimod, a selective SIP1 receptor modulator, on lymphocyte subsets in healthy humans

    This study investigated the effects of ponesimod, a selective SIP1 receptor modulator, on T lymphocyte subsets in 16 healthy subjects. Lymphocyte subset proportions and absolute numbers were determined at baseline and on Day 10, after once-daily administration of ponesimod (10 mg, 20 mg, and 40 mg each consecutively for 3 days) or placebo (ratio 3: 1). The overall change from baseline in lymphocyte count was -1,292 +/- 340x10(6) cells/L and 275 +/- 486x10(6) cells/L in ponesimod- and placebo-treated subjects, respectively. This included a decrease in both T and B lymphocytes following ponesimod treatment. A decrease in naive CD4(+) T cells (CD45RA(+)CCR7(+)) from baseline was observed only after ponesimod treatment (-113 +/- 98x10(6) cells/L, placebo: 0 +/- 18x10(6) cells/L). The number of T-cytotoxic (CD3(+)CD8(+)) and T-helper (CD3(+)CD4(+)) cells was significantly altered following ponesimod treatment compared with placebo. Furthermore, ponesimod treatment resulted in marked decreases in CD4(+) T-central memory (CD45RA(-)CCR7(+)) cells (-437 +/- 164x10(6) cells/L) and CD4(+) T-effector memory (CD45RA(-)CCR7(-)) cells (-131 +/- 57x10(6) cells/L). In addition, ponesimod treatment led to a decrease of -228 +/- 90x10(6) cells/L of gut-homing T cells (CLA(-)integrin beta 7(+)). In contrast, when compared with placebo, CD8(+) T-effector memory and natural killer (NK) cells were not significantly reduced following multiple-dose administration of ponesimod. In summary, ponesimod treatment led to a marked reduction in overall T and B cells. Further investigations revealed that the number of CD4(+) cells was dramatically reduced, whereas CD8(+) and NK cells were less affected, allowing the body to preserve critical viral-clearing functions

    Quantifying the interplay of conversational devices in building mutual understanding

    Humans readily engage in idle chat and heated discussions and negotiate tough joint decisions without ever having to think twice about how to keep the conversation grounded in mutual understanding. However, current attempts at identifying and assessing the conversational devices that make this possible are fragmented across disciplines and investigate single devices within single contexts. We present a comprehensive conceptual framework to investigate conversational devices, their relations, and how they adjust to contextual demands. In two corpus studies, we systematically test the role of three conversational devices: backchannels, repair, and linguistic entrainment. Contrasting affiliative and task-oriented conversations within participants, we find that conversational devices adaptively adjust to the increased need for precision in the latter: We show that low-precision devices such as backchannels are more frequent in affiliative conversations, whereas more costly but higher-precision mechanisms, such as specific repairs, are more frequent in task-oriented conversations. Further, task-oriented conversations involve higher complementarity of contributions in terms of the content and perspective: lower semantic entrainment and less frequent (but richer) lexical and syntactic entrainment. Finally, we show that the observed variations in the use of conversational devices are potentially adaptive: pairs of interlocutors that show stronger linguistic complementarity perform better across the two tasks. By combining motivated comparisons of several conversational contexts and theoretically informed computational analyses of empirical data the present work lays the foundations for a comprehensive conceptual framework for understanding the use of conversational devices in dialogue

    Genetic Correlates of Individual Differences in Sleep Behavior of Free-Living Great Tits (Parus major)

    Within populations, free-living birds display considerable variation in observable sleep behaviors, reflecting dynamic interactions between individuals and their environment. Genes are expected to contribute to repeatable between-individual differences in sleep behaviors, which may be associated with individual fitness. We identified and genotyped polymorphisms in nine candidate genes for sleep, and measured five repeatable sleep behaviors in free-living great tits (Parus major), partly replicating a previous study in blue tits (Cyanistes caeruleus). Microsatellites in the CLOCK and NPAS2 clock genes exhibited an association with sleep duration relative to night length, and morning latency to exit the nest box, respectively. Furthermore, microsatellites in the NPSR1 and PCSK2 genes associated with relative sleep duration and proportion of time spent awake at night, respectively. Given the detection rate of associations in the same models run with random markers instead of candidate genes, we expected two associations to arise by chance. The detection of four associations between candidate genes and sleep, however, suggests that clock genes, a clock-related gene, or a gene involved in the melanocortin system, could play key roles in maintaining phenotypic variation in sleep behavior in avian populations. Knowledge of the genetic architecture underlying sleep behavior in the wild is important because it will enable ecologists to assess the evolution of sleep in response to selection

    Individual variation in age‐dependent reproduction: Fast explorers live fast but senesce young?

    Adaptive integration of life history and behaviour is expected to result in variation in the pace‐of‐life. Previous work focused on whether ‘risky’ phenotypes live fast but die young, but reported conflicting support. We posit that individuals exhibiting risky phenotypes may alternatively invest heavily in early‐life reproduction but consequently suffer greater reproductive senescence. We used a 7‐year longitudinal dataset with >1,200 breeding records of >800 female great tits assayed annually for exploratory behaviour to test whether within‐individual age dependency of reproduction varied with exploratory behaviour. We controlled for biasing effects of selective (dis)appearance and within‐individual behavioural plasticity. Slower and faster explorers produced moderate‐sized clutches when young; faster explorers subsequently showed an increase in clutch size that diminished with age (with moderate support for declines when old), whereas slower explorers produced moderate‐sized clutches throughout their lives. There was some evidence that the same pattern characterized annual fledgling success, if so, unpredictable environmental effects diluted personality‐related differences in this downstream reproductive trait. Support for age‐related selective appearance was apparent, but only when failing to appreciate within‐individual plasticity in reproduction and behaviour. Our study identifies within‐individual age‐dependent reproduction, and reproductive senescence, as key components of life‐history strategies that vary between individuals differing in risky behaviour. Future research should thus incorporate age‐dependent reproduction in pace‐of‐life studies

    Conversation analysis (CA) is an approach to the study of language and social interaction that puts at center stage its sequential development. The chain of initiating and responding actions that characterizes any interaction is a source of internal evidence for the meaning of social behavior as it exposes the understandings that participants themselves give of what one another is doing. Such an analysis requires the close and repeated inspection of audio and video recordings of naturally occurring interaction, supported by transcripts and other forms of annotation. Distributional regularities are complemented by a demonstration of participants' orientation to deviant behavior. CA has long maintained a constructive dialogue and reciprocal influence with linguistic anthropology. This includes a recent convergence on the cross-linguistic and cross-cultural study of social interaction

    Recent models for adaptive personality differences: a review

    In this paper we review recent models that provide adaptive explanations for animal personalities: individual differences in behaviour (or suites of correlated behaviours) that are consistent over time or contexts. We start by briefly discussing patterns of variation in behaviour that have been documented in natural populations. In the main part of the paper we discuss models for personality differences that (i) explain animal personalities as adaptive behavioural responses to differences in state, (ii) investigate how feedbacks between state and behaviour can stabilize initial differences among individuals and (iii) provide adaptive explanations for animal personalities that are not based on state differences. Throughout, we focus on two basic questions. First, what is the basic conceptual idea underlying the model? Second, what are the key assumptions and predictions of the model? We conclude by discussing empirical features of personalities that have not yet been addressed by formal modelling. While this paper is primarily intended to guide empiricists through current adaptive theory, thereby stimulating empirical tests of these models, we hope it also inspires theoreticians to address aspects of personalities that have received little attention up to now
