    Flexible Ultrathin PolyDVB/EVB Composite Membranes for the Optimization of a Whole Blood Glucose Sensor.

    An ultrathin composite membrane has been developed as the outer covering barrier in a model amperometric glucose oxidase enzyme electrode. The membrane was formed by cathodic electropolymerization of divinylbenzene/ethylvinylbenzene at the surface of a gold coated polyester support membrane. Permeability coefficients were determined for O2 and glucose across membranes with a range of polymer thicknesses. Anionic interferents (such as ascorbate), were screened from the working electrode via a charge exclusion mechanism. The enzyme electrode showed an initial 10% signal drift when first exposed to whole human blood over a period of 2 hours, after which responses remained essentially stable. Whole blood patient glucose determinations yielded a correlation coefficient of r2=0.99 compared to standard hospital analyses

    Deep Herschel view of obscured star formation in the Bullet cluster

    We use deep, five band (100–500 μm) data from the Herschel Lensing Survey (HLS) to fully constrain the obscured star formation rate, SFR_(FIR), of galaxies in the Bullet cluster (z = 0.296), and a smaller background system (z = 0.35) in the same field. Herschel detects 23 Bullet cluster members with a total SFRFIR = 144±14 M_☉ yr^(-1). On average, the background system contains brighter far-infrared (FIR) galaxies, with ~50% higher SFRFIR (21 galaxies; 207 ± 9 M_☉ yr^(-1)). SFRs extrapolated from 24 μm flux via recent templates (SFR_(24 µm)) agree well with SFRFIR for ~60% of the cluster galaxies. In the remaining ~40%, SFR24 µm underestimates SFR_(FIR) due to a significant excess in observed S_(100)/S_(24) (rest frame S_(75)/S_(18)) compared to templates of the same FIR luminosity

    The PCR primer GanET has previously been shown to be suitable for the specific amplification of DNA from Ganoderma boninense. A DNA extraction and PCR method has been developed that allows for the amplification of the G. boninense DNA from environmental samples of oil palm tissue. The GanET primer reaction was used in conjunction with a palm-sampling programme to investigate the possible infection of young palms through cut frond base surfaces. Ganoderma DNA was detected in frond base material at a greater frequency than would be expected by comparison with current infection levels. Comparisons are made between the height of the frond base infected, the number of frond bases infected, and subsequent development of basal stem rot. The preliminary results suggest that the development of basal stem rot may be more likely to occur when young lower frond bases are infected

    Mapping Henry: Dendrochronological Analysis of a Sixteenth-Century Panel Painting Based Upon Synchrotron-Sourced X-ray Fluorescence Mapping

    The study of materials that comprise artworks significantly contributes to understanding of age and provenance. While dendrochronology is a particularly valuable and well-established technique for panel paintings comprising oak timber, conventional practices of resurfacing end-grains to reveal tree rings is becoming less acceptable because it removes material, modifying the painting. Recently, application of non-destructive X-ray fluorescence (XRF) spectroscopy to a portrait of Henry VIII held by the Art Gallery of New South Wales (AGNSW), Sydney, Australia revealed tree-ring boundaries in the resulting high-resolution elastic scatter XRF map. In this study, we examine the dendrochronological potential of that mapping with the aim of contributing to resolving the relationship of the AGNSW portrait to similar paintings elsewhere. Examination of the timber revealed affinities with Quercus petraea (Matt.). We measured tree-ring widths along multiple paths in the XRF scatter map and crossdated the resulting 81-year XRF ring-width (XRF portrait ) series against master chronologies of English, western European, and Baltic origin. Rather than an arbitrarily defined threshold, we used a Bonferroni correction to determine a minimum significance level for crossdating. While the XRF portrait series did not crossdate with the continental European chronologies, we identified a single significant (α = 3.4 × 10−6) dating position with the English chronology (1400–1480 CE). Cross-matching with site-level chronologies revealed a cluster of high t-values in central-southern England. The earliest date of felling precedes the documented date of completion for the AGNSW and two similar Tudor portraits of Henry VIII held by the National Portrait Gallery (NPG), London and one at the Society of Antiquaries, London. While the apparent British provenance of timber used in the AGNSW portrait contrasts to Baltic origin of timbers used for the two NPG portraits and the majority of English panel paintings, it is consistent with the provenance of the timber used for the Society of Antiquaries portrait

    A Concept for an STJ-based Spectrograph

    We describe a multi-order spectrograph concept suitable for 8m-class telescopes, using the intrinsic spectral resolution of Superconducting Tunneling Junction detectors to sort the spectral orders. The spectrograph works at low orders, 1-5 or 1-6, and provides spectral coverage with a resolving power of R~8000 from the atmospheric cutoff at 320 nm to the long wavelength end of the infrared H or K band at 1800 nm or 2400 nm. We calculate that the spectrograph would provide substantial throughput and wavelength coverage, together with high time resolution and sufficient dynamic range. The concept uses currently available technology, or technologies with short development horizons, restricting the spatial sampling to two linear arrays; however an upgrade path to provide more spatial sampling is identified. All of the other challenging aspects of the concept - the cryogenics, thermal baffling and magnetic field biasing - are identified as being feasible.Comment: Accepted in Monthly Notices of the Royal Astronomical Society, 12 pages with 10 figure

