    Enhancing Oxygen Uptake Efficiency Through Nasal Breathing in Aerobic Exercise

    An individual’s ability to use oxygen to sustain aerobic performance, as denoted by the oxygen uptake efficiency (OUE), is calculated by dividing oxygen uptake (VO2) by ventilation (VE). Whether nasal breathing (NB) during exercise would improve OUE more significantly compared to combined breathing (CB) remains uncertain. PURPOSE: To determine the effects of utilizing NB during exercise on OUE. METHODS: Fourteen males (age: 20.57 ± 1.22 yrs; BMI: 26.03 ± 3.16 kg/m2) were randomly assigned to the NB (n = 8) or CB (n = 6) group for a 4-week supervised aerobic exercise intervention conducted four times per week for 30 minutes at moderate intensity (70% maximal heart rate). VO2max tests were performed during the pre- (week 0) and post-study (week 4) periods on a recumbent bike to determine the change in OUE, VO2, and VE at varying intensities (40%, 55%, 70%, 85%, and 100% of VO2max). The OUE was calculated as VO2 (L) ÷ VE (L). A one-way ANCOVA, controlling for OUE, VO2, and VE at pre-study, was utilized to examine if NB elicited cardiorespiratory adaptations that were superior to the CB training at post-study. A paired-sample t-test examined changes in OUE, VO2, and VE throughout the GXT from week 0 to week 4. Data are presented as mean ± standard error. RESULTS: The NB group demonstrated significantly greater [F(1,11) = 7.213, p = 0.021] OUE (0.048 ± 0.002) at 85% of VO2max than the CB (0.037 ± 0.003) group. Similarly, the NB group had a significantly greater (p = 0.014) OUE (0.025 ± 0.001) at 100% of VO2max than the CB (0.021 ± 0.001) group. Only the NB group experienced significant improvements after exercise training in VO2 at 40% ( p = 0.003, 12.02 ± 0.50 to 13.72 ± 0.59 mL·kg·min-1], 55% (p = 0.006, 16.68 ± 0.71 to 18.87 ± 0.81 mL·kg·min-1), 70% (p = 0.005, 21.16 ± 0.93 to 24.00 ± 1.01 mL·kg·min-1), 85% (p = 0.004, 25.81 ± 1.11 to 29.03 ± 1.23 mL·kg·min-1), and 100% VO2max (p = 0.005, 30.31 ± 1.30 to 34.18 ± 1.48 mL·kg·min-1). While not statistically significant, the NB group displayed a trend of reduced VE compared to the CB group. CONCLUSION: The integration of NB during aerobic exercise enhanced the OUE, particularly at higher intensities, compared to CB. This adaptation is noteworthy, as the NB group achieved a comparable range of VO2 as the CB group while maintaining a lower VE after just 1-month of training. This denotes that NB could promote enhanced oxygen movement. Future studies are warranted to investigate additional health adaptations resulting from such training benefits

    Examining the cross-cultural validity of the positive affect and negative affect schedule between an Asian (Singaporean) sample and a Western (American) sample

    The positive affect and negative affect schedule (PANAS ) is a popular measure of positive (PA ) and negative affectivity (NA ). Developed and validated in Western contexts, the 20‐item scale has been frequently administered on respondents from Asian countries with the assumption of cross‐cultural measurement invariance. We examine this assumption via a rigorous multigroup confirmatory factor analysis, which allows us to assess between‐group differences in both strength of scale item‐to‐latent factor relationship (metric invariance test) and mean of each scale item (scalar invariance test), on a large sample of 1,065 respondents recruited from Singapore (Asian sample) and the United States (Western sample). We found that two items assessing PA ("excited" and "proud") and three items assessing NA ("guilty," "hostile," and "ashamed") exhibited metric noninvariance whereas 11 of the remaining metric invariant items exhibited scalar noninvariance, suggesting that the PA and NA constructs differ from what the PANAS is expected to measure for Asian respondents. Our findings serve as a cautionary note to researchers who intend to administer the PANAS in future studies as well as to researchers interpreting the results of past studies involving respondents from Asian countries

    A Study of Teacher Growth, Supervision, and Evaluation in Alberta: Policy and Perception in a Collective Case Study

