    The Chemical Origin of Behavior is Rooted in Abiogenesis

    We describe the initial realization of behavior in the biosphere, which we term behavioral chemistry. If molecules are complex enough to attain a stochastic element to their structural conformation in such as a way as to radically affect their function in a biological (evolvable) setting, then they have the capacity to behave. This circumstance is described here as behavioral chemistry, unique in its definition from the colloquial chemical behavior. This transition between chemical behavior and behavioral chemistry need be explicit when discussing the root cause of behavior, which itself lies squarely at the origins of life and is the foundation of choice. RNA polymers of sufficient length meet the criteria for behavioral chemistry and therefore are capable of making a choice

    NOTES: WOODPECKER FORAGE AVAILABILITY IN HABI- TAT DISTURBANCES OF THE BLACK HILLS

    Habitat disturbance events are critical to ecological systems in which some bird species have become specialized. The vegetation community, reduced competition, ability to avoid predators, nest-site characteristics, and forage opportunities within a disturbed ecosystem are all aspects that make it desirable for selection by particular species (Svärdson 1949, Cody 1981, Martin 1998). Specifically, avian species rely on the forest conditions created by fire, insects, and disease (Brawn et al. 2001, Hunter et al. 2001, Devictor et al. 2008). In the Black Hills National Forest (BHNF) of South Dakota,two major types of natural disturbances include wildfires and mountain pine beetle (Dendroctonus ponderosae; MPB) infestations. Dead trees (snags) created by these disturbances attract a suite of insects and wildlife species. Bark beetles (Family: Curculionidae, Scolytinae) and wood borer beetles (Families: Buprestidae and Cerambycidae) are of particular importance to black-backed woodpeckers (Picoides arcticus; BBWO) because they feed almost exclusively on the larvae of these insects (Beal 1911, Murphy and Lehnhausen 1998, Hutto 2006, Bonnot et al. 2008, Bonnot et al. 2009). Black-backed woodpeckers are of key interest to resource management agencies due to their habitat specialization needs and the management activities like wildfire salvage logging and pre-thinning that occur in these disturbance areas (Hutto 1995, 2006). These management activities potentially reduce nest site and food availability for BBWOs and, as a result, they were recently petitioned for protection under the Endangered Species Act (Hanson et al. 2012). Following a fire event or insect infestation, the relative probability of using trees affected by the disturbance increases over surrounding healthy trees (Rota 2013). As a result, we were interested in understanding the food that is available to the woodpeckers following these forest disturbances

    Malignant melanotic nerve sheath tumor with PRKAR1A, KMT2C and GNAQ mutations

    Malignant melanotic nerve sheath tumor (MMNST) is a rare and potentially aggressive lesion defined in the 2021 WHO Classification of Tumors of the Central Nervous System. MMNST demonstrate overlapping histologic and clinical features of schwannoma and melanoma. MMNST often harbor PRKAR1A mutations, especially within the Carney Complex. We present a case of aggressive MMNST of the sacral region in a 48-year-old woman. The tumor contained PRKAR1A frameshift pR352Hfs*89, KMT2C splice site c.7443-1G>T and GNAQ p.R183L missense mutations, as well as BRAF and MYC gains. Genomic DNA methylation analysis using the Illumina 850K EpicBead chip revealed that the lesion did not match an established methylation class; however, uniform manifold approximation and projection (UMAP) placed the tumor very near, or with, schwannomas. The tumor expressed PD-L1, and the patient was treated with radiation and immune checkpoint inhibitors following en bloc resection. Although she had symptomatic improvement, she suffered early disease progression with local recurrence, and distant metastases, and died 18 months after resection. It has been suggested that the presence of GNAQ mutations can differentiate leptomeningeal melanocytic neoplasms and uveal melanoma from MMNST. This case and others demonstrate that GNAQ mutations may exist in malignant nerve sheath tumors; that GNAQ and PRKAR1A mutations are not always mutually exclusive and that neither can be used to differentiate MMNST or MPNST from all melanocytic lesions

    A simple method for construction of pir+ Enterobacterial hosts for maintenance of R6K replicon plasmids

    <p>Abstract</p> <p>Background</p> <p>The R6K replicon is one of the best studied bacterial plasmid replicons. Replication of the R6K plasmid and derivatives harboring its γ origin of replication (<it>ori</it><sub>R6Kγ</sub>) is dependent on the <it>pir </it>gene-encoded π protein. Originally encoded by R6K, this protein is usually provided <it>in trans </it>in hosts engineered to support replication of plasmids harboring <it>ori</it><sub>R6Kγ</sub>. In <it>Escherichia coli </it>this is commonly achieved by chromosomal integration of <it>pir </it>either via lysogenization with a λ<it>pir </it>phage or homologous recombination at a pre-determined locus.</p> <p>Findings</p> <p>Current methods for construction of host strains for <it>ori</it><sub>R6Kγ</sub>-containing plasmids involve procedures that do not allow selection for presence of the <it>pir </it>gene and require cumbersome and time-consuming screening steps. In this study, we established a mini-Tn<it>7</it>-based method for rapid and reliable construction of <it>pir</it><sup>+ </sup>host strains. Using a curable mini-Tn<it>7 </it>delivery plasmid, <it>pir </it>expressing derivatives of several commonly used <it>E. coli </it>cloning and mobilizer strains were isolated using both the wild-type <it>pir<sup>+ </sup></it>gene as well as the copy-up <it>pir-116 </it>allele. In addition, we isolated <it>pir</it><sup>+ </sup>and <it>pir-116 </it>expressing derivatives of a clinical isolate of <it>Salmonella enterica </it>serovar Typhimurium. In both <it>E. coli </it>and <it>S. enterica </it>serovar Typhimurium, the presence of the <it>pir<sup>+ </sup></it>wild-type or <it>pir-116 </it>alleles allowed the replication of <it>ori</it><sub>R6Kγ</sub>-containing plasmids.</p> <p>Conclusions</p> <p>A mini-Tn<it>7 </it>system was employed for rapid and reliable engineering of <it>E. coli </it>and <it>S. enterica </it>serovar Typhimurium host strains for plasmids containing <it>ori</it><sub>R6Kγ</sub>. Since mini-Tn7 elements transpose in most, if not all, Gram negative bacteria, we anticipate that with relatively minor modifications this newly established method will for the first time allow engineering of other bacterial species to enable replication of plasmids with <it>ori</it><sub>R6Kγ</sub>.</p

