Associations between APOE Variants and Metabolic Traits and the Impact of Psychological Stress
In a previous study, we observed that associations between APOE rs439401 and metabolic traits were moderated by chronic stress. Thus, in a population of stressed and non-stressed Danish men, we examined whether associations between APOE rs439401 and a panel of metabolic quantitative traits, all metabolic traits which may lead to T2D and CVD were moderated by psychological stress.Obese young men (n = 475, BMI ≥ 31.0 kg/m(2)) and a randomly selected control group (n = 709) identified from a population of 141,800 men were re-examined in two surveys (S-46: mean age 46, S-49: mean age 49 years) where anthropometric and biochemical measures were available. Psychological stress factors were assessed by a self-administered 7-item questionnaire. Each item had the possible response categories "yes" and "no" and assessed familial problems and conflicts. Summing positive responses constituted a stress item score, which was then dichotomized into stressed and non-stressed. Logistic regression analysis, applying a recessive genetic model, was used to assess odds ratios (OR) of the associations between APOE rs439401 genotypes and adverse levels of metabolic traits. The APOE rs439401 TT-genotype associated positively with BMI (OR = 1.09 [1.01; 1.17]), waist circumference (OR = 1.09 [1.02; 1.17]) in stressed men at S-46. Positive associations were observed for fasting plasma glucose (OR = 1.42 [1.07; 1.87]), serum triglycerides (OR = 1.41 [1.05; 1.91]) and with fasting plasma insulin (OR = 1.48 [1.05; 2.08]) in stressed men at S-49. Rs439401 TT-genotype also associated positively with surrogate measures of insulin resistance (HOMA-IR; OR = 1.21 [1.03; 1.41]) and inversely with insulin sensitivity (Stumvoll index; OR = 0.90 [0.82; 0.99], BIGTT-S(I); OR = 0.60 [0.43; 0.85]) in stressed men. No significant associations were observed in non-stressed men, albeit the estimates showed similar but weaker trends as in stressed men.The present results suggest that the APOE rs439401 TT-genotype is associated with an adverse metabolic profile in a population of psychologically stressed Danish men

Filter Paper: Solution to High Self-Attenuation Corrections in HEPA Filter Measurements
An 8 by 8 by 6 inch High Efficiency Particulate Air (HEPA) filter was measured as part of a uranium holdup survey in June of 2005 as it has been routinely measured every two months since 1998. Although the survey relies on gross gamma count measurements, this was one of a few measurements that had been converted to a quantitative measurement in 1998. The measurement was analyzed using the traditional Generalized Geometry Holdup (GGH) approach, using HMS3 software, with an area calibration and self-attenuation corrected with an empirical correction factor of 1.06. A result of 172 grams of {sup 235}U was reported. The actual quantity of {sup 235}U in the filter was approximately 1700g. Because of this unusually large discrepancy, the measurement of HEPA filters will be discussed. Various techniques for measuring HEPA filters will be described using the measurement of a 24 by 24 by 12 inch HEPA filter as an example. A new method to correct for self attenuation will be proposed for this measurement Following the discussion of the 24 by 24 by 12 inch HEPA filter, the measurement of the 8 by 8 by 6 inch will be discussed in detail
Nonequilibrium phase transition due to social group isolation
We introduce a simple model of a growing system with competing
communities. The model corresponds to the phenomenon of defeats suffered by
social groups living in isolation. A nonequilibrium phase transition is
observed when at critical time the first isolated cluster occurs. In the
one-dimensional system the volume of the new phase, i.e. the number of the
isolated individuals, increases with time as . For a large number
of possible communities the critical density of filled space equals to where is the system size. A similar transition is observed
for Erd\H{o}s-R\'{e}nyi random graphs and Barab\'{a}si-Albert scale-free
networks. Analytic results are in agreement with numerical simulations.Comment: 5 pages, 4 figure
A functional polymorphism in the 5HTR2C gene associated with stress responses also predicts incident cardiovascular events.
