    Evidence for Placental HPV Infection in Both HIV Positive and Negative Women

    Human papillomaviruses (HPVs) have previously been reported to infect epithelial trophoblast cells of the placenta. To investigate this possibility, 200 placental samples from Zambian women were separated into HIV+ and HIV− groups and tested for HPV by redundant primer PCR, using GP5+/GP6+ and CPI/CPII primer sets. Three HPV genotypes (HPV6, 16 and 90) were detected in placental samples. Whereas, 20 different HPV genotypes were detected in vaginal sampling of the same patients, suggesting that compartment specific sub-populations of HPV may exist. The incidence of HPV16 in placental samples was almost 2-fold greater in HIV+ women compared to HIV− (p = 0.0241). HPV16 L1 expression, detected by immunochemistry, was significantly higher in HIV+ than HIV− samples (p = 0.0231). HPV16 DNA was detected in the nuclei of trophoblast cells by in situ hybridization. Overall, these results suggest that HPVs infect the placenta and that HIV significantly influences these infections

    Streptococcus pneumoniae (the 'pneumococcus') is a significant cause of morbidity and mortality worldwide, causing life-threatening diseases such as pneumonia, bacteraemia, and meningitis, with an annual death burden of over one million. Discovered over a century ago, pneumococcal serotype 1 (S1) is a significant cause of these life-threatening diseases. Our understanding of the epidemiology and biology of pneumococcal S1 has significantly improved over the past two decades, informing the development of preventative and surveillance strategies. However, many questions remain unanswered. Here, we review the current state of knowledge of pneumococcal S1, with a special emphasis on clinical epidemiology, genomics, and disease mechanisms

    The effects of metakaolinization and fused-metakaolinization on zeolites from quartz rich natural clays

    A variety of zeolites have been synthesized by performing either metakaolinization or fused-metakaolinization of natural clays prior to the hydrothermal synthesis step. To understand the differences arising from performing fused metakaolinization rather than simple metakaolinization, the calcination conditions, gel composition, gelling and crystallization conditions were kept similar for both methods. The original clay material, a kaolinite group mineral with Si/Al molar ratio of 1, had a high content of quartz as impurity. The two treatment methods gave different zeolites of high crystallinity and relatively similar Si/Al molar ratio. Zeolite Na-A with 98% crystallinity was produced from the metakaolinization method within 8 h, while Na-X with 96% crystallinity was generated from the fused-metakaolinization (fusion) method within 48 h. In the fusion method, sodalite was formed at or beyond 48 h of hydrothermal reaction, while in the metakaolinite method, apart from a slight decrease in intensities of XRD peaks, no phase change was observed even after 168 h of hydrothermal reaction. Modification of the synthesis gel by increasing the Si/Al molar ratio via addition of fumed silica resulted in mixed zeolite products via the metakaolinite method, while Na–Y was the major product for the fusion method. Except for Na-A, all zeolite products had high surface area of up to 600 m2/g and micropore volume of 0.18 cm3/g. Our findings demonstrate the crucial role of the pre-treatment step in the synthesis of zeolites from clay minerals, and that a variety of high quality zeolites are achievable via choice of pre-treatment protocols

    Comparative catching efficiency of traditional prawn fishing gears in Pulicat lake of Tamil Nadu, India

    303-310Prawn fishery in Pulicat Lake has a significant role in livelihood of the fishers, which is being harvested through different fishing gears. Here, we investigated the prawn-fishing gear and their catch composition. The information on various aspect about the gears was collected from the 48 respondents each month through pre-designed interview schedule by adopting random sampling. Results revealed that the quantity of prawn were higher in stake net (209.83 kg), followed by barriers (118.58 kg), drive-in-net (55.58 kg) tangle net (18.25 kg) and was statically significant at 5 %. It was estimated that more than half (52.16 %) of the total prawn catch in Pulicat Lake was obtained through stake net than the barriers (29.48 %), drive-in-net (13.82 %) and tangle net (4.54 %). The maximum quantity of prawn was obtained during the November and December in all the prawn-fishing gear (p < 0.05). This study concludes that non-selective fishing gears resulted in the abundant catch of juvenile fishes and crabs, need to be regulated mesh size, to support the conservation and sustainable harvest of the fishery resources in Pulicat Lake

    Biodiversity of Freshwater Shrimp of the Genus Macrobrachium (Decapoda Palaemonidae) in the Nyong Basin of Cameroon

