    Landscape ideology in the Greater Golden Horseshoe Greenbelt Plan: Negotiating material landscapes and abstract ideals in the city\u27s countryside

    We analyze the role of landscape ideology in the recent Ontario Greater Golden Horseshoe (GGH) Greenbelt Plan. Focusing on the “Protected Countryside,” the major land-use designation in the Plan that structures the Greenbelt framework, we explore tensions between abstract ideals of countryside used by policy makers to elicit support for the Plan and people\u27s lived experience of material landscapes of the peri-urban fringe. Approaching “countryside” from the combined perspectives of landscape studies and political ecology, we show how the abstract ideals used to build support for the protection of countryside in the high-level political arena are in tension with existing material landscapes as people experience them. When implementing the Greenbelt Plan, the abstract ideals have to be applied at the landscape level through negotiation with municipalities, property owners, and other interests. In addition to drawing upon more conventionally legitimate explanations for landscape protection based on environmental science and land-use planning principles, the designation of Protected Countryside and the strategies used to implement the Protected Countryside designation at the local level suggest a tentative commitment to recognizing landscape values and collaborative environmental management processes in policy-making. As with any such normative land-use plan, the success of the Greenbelt Plan hinges on the long-term agreement between planning agencies and diverse publics. We demonstrate the usefulness of approaching environmental management challenges at the urban–rural interface by bringing the perspectives of landscape studies and political ecology into implementation processes for land-use management strategies like the Greenbelt. We argue that public participants deserve legitimate collaborative roles in negotiating just and desirable land uses based on their experiences, and provide observations on ways to bring contested goals and tools for achieving them into reflexive negotiations about how landscapes are and should be produced

    Population of high-spin isomeric states following fragmentation of 238 U

    Isomeric ratios have been determined for 23 metastable states identified in A?200 nuclei from Pt to Rn near the valley of stability following fragmentation of 238U. This includes high-spin states with angular momenta ranging from (39/2) to 25. The experimental results are discussed together with those of similar experiments performed in this mass region. Isomeric ratios are compared with theoretical predictions where the angular momentum of the fragment arises purely due to the angular momentum of nucleons removed from the projectile. The theoretical yield of low-spin states is generally overestimated. In these cases the assumption of 100% feeding of the isomer may require modification. However, the yield of high-spin isomeric states [Im ? (39/2)] is significantly underestimated and highlights the requirement for a more complete theoretical framework in relation to the generation of fragment angular momentum. The enhanced population of high-spin states reported here is advantageous to future studies involving isomeric beams at fragmentation facilities such as the Rikagaku Kenkyusho RI Beam Factory (Japan) and next-generation facilities at the Facility for Antiproton and Ion Research (Germany) and Facility for Rare Isotope Beams (USA). � 2013 American Physical Society

    Subsistence practices, past biodiversity, and anthropogenic impacts revealed by New Zealand-wide ancient DNA survey

    New Zealand's geographic isolation, lack of native terrestrial mammals, and Gondwanan origins make it an ideal location to study evolutionary processes. However, since the archipelago was first settled by humans 750 y ago, its unique biodiversity has been under pressure, and today an estimated 49% of the terrestrial avifauna is extinct. Current efforts to conserve the remaining fauna rely on a better understanding of the composition of past ecosystems, as well as the causes and timing of past extinctions. The exact temporal and spatial dynamics of New Zealand's extinct fauna, however, can be difficult to interpret, as only a small proportion of animals are preserved as morphologically identifiable fossils. Here, we conduct a large-scale genetic survey of subfossil bone assemblages to elucidate the impact of humans on the environment in New Zealand. By genetically identifying more than 5,000 nondiagnostic bone fragments from archaeological and paleontological sites, we reconstruct a rich faunal record of 110 species of birds, fish, reptiles, amphibians, and marine mammals. We report evidence of five whale species rarely reported from New Zealand archaeological middens and characterize extinct lineages of leiopelmatid frog (Leiopelma sp.) and kakapo (Strigops habroptilus) haplotypes lost from the gene pool. Taken together, this molecular audit of New Zealand's subfossil record not only contributes to our understanding of past biodiversity and precontact Maori subsistence practices but also provides a more nuanced snapshot of anthropogenic impacts on native fauna after first human arrival

    Octupole transitions in the 208Pb region

    The 208Pb region is characterised by the existence of collective octupole states. Here we populated such states in 208Pb + 208Pb deep-inelastic reactions. γ-ray angular distribution measurements were used to infer the octupole character of several E3 transitions. The octupole character of the 2318 keV 17− → 14+ in 208Pb, 2485 keV 19/2 − → 13/2 + in 207Pb, 2419 keV 15/2 − → 9/2 + in 209Pb and 2465 keV 17/2 + → 11/2 − in 207Tl transitions was demonstrated for the first time. In addition, shell model calculations were performed using two different sets of two-body matrix elements.     X-rays Studies of the Solar System

    X-ray observatories contribute fundamental advances in Solar System studies by probing Sun-object interactions, developing planet and satellite surface composition maps, probing global magnetospheric dynamics, and tracking astrochemical reactions. Despite these crucial results, the technological limitations of current X-ray instruments hinder the overall scope and impact for broader scientific application of X-ray observations both now and in the coming decade. Implementation of modern advances in X-ray optics will provide improvements in effective area, spatial resolution, and spectral resolution for future instruments.     The Isolation of Nucleic Acids from Fixed, Paraffin-Embedded Tissues–Which Methods Are Useful When?

