    Structure and Metal Binding Properties of ZnuA, a Periplasmic Zinc Transporter from \u3cem\u3eEscherichia coli\u3c/em\u3e

    ZnuA is the periplasmic Zn2+-binding protein associated with the high-affinity ATP-binding cassette ZnuABC transporter from Escherichia coli. Although several structures of ZnuA and its homologs have been determined, details regarding metal ion stoichiometry, affinity, and specificity as well as the mechanism of metal uptake and transfer remain unclear. The crystal structures of E. coli ZnuA (Eco-ZnuA) in the apo, Zn2+-bound, and Co2+-bound forms have been determined. ZnZnuA binds at least two metal ions. The first, observed previously in other structures, is coordinated tetrahedrally by Glu59, His60, His143, and His207. Replacement of Zn2+ with Co2+ results in almost identical coordination geometry at this site. The second metal binding site involves His224 and several yet to be identified residues from the His-rich loop that is unique to Zn2+ periplasmic metal binding receptors. Electron paramagnetic resonance and X-ray absorption spectroscopic data on CoZnuA provide additional insight into possible residues involved in this second site. The second site is also detected by metal analysis and circular dichroism (CD) titrations. Eco-ZnuA binds Zn2+ (estimated K d \u3c 20 nM), Co2+, Ni2+, Cu2+, Cu+, and Cd2+, but not Mn2+. Finally, conformational changes upon metal binding observed in the crystal structures together with fluorescence and CD data indicate that only Zn2+ substantially stabilizes ZnuA and might facilitate recognition of ZnuB and subsequent metal transfer

    Varied Response of Western Pacific Hydrology to Climate Forcings over the Last Glacial Period

    Atmospheric deep convection in the west Pacific plays a key role in the global heat and moisture budgets, yet its response to orbital and abrupt climate change events is poorly resolved. Here, we present four absolutely dated, overlapping stalagmite oxygen isotopic records from northern Borneo that span most of the last glacial cycle. The records suggest that northern Borneo’s hydroclimate shifted in phase with precessional forcing but was only weakly affected by glacial-interglacial changes in global climate boundary conditions. Regional convection likely decreased during Heinrich events, but other Northern Hemisphere abrupt climate change events are notably absent. The new records suggest that the deep tropical Pacific hydroclimate variability may have played an important role in shaping the global response to the largest abrupt climate change events

    Leatherback Turtles in the Eastern Gulf of Mexico: Foraging and Migration Behavior During the Autumn and Winter

    We deployed 19 satellite tags on foraging adult leatherback turtles, including 17 females and 2 males, captured in the northeastern Gulf of Mexico in 2015, 2018, and 2019 in order to study regional distribution and movements. Prior to our study, limited data were available from leatherbacks foraging in the Gulf of Mexico. Tag deployment durations ranged from 63 to 247 days and turtles exhibited three distinct behavior types: foraging, transiting, or rapidly switching between foraging and transiting. Some females were tracked to nesting beaches in the Caribbean. Most of the leatherbacks remained on and foraged along the west Florida continental shelf whereas a few individuals foraged in waters of the central Gulf of Mexico during the autumn and winter. In addition, migration of adult females through the Yucatan Channel indicate that this is a seasonally important area for Caribbean nesting assemblages

    Positive Ecological Interactions and the Success of Seagrass Restoration

    Seagrasses provide multiple ecosystem services including nursery habitat, improved water quality, coastal protection, and carbon sequestration. However, seagrasses are in crisis as global coverage is declining at an accelerating rate. With increased focus on ecological restoration as a conservation strategy, methods that enhance restoration success need to be explored. Decades of work in coastal plant ecosystems, including seagrasses, has shown that positive species relationships and feedbacks are critical for ecosystem stability, expansion, and recovery from disturbance. We reviewed the restoration literature on seagrasses and found few studies have tested for the beneficial effects of including positive species interactions in seagrass restoration designs. Here we review the full suite of positive species interactions that have been documented in seagrass ecosystems, where they occur, and how they might be integrated into seagrass restoration. The few studies in marine plant communities that have explicitly incorporated positive species interactions and feedbacks have found an increase in plant growth with little additional resource investment. As oceans continue to change and stressors become more prevalent, harnessing positive interactions between species through innovative approaches will likely become key to successful seagrass restoration

