    Use of Clumped-Isotope Thermometry To Constrain the Crystallization Temperature of Diagenetic Calcite

    We describe an approach to estimating the crystallization temperatures of diagenetic calcites using clumped-isotope thermometry, a paleothermometer based on the ^(13)C–^(18)O-bond enrichment in carbonates. Application of this thermometer to calcified gastropod shells and calcite cements in an early Eocene limestone from the Colorado Plateau reveals a record of calcite precipitation and replacement at temperatures varying from 14 to 123°C. The early Eocene host sediments were never deeply buried, but they experienced a significant thermal pulse associated with the emplacement of a late Miocene basalt flow. The combination of independent constraints on thermal history with clumped-isotope thermometry, petrographic (including cathodoluminescence) observations, and oxygen isotopic data provides an improved basis for estimation of the temperature and timing of diagenetic events and fluid sources. The petrography and calcite ή^(18)O values, taken alone, suggest that the aragonite-to-calcite transformation of gastropod shell material occurred simultaneously with early formation of cements and lithification of the matrix in the same sample. However, addition of clumped-isotope thermometry demonstrates that this phase transformation of shell material occurred at temperatures of 94–123°C in a highly rock-buffered microenvironment (i.e., with the isotopic composition of fluid buffered by coexisting carbonate), millions of years after lithification of the matrix and formation of initial low-temperature (14–19°C) calcite cements within shell body cavities. Clumped-isotope temperatures in excess of reasonable Earth-surface conditions recorded by later-formed cements demand that cement growth occurred in association with the lava emplacement. Our results illustrate the potential for clumped-isotope thermometry to constrain conditions of diagenesis and guide interpretations that would not be possible on the basis of conventional stable-isotopic and petrographic data alone, and demonstrate how petrographic characterization of clumped-isotope thermometry samples can benefit paleoclimate studies

    Direct measurements of episodic snow accumulation on the Antarctic polar plateau

    This is the published version, also available here: http://dx.doi.org/10.1029/2000JD900099.During a 1-year field experiment at a remote location on the Antarctic polar plateau (85.67°S, 46.38°W) influenced by moderate magnitude katabatic winds, snow accumulation was characterized at three different spatial and temporal scales using snow stakes, tracer material dispersed periodically on the snow surface, and an acoustic depth gauge. The spatial variability of snow accumulation was found to be large, on both annual and intra-annual timescales, and is attributed to the high frequency of moderate to strong winds at the site. Accumulation throughout the year was observed to be episodic in nature, with a small number of snow accumulation events producing the majority of the annual total accumulation for the site, averaging 0.174 m. In the intervals between observed accumulation events (up to several months), negative changes to snow surface height caused by sublimation and densification of the firn were quantified using an acoustic depth gauge. The rate of decrease in snow surface elevation was largest during the austral summer, as expected, and the overall change in snow surface elevation due to sublimation/densification during the year was estimated to be about −0.10 m. Using the precise timing of accumulation events provided by the acoustic depth gauge, meteorological surface observations, numerical model analyses, and satellite imagery were used to gain insights into whether the event was associated with precipitation or related exclusively to blowing snow and to diagnose the meteorological conditions producing the event. Meteorological conditions during the accumulation events were found to strongly support an association with precipitation events caused by mesoscale or synoptic-scale cyclones along the coastal margin. Dating of the accumulation profile using the dispersed tracer technique identified several other accumulation events that were not measured within the target area of the acoustic depth gauge, suggesting that snow accumulation data from a single acoustic depth gauge cannot be extrapolated over a broad area

    Abstract Background Even with well-known sampling biases, the fossil record is key to understanding macro-evolutionary patterns. During the Miocene to Pleistocene in the Caribbean Sea, the fossil record of scleractinian corals shows a remarkable period of rapid diversification followed by massive extinction. Here we combine a time-calibrated molecular phylogeny based on three nuclear introns with an updated fossil stratigraphy to examine patterns of radiation and extinction in Caribbean corals within the traditional family Faviidae. Results Concatenated phylogenetic analysis showed most species of Caribbean faviids were monophyletic, with the exception of two Manicina species. The time-calibrated tree revealed the stem group originated around the closure of the Tethys Sea (17.0 Ma), while the genus Manicina diversified during the Late Miocene (8.20 Ma), when increased sedimentation and productivity may have favored free-living, heterotrophic species. Reef and shallow water specialists, represented by Diploria and Favia, originate at the beginning of the Pliocene (5 – 6 Ma) as the Isthmus of Panama shoaled and regional productivity declined. Conclusions Later origination of the stem group than predicted from the fossil record corroborates the hypothesis of morphological convergence in Diploria and Favia genera. Our data support the rapid evolution of morphological and life-history traits among faviid corals that can be linked to Mio-Pliocene environmental changes.</p