    Teacher effectiveness has long been identified as critical to student success and, more recently, supporting students attaining the skills and dispositions required to be successful in the early 21st century. To do so requires that teachers engage in professional learning characterized as a shift away from conventional models of evaluation and judgment. Accordingly, school and system leaders must create “policies and environments designed to actively support teacher professional growth” (Bakkenes, Vermunt, & Webbels, 2010). This paper reports on the Alberta Teacher Growth, Supervision, and Evaluation (TGSE) Policy (Government of Alberta, 1998) through the eyes of teachers, school leaders, and superintendents. The study sought to answer the following two questions: (1) To what extent, and in what ways, do teachers, principals, and superintendents perceive that ongoing supervision by the principal provides teachers with the guidance and support they need to be successful? and, (2) To what degree, and in what ways, does the TGSE policy provide a foundation to inform future effective policy and implementation of teacher growth, supervision, and evaluation? Results affirm international findings that although a majority of principals consider themselves as instructional leaders, only about one third actually act accordingly (OECD, 2016)

    The Flying Monkey: a Mesoscale Robot that can Run, Fly, and Grasp

    The agility and ease of control make a quadrotor aircraft an attractive platform for studying swarm behavior, modeling, and control. The energetics of sustained flight for small aircraft, however, limit typical applications to only a few minutes. Adding payloads – and the mechanisms used to manipulate them – reduces this flight time even further. In this paper we present the flying monkey, a novel robot platform having three main capabilities: walking, grasping, and flight. This new robotic platform merges one of the world’s smallest quadrotor aircraft with a lightweight, single-degree-of-freedom walking mechanism and an SMA-actuated gripper to enable all three functions in a 30 g package.     Evaluation of the feasibility, diagnostic yield, and clinical utility of rapid genome sequencing in infantile epilepsy (Gene-STEPS): an international, multicentre, pilot cohort study

    BACKGROUND: Most neonatal and infantile-onset epilepsies have presumed genetic aetiologies, and early genetic diagnoses have the potential to inform clinical management and improve outcomes. We therefore aimed to determine the feasibility, diagnostic yield, and clinical utility of rapid genome sequencing in this population. METHODS: We conducted an international, multicentre, cohort study (Gene-STEPS), which is a pilot study of the International Precision Child Health Partnership (IPCHiP). IPCHiP is a consortium of four paediatric centres with tertiary-level subspecialty services in Australia, Canada, the UK, and the USA. We recruited infants with new-onset epilepsy or complex febrile seizures from IPCHiP centres, who were younger than 12 months at seizure onset. We excluded infants with simple febrile seizures, acute provoked seizures, known acquired cause, or known genetic cause. Blood samples were collected from probands and available biological parents. Clinical data were collected from medical records, treating clinicians, and parents. Trio genome sequencing was done when both parents were available, and duo or singleton genome sequencing was done when one or neither parent was available. Site-specific protocols were used for DNA extraction and library preparation. Rapid genome sequencing and analysis was done at clinically accredited laboratories, and results were returned to families. We analysed summary statistics for cohort demographic and clinical characteristics and the timing, diagnostic yield, and clinical impact of rapid genome sequencing. FINDINGS: Between Sept 1, 2021, and Aug 31, 2022, we enrolled 100 infants with new-onset epilepsy, of whom 41 (41%) were girls and 59 (59%) were boys. Median age of seizure onset was 128 days (IQR 46-192). For 43 (43% [binomial distribution 95% CI 33-53]) of 100 infants, we identified genetic diagnoses, with a median time from seizure onset to rapid genome sequencing result of 37 days (IQR 25-59). Genetic diagnosis was associated with neonatal seizure onset versus infantile seizure onset (14 [74%] of 19 vs 29 [36%] of 81; p=0·0027), referral setting (12 [71%] of 17 for intensive care, 19 [44%] of 43 non-intensive care inpatient, and 12 [28%] of 40 outpatient; p=0·0178), and epilepsy syndrome (13 [87%] of 15 for self-limited epilepsies, 18 [35%] of 51 for developmental and epileptic encephalopathies, 12 [35%] of 34 for other syndromes; p=0·001). Rapid genome sequencing revealed genetic heterogeneity, with 34 unique genes or genomic regions implicated. Genetic diagnoses had immediate clinical utility, informing treatment (24 [56%] of 43), additional evaluation (28 [65%]), prognosis (37 [86%]), and recurrence risk counselling (all cases). INTERPRETATION: Our findings support the feasibility of implementation of rapid genome sequencing in the clinical care of infants with new-onset epilepsy. Longitudinal follow-up is needed to further assess the role of rapid genetic diagnosis in improving clinical, quality-of-life, and economic outcomes.