    The truth about snitches: an archival analysis of informant testimony

    Informants are witnesses who often testify in exchange for an incentive (i.e. jailhouse informant, cooperating witness). Despite the widespread use of informants, little is known about the circumstances surrounding their use at trial. This study content-analyzed trials from 22 DNA exoneration cases involving 53 informants. Because these defendants were exonerated, the prosecution informant testimony is demonstrably false. Informant characteristics including motivation for testifying, criminal history, relationship with the defendant and testimony were coded. Most informants were prosecution jailhouse informants; however, there were also defence jailhouse informants and prosecution cooperating witnesses. Regardless of informant type, most denied receiving an incentive, had criminal histories, were friends/acquaintances of the defendant and had testimonial inconsistencies. In closing statements, attorneys relied on informant testimony by either emphasizing or questioning its reliability. The impact of informant testimony on jurors’ decisions is discussed in terms of truth-default theory (TDT), the fundamental attribution error and prosecutorial vouching

    Transformation of ENSO-related rainwater to dripwater δ^(18)O variability by vadose water mixing

    Speleothem oxygen isotopes (δ^(18)O) are often used to reconstruct past rainfall δ^(18)O variability, and thereby hydroclimate changes, in many regions of the world. However, poor constraints on the karst hydrological processes that transform rainfall signals into cave dripwater add significant uncertainty to interpretations of speleothem-based reconstructions. Here we present several 6.5 year, biweekly dripwater δ^(18)O time series from northern Borneo and compare them to local rainfall δ^(18)O variability. We demonstrate that vadose water mixing is the primary rainfall-to-dripwater transformation process at our site, where dripwater δ^(18)O reflects amount-weighted rainfall δ^(18)O integrated over the previous 3–10 months. We document large interannual dripwater δ^(18)O variability related to the El Niño–Southern Oscillation (ENSO), with amplitudes inversely correlated to dripwater residence times. According to a simple stalagmite forward model, asymmetrical ENSO extremes produce significant offsets in stalagmite δ^(18)O time series given different dripwater residence times. Our study highlights the utility of generating multiyear, paired time series of rainfall and dripwater δ^(18)O to aid interpretations of stalagmite δ^(18)O reconstructions

    Northern Borneo stalagmite records reveal West Pacific hydroclimate across MIS 5 and 6

    Over the past decades, tropical stalagmite δ^(18)O records have provided valuable insight on glacial and interglacial hydrological variability and its relationship to a variety of natural climate forcings. The transition out of the penultimate glaciation (MIS 6) represents an important target for tropical hydroclimate reconstructions, yet relatively few such reconstructions resolve this transition. Particularly, comparisons between Termination 1 and 2 provide critical insight on the extent and influence of proposed climate mechanisms determined from paleorecords and model experiments spanning the recent deglaciation. Here we present a new compilation of western tropical Pacific hydrology spanning 0–160 ky BP, constructed from eleven different U/Th-dated stalagmite δ^(18)O records from Gunung Mulu National Park in northern Borneo. The reconstruction exhibits significant precessional power in phase with boreal fall insolation strength over the 0–160 ky BP period, identifying precessional insolation forcing as the dominant driver of hydroclimate variability in northern Borneo on orbital timescales. A comparison with a network of paleoclimate records from the circum-Pacific suggests the insolation sensitivity may arise from changes in the Walker circulation system. Distinct millennial-scale increases in stalagmite δ^(18)O, indicative of reduced regional convection, occur within glacial terminations and may reflect a response to shifts in inter-hemispheric temperature gradients. Our results imply that hydroclimate in this region is sensitive to external forcing, with a response dominated by large-scale temperature gradients
