    Intermediate temperature SOFC single cell test using Nd1.95NiO4+δ as cathode

    This work deals with SOFC single cell tests using neodymium nickelate Nd1.95NiO4+δ as cathode material. This MIEC oxide exhibits high values of both surface exchange coefficient (k) and oxygen diffusion coefficient (D*), as well as high electronic conductivity, which result in an enhanced electrochemical activity with respect to classical materials. The SOFC cells were fabricated from an anode-supported electrolyte half-cell provided by InDEC B.V. Corporation, with a 36mmdiameter. The Nd1.95NiO4+δ cathode was prepared as a two-stage electrode consisting of a thin interlayer of several hundred nanometers covered by a thicker layer of dozens microns. The Nd1.95NiO4+δ oxide powders were synthesized using different routes in order to reduce the final annealing temperature, and to subsequently obtain submicronic powders. I–V characteristics of the single cells were investigated under hydrogen–air conditions. The power densities versus current densities curves are reported and the results of impedance spectroscopy measurements performed under these operating conditions are discussed

    Electronic Densities of States and Pseudo-gaps in Al-based Complex Intermetallics

    The results of an investigation of the electronic structure of the valence band of various Al-based complex intermetallic alloys, carried out using the X-ray emission spectroscopy technique, are presented and discussed. It is shown that the electronic structure results from an interplay between covalency and repulsive interactions that contributes to the formation of a pseudo-gap at the Fermi level in the Al electronic distributions.</p

    Changes in plant metabolism and accumulation of fungal metabolites in response to Esca proper and apoplexy expression in the whole grapevine

    Trunk diseases have become among the most important grapevine diseases worldwide. They are caused by fungal pathogens that attack the permanent woody structure of the vines and cause various symptoms in woody and annual organs. This study examined modifications of plant responses in green stem, cordon and trunk of grapevines expressing Esca proper (E) or apoplexy (A) event, which are the most frequent grapevine trunk disease symptoms observed in Europe. Transcript expression of a set of plant defense- and stress-related genes was monitored by quantitative RT-PCR while plant phytoalexins and fungal metabolites were quantified by HPLC-MS in order to characterize the interaction between the grapevine and trunk disease agents. Expression of genes encoding enzymes of the phenylpropanoid pathway and trans-resveratrol content were altered in the three organs of diseased plants, especially in the young tissues of A plants. PR proteins and the antioxidant system were severely modulated in A plants, which indicates a drastic stress effect. In the meantime, fungal polyketides 6-MSA, (R)-mellein and (3R,4R)-4-hydroxymellein, were accumulated in A plants that suggests their potential effect on plant metabolism during the appearance of foliar symptoms

    Does a rapid antibiotic susceptibility test optimize antibiotic therapy of patients with bacteremia ?

    editorial reviewedNous avons évalué la contribution d’un antibiogramme rapide réalisé directement à partir d’une hémoculture positive (HP), le dRAST™, dans la prise en charge des patients présentant une bactériémie. Méthodes: Nous avons comparé, rétrospectivement, le délai entre le prélèvement et la disponibilité des résultats d’antibiogramme («temps-pour-résultats», TPR) entre le dRAST™ et l’antibiogramme classique (Vitek®2), auprès de 150 patients présentant une bactériémie. Les antibiothérapies de ces 150 patients ont été classés en trois catégories (optimale, suboptimale, inefficace) en fonction du moment d’obtention des résultats de l’antibiogramme. Résultats : L’adaptation du traitement antibiotique en thérapie optimale suite au résultat de l’antibiogramme est survenue chez 46/100 (46 %) des HP à Gram négatif et chez 4/50 (2 %) des HP à Gram positif. Le TPR était significativement plus faible avec le dRAST™ par rapport à l’antibiogramme classique (29:35 (± 08:48) heures versus 50:55 (± 12:45) heures, p < 0,001). Conclusion : Pour les patients avec bactériémie nécessitant une adaptation de l’antibiothérapie empirique basée sur l’antibiogramme, le dRAST™ permettrait une administration plus rapide du traitement optimal

    β(1,3)-Glucanosyl-Transferase Activity Is Essential for Cell Wall Integrity and Viability of Schizosaccharomyces pombe

    13 páginas, 7 figuras, 2 tablas.[Background]: The formation of the cell wall in Schizosaccharomyces pombe requires the coordinated activity of enzymes involved in the biosynthesis and modification of b-glucans. The b(1,3)-glucan synthase complex synthesizes linear b(1,3)- glucans, which remain unorganized until they are cross-linked to other b(1,3)-glucans and other cell wall components. Transferases of the GH72 family play important roles in cell wall assembly and its rearrangement in Saccharomyces cerevisiae and Aspergillus fumigatus. Four genes encoding b(1,3)-glucanosyl-transferases -gas1+, gas2+, gas4+ and gas5+- are present in S. pombe, although their function has not been analyzed. [Methodology/Principal Findings]: Here, we report the characterization of the catalytic activity of gas1p, gas2p and gas5p together with studies directed to understand their function during vegetative growth. From the functional point of view, gas1p is essential for cell integrity and viability during vegetative growth, since gas1D mutants can only grow in osmotically supported media, while gas2p and gas5p play a minor role in cell wall construction. From the biochemical point of view, all of them display b(1,3)-glucanosyl-transferase activity, although they differ in their specificity for substrate length, cleavage point and product size. In light of all the above, together with the differences in expression profiles during the life cycle, the S. pombe GH72 proteins may accomplish complementary, non-overlapping functions in fission yeast.     The Middle Ear Microbiota in Healthy Dogs Is Similar to That of the External Ear Canal

    peer reviewedOtitis media can be a consequence of chronic otitis externa and could represent a perpetuating factor. While the microbiota of the EEC in healthy dogs and in the presence of otitis externa has been described, only sparse information is available concerning the normal microbiota of the middle ear. The objective was to compare the tympanic bulla (TB) with the external ear canal (EEC) microbiota in healthy dogs. Six healthy experimental Beagle dogs were selected based on the absence of otitis externa, negative cytology and bacterial culture from the TB. Samples from the EEC and TB were collected directly after death using a total ear canal ablation and lateral bulla osteotomy. The hypervariable segment V1–V3 of the 16S rDNA was amplified and sequenced with a MiSeq Illumina. The sequences were analyzed by the Mothur software using the SILVA database. No significant differences between the EEC and TB microbiota for the Chao1 richness index (p = 0.6544), the Simpson evenness index (p = 0.4328) and the reciprocal Simpson alpha diversity (p = 0.4313) were noted (Kruskal-Wallis test). A significant difference (p = 0.009) for the Chao1 richness index between the right and left EEC was observed. The microbiota profile was similar in the EEC and the TB of the Beagles