    The arrival of New Labour into Government witnessed the prominent re-emergence of place onto the policy agenda. This heralded a range of area-based-initiatives designed to both tackle neighbourhood forms of deprivation and to re-establish a sense of identity and connection between individuals and their local community. In terms of place-making, effective and inclusive participation, representation and leadership were all identified as prerequisites for the creation of sustainable communities . But how important is local leadership and strategic vision within local public service organisations in achieving the desired place-making outcomes? This paper examines the extent to which local leadership and strategic vision represents a significant factor in promoting higher levels of satisfaction, belonging, cohesion and participation across single tier councils in England. The ensuing empirical evidence raises significant questions not only about the importance of local leadership in place-making, but also the environmental and organizational factors that shape local places

    Heterologous prime-boost-boost immunisation of Chinese cynomolgus macaques using DNA and recombinant poxvirus vectors expressing HIV-1 virus-like particles

    Background: There is renewed interest in the development of poxvirus vector-based HIV vaccines due to the protective effect observed with repeated recombinant canarypox priming with gp120 boosting in the recent Thai placebo-controlled trial. This study sought to investigate whether a heterologous prime-boost-boost vaccine regimen in Chinese cynomolgus macaques with a DNA vaccine and recombinant poxviral vectors expressing HIV virus-like particles bearing envelopes derived from the most prevalent clades circulating in sub-Saharan Africa, focused the antibody response to shared neutralising epitopes. Methods: Three Chinese cynomolgus macaques were immunised via intramuscular injections using a regimen composed of a prime with two DNA vaccines expressing clade A Env/clade B Gag followed by boosting with recombinant fowlpox virus expressing HIV-1 clade D Gag, Env and cholera toxin B subunit followed by the final boost with recombinant modified vaccinia virus Ankara expressing HIV-1 clade C Env, Gag and human complement protein C3d. We measured the macaque serum antibody responses by ELISA, enumerated T cell responses by IFN-gamma ELISpot and assessed seroneutralisation of HIV-1 using the TZM-bl beta-galactosidase assay with primary isolates of HIV-1. Results: This study shows that large and complex synthetic DNA sequences can be successfully cloned in a single step into two poxvirus vectors: MVA and FPV and the recombinant poxviruses could be grown to high titres. The vaccine candidates showed appropriate expression of recombinant proteins with the formation of authentic HIV virus-like particles seen on transmission electron microscopy. In addition the b12 epitope was shown to be held in common by the vaccine candidates using confocal immunofluorescent microscopy. The vaccine candidates were safely administered to Chinese cynomolgus macaques which elicited modest T cell responses at the end of the study but only one out of the three macaques elicited an HIV-specific antibody response. However, the antibodies did not neutralise primary isolates of HIV-1 or the V3-sensitive isolate SF162 using the TZM-bl b-galactosidase assay. Conclusions: MVA and FP9 are ideal replication-deficient viral vectors for HIV-1 vaccines due to their excellent safety profile for use in humans.     The bright-end galaxy candidates at z ~ 9 from 79 independent HST fields

    We present a full data analysis of the pure-parallel Hubble Space Telescope (HST) imaging observations in the Brightest of Reionizing Galaxies Survey (BoRG[z9]) in Cycle 22. The medium-deep exposures with five HST/WFC3IR+UVIS filter bands from 79 independent sightlines (~370 arcmin^2) provide the least biased determination of number density for z>9 bright galaxies against cosmic variance. After a strict two-step selection for candidate galaxies, including dropout color and photometric redshift analyses, and revision of previous BoRG candidates, we identify one source at z~10 and two sources at z~9. The z~10 candidate shows evidence of line-of-sight lens magnification (mu~1.5), yet it appears surprisingly luminous (MUV ~ -22.6\pm0.3 mag), making it one of the brightest candidates at z > 8 known (~ 0.3 mag brighter than the z = 8.68 galaxy EGSY8p7, spectroscopically confirmed by Zitrin and collaborators). For z ~ 9 candidates, we include previous data points at fainter magnitudes and find that the data are well fitted by a Schechter luminosity function with alpha ~ -2.1, MUV ~ -21.5 mag, and log phi ~ -4.5 Mpc^-3mag^-1, for the first time without fixing any parameters. The inferred cosmic star formation rate density is consistent with unaccelerated evolution from lower redshift.Comment: 18pages, 7figures, 6tables. accepted to the Astrophysical Journa

    Deposit of microbial strains in public service collections as part of the publication process to underpin good practice in science

    Despite recommendations to release microbial resources to the community post-publication, the reality is far from satisfying. A workshop discussed the need for a coordinated and effective deposition policy for 'key' microbial strains and proposes a set of criteria to facilitate their deposition into public service collections. The majority of authors either contacted directly or during submission of manuscripts to several international, mainly European bacteriology journals agreed to this set of 'key strain' criteria and to the voluntarily deposition of resources into public resource centres.The authors thank additional members of the 2011 Braunschweig workshop: Jorg Overmann, Esperanza Garay-Auban, Peter Kampfer, Yohan Lecuona, James I Prosser, Ramon Rosello-Mora, Karl-Heinz Schleifer, and Kornelia Smalla.     Constructing a WISE High Resolution Galaxy Atlas

    After eight months of continuous observations, the Wide-field Infrared Survey Explorer (WISE) mapped the entire sky at 3.4 {\mu}m, 4.6 {\mu}m, 12 {\mu}m and 22 {\mu}m. We have begun a dedicated WISE High Resolution Galaxy Atlas (WHRGA) project to fully characterize large, nearby galaxies and produce a legacy image atlas and source catalogue. Here we summarize the deconvolution technique used to significantly improve the spatial resolution of WISE imaging, specifically designed to study the internal anatomy of nearby galaxies. As a case study, we present results for the galaxy NGC 1566, comparing the WISE super-resolution image processing to that of Spitzer, GALEX and ground-based imaging.