    A detailed assessment of snow accumulation in katabatic wind areas on the Ross Ice Shelf, Antarctica

    This is the published version, also available here: http://dx.doi.org/10.1029/97JD02337.An investigation of time dependent snow accumulation and erosion dynamics in a wind-swept environment was undertaken at two automatic weather stations sites on the Ross Ice Shelf between January 1994 and November 1995 using newly developed instrumentation employing a technique which automatically disperses inert, colored (high albedo) glass microspheres onto the snow surface at fixed intervals throughout the year. The microspheres act as a time marker and tracer to allow the accumulation rate and wind erosion processes to be quantified with a high temporal resolution. Snow core and snow pit sampling was conducted twice during the study period to identify microsphere horizons in the annual snow accumulation profile, allowing the snow accumulation/erosion events to be reconstructed. The two sites chosen for this investigation have characteristically different mean wind speeds and therefore allow a comparative examination on the role of wind on ice sheet growth. Mass accumulation rate at the two sites for the 14-day integration periods available ranged from 0.0 to >2.0 kg m−2 d−1. The mean mass accumulation rate during the study period was greater at the site with stronger winds (0.69 kg m−2 d−1) than the site with lower mean wind speeds (0.61 kg m−2 d−1); however, the difference between the two means is not statistically significant. Accumulation rates derived from an ultrasonic snow depth gauge operated at one of the sites are compared to the actual tracer-derived accumulation rates and show the limitations of only having a measure of snow surface height with no instantaneous measurements of the snow density profile. Snow depth gauge derived accumulation rates were found to be greatly overestimated during high-accumulation periods and were greatly underestimated during low-accumulation periods

    The New Deal is the Labour government's flagship programme to "end the tragic waste of youth and long-term unemployment" by getting people off welfare benefits and into work. This paper argues that the principal weakness of the New Deal is that it seeks to influence the character of labour supply (i.e. the motivation and skills of the unemployed) while neglecting the state of labour demand, which varies greatly between places. The uneven geography of unemployment in the UK is likely to have a crucial bearing on the programme's impact and effectiveness, but this has been largely ignored in its development. The paper outlines some of the practical consequences of this imbalance and suggests how it could be rectified for the programme to be more effective

    The expression of TRMT2A, a novel cell cycle regulated protein, identifies a subset of breast cancer patients with HER2 over-expression that are at an increased risk of recurrence

    <p>Abstract</p> <p>Background</p> <p>Over-expression of <it>HER2 </it>in a subset of breast cancers (<it>HER2</it>+) is associated with high histological grade and aggressive clinical course. Despite these distinctive features, the differences in response of <it>HER2</it>+ patients to both adjuvant cytotoxic chemotherapy and targeted therapy (e.g. trastuzumab) suggests that unrecognized biologic and clinical diversity is confounding treatment strategies. Furthermore, the small but established risk of cardiac morbidity with trastuzumab therapy compels efforts towards the identification of biomarkers that might help stratify patients.</p> <p>Methods</p> <p>A single institution tissue array cohort assembled at the Clearview Cancer Institute of Huntsville (CCIH) was screened by immunohistochemistry staining using a large number of novel and commercially available antibodies to identify those with a univariate association with clinical outcome in <it>HER2</it>+ patients. Staining with antibody directed at TRMT2A was found to be strongly associated with outcome in <it>HER2</it>+ patients. This association with outcome was tested in two independent validation cohorts; an existing staining dataset derived from tissue assembled at the Cleveland Clinic Foundation (CCF), and in a new retrospective study performed by staining archived paraffin blocks available at the Roswell Park Cancer Institute (RPCI).</p> <p>Results</p> <p>TRMT2A staining showed a strong correlation with likelihood of recurrence at five years in 67 <it>HER2</it>+ patients from the CCIH discovery cohort (HR 7.0; 95% CI 2.4 to 20.1, p < 0.0004). This association with outcome was confirmed using 75 <it>HER2</it>+ patients from the CCF cohort (HR 3.6; 95% CI 1.3 to 10.2, p < 0.02) and 64 patients from the RPCI cohort (HR 3.4; 95% CI 1.3-8.9, p < 0.02). In bivariable analysis the association with outcome was independent of grade, tumor size, nodal status and the administration of conventional adjuvant chemotherapy in the CCIH and RPCI cohorts.</p> <p>Conclusions</p> <p>Studies from three independent single institution cohorts support TRMT2A protein expression as a biomarker of increased risk of recurrence in <it>HER2+ </it>breast cancer patients. These results suggest that TRMT2A expression should be further studied in the clinical trial setting to explore its predictive power for response to adjuvant cytotoxic chemotherapy in combination with <it>HER2 </it>targeted therapy.</p
