Screening in YBCO at large wave vectors
We present experimental inelastic x-ray scattering (IXS) and ab initio
time-dependent density-functional-theory (TDDFT) studies of YBa2Cu3O7-{\delta}.
The response of the low-lying Ba 5p and Y 4p core electrons is shown to
interact strongly with the Cu 3d and O 2p excitations, with important
consequences on screening. The agreement between IXS and TDDFT results is
excellent, apart from a new type of excitations, mainly related to loosely
bound Ba electrons and significantly affected by correlations. This points to
correlation mechanisms not fully described by TDDFT that might have a role in
giving rise to antiscreening.Comment: 6 pages, 3 figure
Nicotinatfermentation in Eubacterium barkeri
Aus EcoRI- und BamHI/BglII-Phagen-Genbänken von Eubacterium barkeri wurde mit Hilfe Digoxigenin-markierten homologen DNA-Sonden, direkter genomischer Sequenzierung und Seegene Speedwalking ein Gencluster von 23 kbp kloniert, das fĂĽr die Enzyme der Nicotinatfermentation kodierte. Nach Sequenzierung und Analyse wurden die folgenden 22 offenen Leserahmen identifiziert. Die ndhFSLM Gene kodieren fĂĽr die Untereinheiten der Nicotinat-Dehydrogenase, deren Aminosäuresequenzen signifikante Identitäten zu Xanthin-Dehydrogenase aufweisen. hnr kodiert fĂĽr die 6-Hydroxynicotinat-Reduktase, ein [2Fe-2S]+x 2 x [4Fe-4S] Enzym mit kovalent gebundenem Flavin. ena kodiert fĂĽr die Enamidase, deren Aminosäuresequenz eine Zugehörigkeit zur Amidohydrolase Superfamilie aufweist, und hgd fĂĽr die 2-(Hydroxymethyl)glutarat-Dehydrogenase. Die dmdAB Gene (60-73 % Aminosäuresequenz-Identität zu Isopropylmalat-Isomerase) kodieren fĂĽr die beiden Untereinheiten der Dimethylmalat-Dehydratase. Ăśber die Aminosäuresequenzenidentität zur α-Untereinheit der [4Fe-4S]-Serin-Dehydratase (23-26 %) wurde hmd als Gen potentiell identifiziert, das fĂĽr die 2-(Hydroxymethyl)glutarat-Dehydratase kodiert. Mit den bereits vorher charakterisierten mgm (2-Methylglutarat-Mutase), mii (3-Methylitaconat-Isomerase), mgmL (2-Methylglutarat-Mutase Reaktivase/Stabilisierungsfaktor) und dml (Dimethylmalat-Lyase) Genen sind jetzt die Primärstrukturen aller Enzyme der Nicotinatfermentation identifiziert. Neben diesen Genen wurden zwischen den Nicotinat-Fermentationsgenen die orfC, orfD und orfE Gene, stromaufwärts von hnr die nicR, orfA und orfB Gene, und stromabwärts von dmdAB die orfF und orfG Gene angetroffen. AuĂźer fĂĽr nicR (potentieller Regulator) und orfC und orfE (grenzend an hnr-Homologen) konnte eine Beteiligung am Nicotinatabbau noch nicht aufgeklärt werden. Ăśber die Sequenzen von Hnr und Ena wurden in 6 Proteobakterien Gencluster fĂĽr den Abbau von Nicotinat zu 2-Formylglutarat gefunden.
Für biochemische Untersuchungen wurden ena und hgd heterolog in E. coli exprimiert. Die mit N-terminalen Streptags versetzte Enamidase und 2-(Hydroxymethyl)glutarat-Dehydrogenase wurden mit Streptactin-Affinitätschromatographie gereinigt. Die kinetischen Parameter wurden bestimmt.
Die Kristallstruktur der Enamidase von E. barkeri wurde in Zusammenarbeit mit der AG Essen zu einer Auflösung von 1,89 Å bestimmt. Gemäss Metallanalyse wurden im Homotetramer vier binukleäre Eisen-Zink Zentren gefunden. Glutamat, Chlorid und ein Sauerstoff Atom verbrückten die Metallionen
Low-T_c Josephson junctions with tailored barrier
Nb/Al_2O_3/Ni_{0.6}Cu_{0.4}/Nb based
superconductor-insulator-ferromagnet-superconductor (SIFS) Josephson tunnel
junctions with a thickness step in the metallic ferromagnetic
\Ni_{0.6}\Cu_{0.4} interlayer were fabricated. The step was defined by optical
lithography and controlled etching. The step height is on the scale of a few
angstroms. Experimentally determined junction parameters by current-voltage
characteristics and Fraunhofer pattern indicate an uniform F-layer thickness
and the same interface transparencies for etched and non-etched F-layers. This
technique could be used to tailor low-T_c Josephson junctions having controlled
critical current densities at defined parts of the junction area, as needed for
tunable resonators, magnetic-field driven electronics or phase modulated

Purification and structural characterization of the Na<sup>+</sup>-translocating ferredoxin: NAD<sup>+</sup> reductase (Rnf) complex of Clostridium tetanomorphum
Various microbial metabolisms use H+/Na+-translocating ferredoxin:NAD+ reductase (Rnf) either to exergonically oxidize reduced ferredoxin by NAD+ for generating a transmembrane electrochemical potential or reversely to exploit the latter for producing reduced ferredoxin. For cryo-EM structural analysis, we elaborated a quick four-step purification protocol for the Rnf complex from Clostridium tetanomorphum and integrated the homogeneous and active enzyme into a nanodisc. The obtained 4.27 Ă… density map largely allows chain tracing and redox cofactor identification complemented by biochemical data from entire Rnf and single subunits RnfB, RnfC and RnfG. On this basis, we postulated an electron transfer route between ferredoxin and NAD via eight [4Fe-4S] clusters, one Fe ion and four flavins crossing the cell membrane twice related to the pathway of NADH:ubiquinone reductase. Redox-coupled Na+ translocation is provided by orchestrating Na+ uptake/release, electrostatic effects of the assumed membrane-integrated FMN semiquinone anion and accompanied polypeptide rearrangements mediated by different redox steps
