    Exploring the cytotoxic mechanisms of Pediocin PA-1 towards HeLa and HT29 cells by comparison to known bacteriocins: Microcin E492, enterocin heterodimer and Divercin V41.

    The purpose of this study was to explore potential mechanisms of cytotoxicity towards HeLa and HT29 cells displayed by Pediocin PA-1. We did this by carrying out sequence alignments and 3D modelling of related bacteriocins which have been studied in greater detail: Microcin E492, Enterocin AB heterodimer and Divercin V41. Microcin E492 interacts with Toll-Like Receptor 4 in order to activate an apoptosis reaction, sequence alignment showed a high homology between Pediocin PA-1 and Microcin E492 whereas 3D modelling showed Pediocin PA-1 interacting with TLR-4 in a way reminiscent of Microcin E492. Furthermore, Pediocin PA-1 had the highest homology with the Enterocin heterodimer, particularly chain A; Enterocin has also shown to cause an apoptotic response in cancer cells. Based on this we are led to strongly believe Pediocin PA-1 interacts with TLRs in order to cause cell death. If this is the case, it would explain the difference in cytotoxicity towards HeLa over HT29 cells, due to difference in expression of particular TLRs. Overall, we believe Pediocin PA-1 exhibits a dual effect which is dose dependant, like that of Microcin. Unfortunately, due to the COVID-19 pandemic, we were unable to carry out experiments in the lab, and the unavailability of important data meant we were unable to provide and validate out solid conclusions, but rather suggestions. However, bioinformatic analysis is still able to provide information regarding structure and sequence analysis to draw plausible and evidence based conclusions. We have been able to highlight interesting findings and how these could be translated into future research and therapeutics in order to improve the quality of treatment and life of cancer patients

    Strategies for producing biochars with minimum PAH contamination

    With the aim to develop initial recommendations for production of biochars with minimal contamination with polycyclic aromatic hydrocarbons (PAHs), we analysed a systematic set of 46 biochars produced under highly controlled pyrolysis conditions. The effects of the highest treatment temperature (HTT), residence time, carrier gas flow and typical feedstocks (wheat / oilseed rape straw pellets (WSP), softwood pellets (SWP)) on 16 US EPA PAH concentration in biochar were investigated. Overall, the PAH concentrations ranged between 1.2 and 100 mg kg-1. On average, straw-derived biochar contained 5.8 times higher PAH concentrations than softwood-derived biochar. In a batch pyrolysis reactor, increasing carrier gas flow significantly decreased PAH concentrations in biochar; in case of straw, the concentrations dropped from 43.1 mg kg-1 in the absence of carrier gas to 3.5 mg kg-1 with a carrier gas flow of 0.67 L min-1; for woody biomass PAHs concentrations declined from 7.4 mg kg-1 to 1.5 mg kg-1 with the same change of carrier gas flow. In the temperature range of 350-650°C the HTT did not have any significant effect on PAH content in biochars, irrespective of feedstock type, however, in biochars produced at 750°C the PAH concentrations were significantly higher. After detailed investigation it was deduced that this intensification in PAH contamination at high temperatures was most likely down to the specifics of the unit design of the continuous pyrolysis reactor used. Overall, it was concluded that besides PAH formation, vaporisation is determining the PAH concentration in biochar. The fact that both of these mechanisms intensify with pyrolysis temperature (one increasing and the other one decreasing the PAH concentration in biochar) could explain why no consistent trend in PAH content in biochar with temperature has been found in the literature

