    The Use of a Novel Heart Failure Agent in the Treatment of Pregnancy-Associated Cardiomyopathy

    Peripartum cardiomyopathy is an uncommon, pregnancy-related form of dilated cardiomyopathy that is associated with development of new-onset left ventricular dysfunction. Its etiology is presently unknown, but current standard of care involves the use of typical drug therapy for the treatment of heart failure. Pregnancy-associated cardiomyopathy (PACM) is a similar condition that refers to patients who develop such symptoms prior to the last month of pregnancy. We report the case of a nulliparous Caucasian female who develops early, severe PACM during her first pregnancy with postpartum persistence of New York Heart Association class II-III symptoms despite medical therapy. The use of the novel heart failure agent, sacubitril/valsartan (Entresto), is initiated with near-complete resolution of her symptoms

    Computerized Tomography Detects Pulmonary Lesions in Children with Normal Radiographs Diagnosed to have Tuberculosis

    This report is based on observations during the conduct of a larger study to develop diagnostic criteria for childhood tuberculosis (TB). Of 20l children confirmed to have pulmonary or lymph node TB, 84 had normal chest radiographs. Computerized tomography (CT) of the chest was performed in nine of them, seven of whom had normal chest radiographs while two had visible calcification. Eight of the nine children had definitive lesions detected by computerized tomography of the chest. While five children had primarily hilar lymph node enlargement, three had pulmonary parenchymal lesions. The use of more sensitive diagnostic tests like computed tomography helps to detect tuberculosis lesions not otherwise visualized on chest radiographs. This report highlights the difficulty in excluding active tuberculosis in children. More studies are required on the role of CT scans in the diagnosis of tuberculosis in children

    Incorporating a multiple discrete-continuous outcome in the generalized heterogeneous data model: Application to residential self-selection effects analysis in an activity time-use behavior model

    This paper makes both a methodological contribution as well as an empirical contribution. From a methodological perspective, we propose a new econometric approach for the estimation of joint mixed models that include a multiple discrete choice outcome and a nominal discrete outcome, in addition to the count, binary/ordinal outcomes, and continuous outcomes considered in traditional structural equation models. These outcomes are modeled together by specifying latent underlying unobserved individual lifestyle, personality, and attitudinal factors that impact the many outcomes, and generate the jointness among the outcomes. From an empirical perspective, we analyze residential location choice, household vehicle ownership choice, as well as time-use choices, and investigate the extent of association versus causality in the effects of residential density on activity participation and mobility choices. The sample for the empirical application is drawn from a travel survey conducted in the Puget Sound Region in 2014. The results show that residential density effects on activity participation and motorized auto ownership are both associative as well as causal, emphasizing that accounting for residential self-selection effects are not simply esoteric econometric pursuits, but can have important implications for land-use policy measures that focus on neo-urbanist design

    Antihypertensive and Antioxidant Action of Amlodipine and Vitamin C in Patients of Essential Hypertension

    The etiology of essential hypertension includes increased oxidative stress. The role of antihypertensive drug amlodipine as an antioxidant and the benefit of addition of vitamin C, an antioxidant to antihypertensive therapy were studied. Forty male patients of essential hypertension were randomly divided into two groups and treated with 5 mg amlodipine. In addition one group also received 1000 mg vitamin C (as two 500 mg tablets) once daily for three months. Although blood pressure decreased in both groups, the systolic blood pressure in patients given vitamin C was less (126.4 ± 7.47) compared to the other group (130.9 ± 7.27). A decrease in malondialdehyde, an increase in erythrocyte sodium-potassium adenosine triphosphatase (Na+ K+ ATPase) and an increase in the superoxide dismutase levels were observed in both groups. The increase in SOD was statistically more in the patients given vitamin C in addition to amlodipine (0.1717 ± 0.0150 compared to 0.152 ± 0.0219 units/100 ml assay). In spite of the known antihypertensive, antioxidant activity, similarity in correcting endothelial dysfunction independently, giving the two drugs together and early introduction of vitamin C perhaps decreases oxidative stress and augments the antioxidant status. This may prevent further vascular damage due to oxidative stress, leading to a better prognosis in essential hypertension patients

    An RNA aptamer that interferes with the DNA binding of the HSF transcription activator