    Museums and pathology collections around the world represent an archive of genetic material to study populations and diseases. For preservation purposes, a large portion of these collections has been fixed in formalin-containing solutions, a treatment that results in cross-linking of biomolecules. Cross-linking not only complicates isolation of nucleic acid but also introduces polymerase “blocks” during PCR. A wide variety of methods exists for the recovery of DNA and RNA from archival tissues, and although a number of previous studies have qualitatively compared the relative merits of the different techniques, very few have undertaken wide scale quantitative comparisons. To help address this issue, we have undertaken a study that investigates the quality of nucleic acids recovered from a test panel of fixed specimens that have been manipulated following a number of the published protocols. These include methods of pre-treating the samples prior to extraction, extraction and nucleic acid purification methods themselves, and a post-extraction enzymatic repair technique. We find that although many of the published methods have distinct positive effects on some characteristics of the nucleic acids, the benefits often come at a cost. In addition, a number of the previously published techniques appear to have no effect at all. Our findings recommend that the extraction methodology adopted should be chosen carefully. Here we provide a quick reference table that can be used to determine appropriate protocols for particular aims

    Recommended practices for computerized clinical decision support and knowledge management in community settings: a qualitative study

    <p>Abstract</p> <p>Background</p> <p>The purpose of this study was to identify recommended practices for computerized clinical decision support (CDS) development and implementation and for knowledge management (KM) processes in ambulatory clinics and community hospitals using commercial or locally developed systems in the U.S.</p> <p>Methods</p> <p>Guided by the Multiple Perspectives Framework, the authors conducted ethnographic field studies at two community hospitals and five ambulatory clinic organizations across the U.S. Using a Rapid Assessment Process, a multidisciplinary research team: gathered preliminary assessment data; conducted on-site interviews, observations, and field surveys; analyzed data using both template and grounded methods; and developed universal themes. A panel of experts produced recommended practices.</p> <p>Results</p> <p>The team identified ten themes related to CDS and KM. These include: 1) workflow; 2) knowledge management; 3) data as a foundation for CDS; 4) user computer interaction; 5) measurement and metrics; 6) governance; 7) translation for collaboration; 8) the meaning of CDS; 9) roles of special, essential people; and 10) communication, training, and support. Experts developed recommendations about each theme. The original Multiple Perspectives framework was modified to make explicit a new theoretical construct, that of Translational Interaction.</p> <p>Conclusions</p> <p>These ten themes represent areas that need attention if a clinic or community hospital plans to implement and successfully utilize CDS. In addition, they have implications for workforce education, research, and national-level policy development. The Translational Interaction construct could guide future applied informatics research endeavors.</p

    Staphylococcus aureus Panton-Valentine Leukocidin Contributes to Inflammation and Muscle Tissue Injury

    Community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) threatens public health worldwide, and epidemiologic data suggest that the Panton-Valentine Leukocidin (PVL) expressed by most CA-MRSA strains could contribute to severe human infections, particularly in young and immunocompetent hosts. PVL is proposed to induce cytolysis or apoptosis of phagocytes. However, recent comparisons of isogenic CA-MRSA strains with or without PVL have revealed no differences in human PMN cytolytic activity. Furthermore, many of the mouse studies performed to date have failed to demonstrate a virulence role for PVL, thereby provoking the question: does PVL have a mechanistic role in human infection? In this report, we evaluated the contribution of PVL to severe skin and soft tissue infection. We generated PVL mutants in CA-MRSA strains isolated from patients with necrotizing fasciitis and used these tools to evaluate the pathogenic role of PVL in vivo. In a model of necrotizing soft tissue infection, we found PVL caused significant damage of muscle but not the skin. Muscle injury was linked to induction of pro-inflammatory chemokines KC, MIP-2, and RANTES, and recruitment of neutrophils. Tissue damage was most prominent in young mice and in those strains of mice that more effectively cleared S. aureus, and was not significant in older mice and mouse strains that had a more limited immune response to the pathogen. PVL mediated injury could be blocked by pretreatment with anti-PVL antibodies. Our data provide new insights into CA-MRSA pathogenesis, epidemiology and therapeutics. PVL could contribute to the increased incidence of myositis in CA-MRSA infection, and the toxin could mediate tissue injury by mechanisms other than direct killing of phagocytes