Previously we have shown that a functional nonsynonymous single nucleotide polymorphism (rs6318) of the 5HTR2C gene located on the X-chromosome is associated with hypothalamic-pituitary-adrenal axis response to a stress recall task, and with endophenotypes associated with cardiovascular disease (CVD). These findings suggest that individuals carrying the rs6318 Ser23 C allele will be at higher risk for CVD compared to Cys23 G allele carriers. The present study examined allelic variation in rs6318 as a predictor of coronary artery disease (CAD) severity and a composite endpoint of all-cause mortality or myocardial infarction (MI) among Caucasian participants consecutively recruited through the cardiac catheterization laboratory at Duke University Hospital (Durham, NC) as part of the CATHGEN biorepository. Study population consisted of 6,126 Caucasian participants (4,036 [65.9%] males and 2,090 [34.1%] females). A total of 1,769 events occurred (1,544 deaths and 225 MIs; median follow-up time = 5.3 years, interquartile range = 3.3-8.2). Unadjusted Cox time-to-event regression models showed, compared to Cys23 G carriers, males hemizygous for Ser23 C and females homozygous for Ser23C were at increased risk for the composite endpoint of all-cause death or MI: Hazard Ratio (HR) = 1.47, 95% confidence interval (CI) = 1.17, 1.84, p = .0008. Adjusting for age, rs6318 genotype was not related to body mass index, diabetes, hypertension, dyslipidemia, smoking history, number of diseased coronary arteries, or left ventricular ejection fraction in either males or females. After adjustment for these covariates the estimate for the two Ser23 C groups was modestly attenuated, but remained statistically significant: HR = 1.38, 95% CI = 1.10, 1.73, p = .005. These findings suggest that this functional polymorphism of the 5HTR2C gene is associated with increased risk for CVD mortality and morbidity, but this association is apparently not explained by the association of rs6318 with traditional risk factors or conventional markers of atherosclerotic disease
Systolic Blood Pressure, Socioeconomic Status, and Biobehavioral Risk Factors in a Nationally Representative US Young Adult Sample
In the National Longitudinal Study of Adolescent Health, a US longitudinal study of over 15,000 young adults, we examined the extent to which socioeconomic status is linked to systolic blood pressure, and whether biobehavioral risk factors mediate the association. Over 62% of the participants had systolic blood pressure >120 mmHg and 12% with systolic blood pressure >140 mmHg. Over 66% were classified as at least overweight (Body Mass Index>25 kg/m2), with over 36% meeting criteria for at least Class I obesity (Body Mass Index>30 kg/m2). Multivariate models showed that higher household income and being married were independently associated with lower systolic blood pressure. Higher body mass index, greater waist circumference, smoking, and higher alcohol intake were each independently associated with higher systolic blood pressure. Meditational analyses suggested that higher education level was associated with lower systolic blood pressure by way of lower body mass, smaller waist circumference, and lower resting heart rate. When these indirect effects were accounted for, education was not significantly associated with systolic blood pressure. In contrast, household income remained associated with systolic blood pressure even with control for all covariates. Results reinforce current public health concerns about rates of obesity and high blood pressure among young adults and suggest that disparities in education level and household income may play an important role the observed decrements in health. Identifying modifiable mechanisms that link socioeconomic status to systolic blood pressure using data from a large representative sample may improve risk stratification and guide the development of effective interventions
The impact of emotional well-being on long-term recovery and survival in physical illness: a meta-analysis
This meta-analysis synthesized studies on emotional well-being as predictor of the prognosis of physical illness, while in addition evaluating the impact of putative moderators, namely constructs of well-being, health-related outcome, year of publication, follow-up time and methodological quality of the included studies. The search in reference lists and electronic databases (Medline and PsycInfo) identified 17 eligible studies examining the impact of general well-being, positive affect and life satisfaction on recovery and survival in physically ill patients. Meta-analytically combining these studies revealed a Likelihood Ratio of 1.14, indicating a small but significant effect. Higher levels of emotional well-being are beneficial for recovery and survival in physically ill patients. The findings show that emotional well-being predicts long-term prognosis of physical illness. This suggests that enhancement of emotional well-being may improve the prognosis of physical illness, which should be investigated by future research
