    Are All These Rules Necessary? Extension Pesticide Programming with a Regulatory Purpose

    Indiana\u27s private applicator recertification program includes state-required, pesticide regulatory topics. This article explores the relationship between Indiana private applicators\u27 dual attitudes towards pesticide handling practices and the pesticide regulations that mandate those practices. Newly recertified private applicators in northwest Indiana were surveyed by a mailed questionnaire. Respondents valued responsible pesticide management practices, but were collectively undecided about regulatory oversight of their pesticide handling activities. These results suggest that Extension pesticide safety educators involved in compliance education may improve their training curriculum by including material on the underlying benefits, personal and social, of pesticide regulation

    CP43, a component of Photosystem II (PSII) in higher plants, algae and cyanobacteria, is encoded by the psbC gene. Previous work demonstrated that alteration of an arginine residue occurring at position 305 to serine produced a strain (R305S) with altered PSII characteristics including lower oxygen-evolving activity, fewer assembled reaction centers, higher sensitivity to photoinactivation, etc. [Biochemistry 38 (1999) 1582]. Additionally, it was determined that the mutant exhibited an enhanced stability of its S2 state. Recently, we observed a significant chloride effect under chloride-limiting conditions. The mutant essentially lost the ability to grow photoautotrophically, assembled fewer fully functional PSII reaction centers and exhibited a very low rate of oxygen evolution. Thus, the observed phenotype of this mutation is very similar to that observed for the psbV mutant, which lacks cytochrome c550 (Biochemistry 37 (1998) 1551). A His-tagged version of the R305S mutant was produced to facilitate the isolation of PSII particles. These particles were analyzed for the presence of cytochrome c550. Reduced minus oxidized difference spectroscopy and chemiluminescence examination of Western blots indicated that cytochrome c550 was absent in these PSII particles. Whole cell extracts from the R305S mutant, however, contained a similar amount of cytochrome c550 to that observed in the control strain. These results indicate that the mutation R305S in CP43 prevents the strong association of cytochrome c550 with the PSII core complex. We hypothesize that this residue is involved in the formation of the binding domain for the cytochrome. © 2002 Elsevier Science B.V. All rights reserved

    Reading fiction and reading minds : the role of simulation in the default network

    Research in psychology has suggested that reading fiction can improve individuals’ social-cognitive abilities. Findings from neuroscience show that reading and social cognition both recruit the default network, a network which is known to support our capacity to simulate hypothetical scenes, spaces and mental states. The current research tests the hypothesis that fiction reading enhances social cognition because it serves to exercise the default subnetwork involved in theory of mind. While undergoing functional neuroimaging, participants read literary passages that differed along two dimensions: (i) vivid vs abstract and (ii) social vs non-social. Analyses revealed distinct subnetworks of the default network respond to the two dimensions of interest: the medial temporal lobe subnetwork responded preferentially to vivid passages, with or without social content; the dorsomedial prefrontal cortex (dmPFC) subnetwork responded preferentially to passages with social and abstract content. Analyses also demonstrated that participants who read fiction most often also showed the strongest social cognition performance. Finally, mediation analysis showed that activity in the dmPFC subnetwork in response to the social content mediated this relation, suggesting that the simulation of social content in fiction plays a role in fiction’s ability to enhance readers’ social cognition

    Asia is the second most rapidly urbanising region globally, currently 48% of the Asian population resides in urban areas. Disturbance to the natural environment as a result of urbanisation is expected to be significant since natural resource consumption (e.g. water, energy) and the physical expansion of cities are both outpacing population growth. While cities have been characterised in terms of their economic, social and environmental situation, the role of geology for city resilience and sustainability is under-appreciated. Using Greater Hanoi - a soft-sediment city lying in the Red River Catchment, contrasted with Greater London, a riverine city located in the Thames River Catchment - we illustrate how an understanding of the urban geological environment supported by data informatics, sensor technologies and modelling systems, may be used to underpin urban development and sustainable use of the subsurface. The priority urban challenges identified as part of Hanoi’s Master Planning - including future transport infrastructure, groundwater management & drainage, subsidence, and shallow geothermal energy utilisation are used as case studies to highlight the potential benefits of 3D urban geology approaches and digital data workflows. The benefits of geoscience knowledge-exchange networks across government and private sector partners are also highlighted

    Therapeutic efficacy of favipiravir against Bourbon virus in mice

    Bourbon virus (BRBV) is an emerging tick-borne RNA virus in the orthomyxoviridae family that was discovered in 2014. Although fatal human cases of BRBV have been described, little is known about its pathogenesis, and no antiviral therapies or vaccines exist. We obtained serum from a fatal case in 2017 and successfully recovered the second human infectious isolate of BRBV. Next-generation sequencing of the St. Louis isolate of BRBV (BRBV-STL) showed >99% nucleotide identity to the original reference isolate. Using BRBV-STL, we developed a small animal model to study BRBV-STL tropism in vivo and evaluated the prophylactic and therapeutic efficacy of the experimental antiviral drug favipiravir against BRBV-induced disease. Infection of Ifnar1-/- mice lacking the type I interferon receptor, but not congenic wild-type animals, resulted in uniformly fatal disease 6 to 10 days after infection. RNA in situ hybridization and viral yield assays demonstrated a broad tropism of BRBV-STL with highest levels detected in liver and spleen. In vitro replication and polymerase activity of BRBV-STL were inhibited by favipiravir. Moreover, administration of favipiravir as a prophylaxis or as post-exposure therapy three days after infection prevented BRBV-STL-induced mortality in immunocompromised Ifnar1-/- mice. These results suggest that favipiravir may be a candidate treatment for humans who become infected with BRBV

    Datos del subsuelo y su conocimiento para las Ciudades del Mañana: lecciones aprendidas de Glasgow y su aplicabilidad en otros lugares

    El conocimiento del subsuelo es de vital importancia en la planificación y ejecución exitosa de proyectos de construcción y regeneración urbanas. Para abordar en el área de Glasgow éste y otros temas del subsuelo urbano (por ejemplo, la planificación, las inundaciones, la contaminación), el proyecto Clyde-Urban Super-Project (CUSP) del Servicio Geológico Británico (BGS, por sus siglas en inglés) ha desarrollado modelos 3D y 4D del subsuelo. Asimismo, se han producido otros conjuntos de datos de geociencias (geoquímica, agua subterránea, geología de ingeniería). Los modelos basados en información obtenida de decenas de miles de perforaciones y otras fuentes, proporcionan nuevos conocimientos sobre: la geología compleja de Glasgow, los impactos de su legado industrial, y las oportunidades para aprovechar el calor de las explotaciones mineras abandonadas. Para que los modelos y datos del proyecto CUSP fueran más accesibles, el BGS y el Ayuntamiento de Glasgow, socio clave, han establecido una red para acceder al conocimiento del subsuelo (ASK, por sus siglas en inglés). Esta red permite el intercambio de datos y conocimientos, implicando a socios de los sectores público y privado. ASK promueve el libre flujo digital de datos del subsuelo y el conocimiento entre sus socios. Las lecciones aprendidas en Glasgow se comparten a través de la Acción Europea COST (Sub-Urban), centrada en el uso sostenible del subsuelo urbano, y en la transformación de las relaciones entre los que desarrollan el conocimiento del subsuelo urbano y los que pueden beneficiarse más de él, los planificadores y promotores de las ciudades del futuro