    The origin of micrograins

    Using ultraviolet and infrared techniques, researchers investigated the origins of the tiny (approx. 10A) grains whose presence in the interstellar medium (ISM) is inferred from near-infrared photometry (Sellgren, Werner, and Dinerstein 1983; Sellgren 1984). The authors consider two possibilities: (1) that the grains are formed by condensation in stellar atmospheres; or (2) that they are formed by fragmentation of larger grains in interstellar shocks. They searched for evidence of very small grains in circumstellar environments by analyzing ultraviolet extinction curves in binaries containing hot companions, and by searching for the 3.3-micron emission feature in similar systems. The ultraviolet extinction curve analysis could be applied only to oxygen-rich systems, where small carbonaceous grains would not be expected, so these results provide only indirect information. Researchers find a deficiency of grains smaller than 800A in oxygen-rich systems, consistent with theoretical models of grain condensation which suggest that grains grow to large sizes before injection into the interstellar medium. More direct information on carbonaceous micrograins was obtained from the search for the 3.3-micron feature in carbon-rich binaries with hot companions, whose ultraviolet flux should excite the tiny grains to emit in the infrared. No 3.3-micron feature was found, suggesting that the micrograins are absent in these systems. In addition to the negative search for micrograins in circumstellar environments, researchers have also studied the possible association of these grains with shocks in the diffuse interstellar medium. Using Infrared Astronomy Satellite (IRAS) colors as indicators of the presence or absence of the small grains (e.g., Ryter, Puget, and Perault 1987 and references cited therein), researchers systematically searched for them in regions (reflection nebulae) expected to have sufficient ultraviolet flux to make them glow in the infrared. They found that the distribution is not uniform. The researchers propose that production of micrograins by fragmentation of larger grains in shocks could explain this uneven distribution

    Low power, compact charge coupled device signal processing system

    A variety of charged coupled devices (CCDs) for performing programmable correlation for preprocessing environmental sensor data preparatory to its transmission to the ground were developed. A total of two separate ICs were developed and a third was evaluated. The first IC was a CCD chirp z transform IC capable of performing a 32 point DFT at frequencies to 1 MHz. All on chip circuitry operated as designed with the exception of the limited dynamic range caused by a fixed pattern noise due to interactions between the digital and analog circuits. The second IC developed was a 64 stage CCD analog/analog correlator for performing time domain correlation. Multiplier errors were found to be less than 1 percent at designed signal levels and less than 0.3 percent at the measured smaller levels. A prototype IC for performing time domain correlation was also evaluated

    Patients often request a general check-up with blood tests. In the UK these are often referred to as an MOT, in allusion to the annual motor vehicle check. Some patients may, however, have unrealistic expectations of medical tests1 and underestimate their potential harms. While agreeing to some blood tests can be an easy way out for a busy clinician, it can expose patients to the harms of over-testing and produce extra workload downstream. We provide a framework for navigating these requests constructively, some elements of which are feasible within a 10 minute consultation

    Boerma and Kuhn (1976) established that \u27Dorman\u27 and \u27CNS\u27 each contain a single dominant gene conditioning resistance to peanut mottle virus (PMV). The gene was labelled Rpv, but it was not demonstrated that the genes in the cultivars were allelic. Shipe et al. (1979) also demonstrated the presence of single dominant genes for PMV resistance in each of \u27Arksoy\u27, \u27PI 89784\u27 and \u27PI 219789\u27, but made no tests for allelic relationships

    Effects of cochlear implantation on binaural hearing in adults with unilateral hearing loss

    A FDA clinical trial was carried out to evaluate the potential benefit of cochlear implant (CI) use for adults with unilateral moderate-to-profound sensorineural hearing loss. Subjects were 20 adults with moderate-to-profound unilateral sensorineural hearing loss and normal or near-normal hearing on the other side. A MED-EL standard electrode was implanted in the impaired ear. Outcome measures included: (a) sound localization on the horizontal plane (11 positions, −90° to 90°), (b) word recognition in quiet with the CI alone, and (c) masked sentence recognition with the target at 0° and the masker at −90°, 0°, or 90°. This battery was completed preoperatively and at 1, 3, 6, 9, and 12 months after CI activation. Normative data were also collected for 20 age-matched control subjects with normal or near-normal hearing bilaterally. The CI improved localization accuracy and reduced side bias. Word recognition with the CI alone was similar to performance of traditional CI recipients. The CI improved masked sentence recognition when the masker was presented from the front or from the side of normal or near-normal hearing. The binaural benefits observed with the CI increased between the 1- and 3-month intervals but appeared stable thereafter. In contrast to previous reports on localization and speech perception in patients with unilateral sensorineural hearing loss, CI benefits were consistently observed across individual subjects, and performance was at asymptote by the 3-month test interval. Cochlear implant settings, consistent CI use, and short duration of deafness could play a role in this result