    Heat shock factor (HSF) is a conserved and highly potent transcription activator. It is involved in a wide variety of important biological processes including the stress response and specific steps in normal development. Reagents that interfere with HSF function would be useful for both basic studies and practical applications. We selected an RNA aptamer that binds to HSF with high specificity. Deletion analysis defined the minimal binding motif of this aptamer to be two stems and one stem–loop joined by a three-way junction. This RNA aptamer interferes with normal interaction of HSF with its DNA element, which is a key regulatory step for HSF function. The DNA-binding domain plus a flanking linker region on the HSF (DL) is essential for the RNA binding. Additionally, this aptamer inhibits HSF-induced transcription in vitro in the complex milieu of a whole cell extract. In contrast to the previously characterized NF-κB aptamer, the HSF aptamer does not simply mimic DNA binding, but rather binds to HSF in a manner distinct from DNA binding to HSF

    Bioactivity of Locally Available Plants on Cotton Whitefly, Bemisia tabaci and the fungus isolated from cotton

    Aqueous, diethyl ether, chloroform, petroleum ether, N-hexane and benzene extracts of locally available plant species were tested for phytochemical and insecticidal bioactivity against cotton whitefly, Bemisia tabaci, under controlled conditions. This study is within bioprospection context, for utilizing local plant species as alternative in sustainable agriculture development. The leaf and stem extract was used. The whole plant extract of T.procumbens followed by N.oleander and V.rosea showed repellent and toxic effect against adult and second nymphal instars. Leaf extract of all three plants showed high inhibition activity against nymphal instars. In case of flower extract less inhibition activity was shown respectively. Fungi which grow on the cotton plant was screened, characterized and checked for antifungal activity against the extracts of the plant material. Phytochemical analysis was also carried out by standard protocols

    Evidence for Tankyrases as Antineoplastic Targets in Lung Cancer

    Background: New pharmacologic targets are urgently needed to treat or prevent lung cancer, the most common cause of cancer death for men and women. This study identified one such target. This is the canonical Wnt signaling pathway, which is deregulated in cancers, including those lacking adenomatous polyposis coli or β -catenin mutations. Two poly-ADP-ribose polymerase (PARP) enzymes regulate canonical Wnt activity: tankyrase (TNKS) 1 and TNKS2. These enzymes poly-ADP-ribosylate (PARsylate) and destabilize axin, a key component of the β -catenin phosphorylation complex. Methods: This study used comprehensive gene profiles to uncover deregulation of the Wnt pathway in murine transgenic and human lung cancers, relative to normal lung. Antineoplastic consequences of genetic and pharmacologic targeting of TNKS in murine and human lung cancer cell lines were explored, and validated in vivo in mice by implantation of murine transgenic lung cancer cells engineered with reduced TNKS expression relative to controls. Results: Microarray analyses comparing Wnt pathway members in malignant versus normal tissues of a murine transgenic cyclin E lung cancer model revealed deregulation of Wnt pathway components, including TNKS1 and TNKS2. Real-time PCR assays independently confirmed these results in paired normal-malignant murine and human lung tissues. Individual treatments of a panel of human and murine lung cancer cell lines with the TNKS inhibitors XAV939 and IWR-1 dose-dependently repressed cell growth and increased cellular axin 1 and tankyrase levels. These inhibitors also repressed expression of a Wnt-responsive luciferase construct, implicating the Wnt pathway in conferring these antineoplastic effects. Individual or combined knockdown of TNKS1 and TNKS2 with siRNAs or shRNAs reduced lung cancer cell growth, stabilized axin, and repressed tumor formation in murine xenograft and syngeneic lung cancer models. Conclusions: Findings reported here uncovered deregulation of specific components of the Wnt pathway in both human and murine lung cancer models. Repressing TNKS activity through either genetic or pharmacological approaches antagonized canonical Wnt signaling, reduced murine and human lung cancer cell line growth, and decreased tumor formation in mouse models. Taken together, these findings implicate the use of TNKS inhibitors to target the Wnt pathway to combat lung cancer

    ABSTRACT The photocatalytic degradation of methylene blue dye under visible light has been investigated using chromium modified titanium dioxide supported on zeolite (Cr- TiO:lzeolite). The photocatalyst was prepared by sol-gel method and characterized .by X-ray diffraction and SEM. The\u27 rate of photodegradation of dye was monitored spectrophotometrical/y.The effect of pH, dye concentration,amount of photocatalyst and intensity of light on the rate of photocatalytic reaction was observed. The results showed that the use of Cr-doped Ti02 increased the rate of photocatalytic degradation of methylene blue as compared to untreated Ti02.
