    The Effect of Substratum Roughness on Osteoclast-like Cells In Vitro

    Calcium phosphate powders were used to produce three groups of experimental substrata for the culture of primary rat bone marrow cells in conditions which permitted the survival and function of osteoclasts. Each of the three experimental groups were subdivided by differences in substratum surface roughness and following a culture period of 7 to 11 days, the culture units were stained for tartrate-resistant acid phosphatase activity. In all samples both small, sometimes mononuclear, and large multinucleate cells stained positive for tartrate-resistant acid phosphatase activity and the numbers and types of cells were quantified and statistically analyzed. Following histochemical staining the samples were dehydrated and gold coated for examination by scanning electron microscopy. Cells were found to create distinct resorption lacunae in most substrata, but not on the dense, high temperature sintered hydroxyapatite, and cells responsible for this activity were confirmed as exhibiting positive tartrate resistant acid phosphatase activity. Statistical analyses showed that both the total number of tartrate-resistant acid phosphatase positive cells and the number of multinucleate tartrate-resistant acid phosphatase positive cells was greater on the rougher than the smoother surfaces

    Web Service Discovery in a Semantically Extended UDDI Registry: the Case of FUSION

    Service-oriented computing is being adopted at an unprecedented rate, making the effectiveness of automated service discovery an increasingly important challenge. UDDI has emerged as a de facto industry standard and fundamental building block within SOA infrastructures. Nevertheless, conventional UDDI registries lack means to provide unambiguous, semantically rich representations of Web service capabilities, and the logic inference power required for facilitating automated service discovery. To overcome this important limitation, a number of approaches have been proposed towards augmenting Web service discovery with semantics. This paper discusses the benefits of semantically extending Web service descriptions and UDDI registries, and presents an overview of the approach put forward in project FUSION, towards semantically-enhanced publication and discovery of services based on SAWSDL

    Energy-Filtering Transmission Electron Microscopy of Biological Specimens

    By energy-filtering transmission electron microscopy (EFTEM) electrons can be separated by their energy losses. An electron-energy filter, added to the microscope column allows the measurement of the energy distribution of transmitted electrons that have lost energy (\u3c 2,000 eV, with an energy resolution of ~ 1 eV). These filtered electrons, recorded either as a spectrum or as an image, are composed of two parts superimposed on top of each other: (a) the unspecific energy-loss population (= the continuum) and (b) the specific element-related energy-loss population (= the edges). At the edges, electron data in spectra and images are mathematically processed, to obtain the desired element-related net-intensity values or images. These data are related to the total transmitted electron intensity, from the zero-and low-loss spectral region giving the relative spectral-or image intensity ratios (SR*x, IR*x), which can be related to the element concentration. The acquisition of the zero-loss and low-loss data is hampered by the restricted dynamic range of the TV camera. By improvements through the introduction of calibrated attenuation filters in the optical path to the TV-camera, more reliable values for SR*x and IR*x can be acquired. By addition of Bio-standards adjacent to the tissue, a known and unknown concentration of the element present in the same ultrathin section and the bias in the concentration estimation, can be obtained. Some practical examples are given for the estimation of the iron concentration in siderosomes, boron in melanosomes and calcium in calcium oxalate monohydrate crystals

    What lies beneath? The role of informal and hidden networks in the management of crises

    Crisis management research traditionally focuses on the role of formal communication networks in the escalation and management of organisational crises. Here, we consider instead informal and unobservable networks. The paper explores how hidden informal exchanges can impact upon organisational decision-making and performance, particularly around inter-agency working, as knowledge distributed across organisations and shared between organisations is often shared through informal means and not captured effectively through the formal decision-making processes. Early warnings and weak signals about potential risks and crises are therefore often missed. We consider the implications of these dynamics in terms of crisis avoidance and crisis management

    Objective. To acknowledge the dinical syndrome chronic fatigue syndrome (CFS) and outline the diagnostic criteria and reasonable management.Outcomes. Attempt at containment of treatment cost and improvement of the quality of care of patients with CFS.Evidence. Delphi-type commentary from 20 expert clinicians and appropriate organisations. Limited literature survey.Values. To clarify the reasonable management of CFS amid conflicting clinical opinion on a condition qf concern to patients, funders and doctors, An adaptation:of an existing guideline was sent to organisations and individuals for comment. Comments received were included in this guideline where possible.Benefits, harms and costs. To acknowledge a clinical syndrome with a reasonable approach to management considering the cost implications. No cost analysis was done.Recommendations. To recommend the following: (I) diagnostic criteria for CFS; (il) potential differential diagnoses and possible investigations; and (iil) management protocol.Validation. The draft guidelines were subjected to external review by individual doctors who are acknowledged CFS treaters, doctor groups and the patient support group. There were major disputes about the content, with the responses falling into two groups: thosewho do not believe CFS is a distinguishable illness, and those who do.Developer and funding.     Flip dynamics in octagonal rhombus tiling sets

    We investigate the properties of classical single flip dynamics in sets of two-dimensional random rhombus tilings. Single flips are local moves involving 3 tiles which sample the tiling sets {\em via} Monte Carlo Markov chains. We determine the ergodic times of these dynamical systems (at infinite temperature): they grow with the system size NTN_T like Cst.NT2lnNTCst. N_T^2 \ln N_T; these dynamics are rapidly mixing. We use an inherent symmetry of tiling sets and a powerful tool from probability theory, the coupling technique.     A theorem on the absence of phase transitions in one-dimensional growth models with onsite periodic potentials

    We rigorously prove that a wide class of one-dimensional growth models with onsite periodic potential, such as the discrete sine-Gordon model, have no phase transition at any temperature T>0T>0. The proof relies on the spectral analysis of the transfer operator associated to the models.     Cytocompatibility and response of osteoblastic-like cells to starch-based polymers : effect of several additives and processing conditions

    This work reports on the biocompatibility evaluation of new biodegradable starch-based polymers that are under consideration for use in orthopaedic temporary applications and as tissue engineering scaffolds. It has been shown in previous works that by using these polymers it is both possible to produce polymer/hydroxyapatite (HA) composites (with or without the use of coupling agents) with mechanical properties matching those of the human bone, and to obtain 3D structures generated by solid blowing agents, that are suitable for tissue engineering applications. This study was focused on establishing the influence of several additives (ceramic fillers, blowing agents and coupling agents) and processing methods/conditions on the biocompatibility of the materials described above. The cytotoxicity of the materials was evaluated using cell culture methods, according to ISO/EN 109935 guidelines. A cell suspension of human osteosarcoma cells (HOS) was also seeded on a blend of corn starch with ethylene vinyl alcohol (SEVA-C) and on SEVA-C/HA composites, in order to have a preliminary indication on cell adhesion and proliferation on the materials surface. In general, the obtained results show that all the different materials based on SEVA-C, (which are being investigated for use in several biomedical applications), as well as all the additives (including the novel coupling agents) and different processing methods required to obtain the different properties/products, can be used without inducing a cytotoxic behaviour to the developed biomaterial

    We give an expression for the partition function of a one-dimensional log-gas comprised of particles of (possibly) different integer charge at inverse temperature {\beta} = 1 (restricted to the line in the presence of a neutralizing field) in terms of the Berezin integral of an associated non- homogeneous alternating tensor. This is the analog of the de Bruijn integral identities [3] (for {\beta} = 1 and {\beta} = 4) ensembles extended to multicomponent ensembles.Comment: 14 page