    Muscle recruitment patterns during the prone leg extension

    BACKGROUND: The prone leg extension (PLE) is a clinical test used to evaluate the function of the lumbopelvis. It has been theorized that a normal and consistent pattern of muscle activation exists. Previous research has found two contradictory patterns of muscle activation during PLE in normal individuals. One study shows an almost simultaneous activation of the lower erector spinae and hamstring muscle group with a delayed activation of the gluteus maximus, while the second describes the order of activation being ipsilateral erector spinae (to the leg being extended), hamstrings, contralateral erector spinae and gluteus maximus. Due to the different conclusions from these two studies and the lack of quantified muscle onset times, expressed in absolute time this study attempted to quantify the muscle onset times (in milliseconds) during the prone leg extension, while noting if a consistent order of activation exists and whether a timing relationship also exists between the gluteus maximus and contralateral latissimus dorsi. METHODS: 10 asymptomatic males (Average height: 175.2 cm (SD 6.5), Average Weight 75.9 kg (SD 6.5), Average Age: 27.1(SD 1.28)) and 4 asymptomatic females (Average height 164.5 (SD 2.9), weight: 56.2 (SD 8.9), Average Age: 25 (SD 1)) performed the prone leg extension task while the myoelectric signal was recorded from the bilateral lower erector spinae, gluteus maximus and hamstring muscle groups. Activation onsets were determined from the rectified EMG signal relative to the onset of the hamstrings muscle group. RESULTS: No consistent recruitment patterns were detected for prone leg extension among the hamstring muscle group and the erector spinae. However, a consistent delay in the Gluteus Maximus firing of approximately 370 ms after the first muscle activated was found. Five out of 14 asymptomatic subjects showed a delay in gluteus maximus firing exceeding the average delay found in previous research of subjects considered to have a dysfunctional firing pattern. CONCLUSION: A consistent pattern of activation was not found. Variability was seen across subjects. These findings suggest the PLE is not sufficient for a diagnostic test due to the notable physiological variation. An overlap between normal and potentially abnormal activation patterns may exist

    National Performance Benchmarks for Modern Screening Digital Mammography: Update from the Breast Cancer Surveillance Consortium

    Purpose To establish performance benchmarks for modern screening digital mammography and assess performance trends over time in U.S. community practice. Materials and Methods This HIPAA-compliant, institutional review board-approved study measured the performance of digital screening mammography interpreted by 359 radiologists across 95 facilities in six Breast Cancer Surveillance Consortium (BCSC) registries. The study included 1 682 504 digital screening mammograms performed between 2007 and 2013 in 792 808 women. Performance measures were calculated according to the American College of Radiology Breast Imaging Reporting and Data System, 5th edition, and were compared with published benchmarks by the BCSC, the National Mammography Database, and performance recommendations by expert opinion. Benchmarks were derived from the distribution of performance metrics across radiologists and were presented as 50th (median), 10th, 25th, 75th, and 90th percentiles, with graphic presentations using smoothed curves. Results Mean screening performance measures were as follows: abnormal interpretation rate (AIR), 11.6 (95% confidence interval [CI]: 11.5, 11.6); cancers detected per 1000 screens, or cancer detection rate (CDR), 5.1 (95% CI: 5.0, 5.2); sensitivity, 86.9% (95% CI: 86.3%, 87.6%); specificity, 88.9% (95% CI: 88.8%, 88.9%); false-negative rate per 1000 screens, 0.8 (95% CI: 0.7, 0.8); positive predictive value (PPV) 1, 4.4% (95% CI: 4.3%, 4.5%); PPV2, 25.6% (95% CI: 25.1%, 26.1%); PPV3, 28.6% (95% CI: 28.0%, 29.3%); cancers stage 0 or 1, 76.9%; minimal cancers, 57.7%; and node-negative invasive cancers, 79.4%. Recommended CDRs were achieved by 92.1% of radiologists in community practice, and 97.1% achieved recommended ranges for sensitivity. Only 59.0% of radiologists achieved recommended AIRs, and only 63.0% achieved recommended levels of specificity. Conclusion The majority of radiologists in the BCSC surpass cancer detection recommendations for screening mammography; however, AIRs continue to be higher than the recommended rate for almost half of radiologists interpreting screening mammograms. © RSNA, 2016 Online supplemental material is available for this article