    The population of shrimps, especially the macrobrachium genus, has been affected with habitat pollution and overexploitation. Shrimps provide a major protein source to animals higher in the food chain. The importance of studying the ecology of shrimps in their different habitats helps in their preservation and provides possibilities of carcinoculture. This study examines diversity, ecology and reproduction parameters of fresh water shrimps of the genus macrobrachium in the basins of Lepmassoun and Ondoamedza rivers. Lepmassoun and Ondoamedza rivers form part of the larger Nyong river basin in Cameroon. In situ water physicochemical parameters were determined on site, preserved water samples used in mineral analysis and subsequent ecological health assessment. Shrimps were sampled using the scoop method with hand nets and analyzed. Sampling was done in 5 seasons distributed from January 2017 to February 2018. The results of physicochemical parameters indicate that water in the two rivers was slightly mineralized, sufficiently oxygenated and slightly turbid. It was generally a non-polluted ecological zone for shrimps. A total of 113 shrimps distributed among 14 species were collected. The shrimp population included Macrobrachium macrobrachion Macrobrachium idae, Macrobrachium rude, Macrobrachium niloticus , Macrobrachium dux Macrobrachium sp1, Macrobrachium sp2, Macrobrachium sp3,Macrobrachium sp4, Macrobrachium sp4, Macrobrachium sp5, Macrobrachium sp6, Macrobrachium sp7, Macrobrachium sp8 and Macrobrachium sp9.In the river Odoameza 11 species were collected among which Macrobrachium sp8 (26.02%) is abundant, though,in the river Lepmassoun we collected 10 species dominanced by Macrobrachium macrobrachion (22,5%). The species Macrobrachium macrobrachion is most ubiquiste throughout river Lepmassoun (%O = 83.33%) The river basin of Lepmassoun had a higher population of Macrobrachium macrobrachion with a percentage occurrence of 66.67%. then It is concluded that the water in the two river basins is of good ecological quality for shrimp survival with species Macrobrachium macrobrachion dominating in the river lepmassoun. In the river Ondoameza Macrobrachium sp8, Macrobrachium idea, Macrobrachium macrobrachion,Macrobrachium sp4 having all the percentage of occurrence (%O) of 66.67% are relatively represented. Only Macrobrachium dux have Ovigerous females in our study. This Ovigerous females ranged from 50.38 to 64.18 mm and egg size varied from 1.7 to 2 mm. The prawn attained a maximum total length and weight of 64,18 m and 2,96 g respectively. In our basins slope the Macrobrachium dux reproduces twice year during the big dry season and small dry season since production is early and the eggs are rather broad by consequence the larval development is shortened

    'A bite before bed': exposure to malaria vectors outside the times of net use in the highlands of western Kenya.

    BACKGROUND: The human population in the highlands of Nyanza Province, western Kenya, is subject to sporadic epidemics of Plasmodium falciparum. Indoor residual spraying (IRS) and long-lasting insecticide treated nets (LLINs) are used widely in this area. These interventions are most effective when Anopheles rest and feed indoors and when biting occurs at times when individuals use LLINs. It is therefore important to test the current assumption of vector feeding preferences, and late night feeding times, in order to estimate the extent to which LLINs protect the inhabitants from vector bites. METHODS: Mosquito collections were made for six consecutive nights each month between June 2011 and May 2012. CDC light-traps were set next to occupied LLINs inside and outside randomly selected houses and emptied hourly. The net usage of residents, their hours of house entry and exit and times of sleeping were recorded and the individual hourly exposure to vectors indoors and outdoors was calculated. Using these data, the true protective efficacy of nets (P*), for this population was estimated, and compared between genders, age groups and from month to month. RESULTS: Primary vector species (Anopheles funestus s.l. and Anopheles arabiensis) were more likely to feed indoors but the secondary vector Anopheles coustani demonstrated exophagic behaviour (p < 0.05). A rise in vector biting activity was recorded at 19:30 outdoors and 18:30 indoors. Individuals using LLINs experienced a moderate reduction in their overall exposure to malaria vectors from 1.3 to 0.47 bites per night. The P* for the population over the study period was calculated as 51% and varied significantly with age and season (p < 0.01). CONCLUSIONS: In the present study, LLINs offered the local population partial protection against malaria vector bites. It is likely that P* would be estimated to be greater if the overall suppression of the local vector population due to widespread community net use could be taken into account. However, the overlap of early biting habit of vectors and human activity in this region indicates that additional methods of vector control are required to limit transmission. Regular surveillance of both vector behaviour and domestic human-behaviour patterns would assist the planning of future control interventions in this region

    Accelerated SARS-CoV-2 Intrahost Evolution Leading to Distinct Genotypes During Chronic Infection

    The chronic infection hypothesis for novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variant emergence is increasingly gaining credence following the appearance of Omicron. Here, we investigate intrahost evolution and genetic diversity of lineage B.1.517 during a SARS-CoV-2 chronic infection lasting for 471 days (and still ongoing) with consistently recovered infectious virus and high viral genome copies. During the infection, we find an accelerated virus evolutionary rate translating to 35 nucleotide substitutions per year, approximately 2-fold higher than the global SARS-CoV-2 evolutionary rate. This intrahost evolution results in the emergence and persistence of at least three genetically distinct genotypes, suggesting the establishment of spatially structured viral populations continually reseeding different genotypes into the nasopharynx. Finally, we track the temporal dynamics of genetic diversity to identify advantageous mutations and highlight hallmark changes for chronic infection. Our findings demonstrate that untreated chronic infections accelerate SARS-CoV-2 evolution, providing an opportunity for the emergence of genetically divergent variants
